PURPOSE: An assay for radiosensitivity has numerous applications in the clinic. Avoidance of acute responses, prediction of normal tissue toxicity, and individualization of patient radiotherapy are included among these. We have developed a rapid assay (about 24 h) able to predict intrinsic radiosensitivity of CD4 and CD8 T-lymphocytes based on radiation-induced apoptosis.

PURPOSE: Continuous chromosome missegregation over successive mitotic divisions, known as chromosomal instability (CIN), is common in cancer. Though it has been associated with treatment resistance and poor prognosis, increasing CIN above a maximally tolerated threshold leads to cell death because of loss of essential chromosomes. A patient previously treated with radiation for base-of-tongue cancer presented with recurrent disease seven years later. The spinal cord had received tolerance dose. Using state-of-the-art treatment planning techniques, including beam's-eye-view and volumetrics, dose-volume histograms, split field technique, mixed energies, and beam intensity modulation (with a compensator), we achieved uniform dose coverage of the target in 3-D. This was verified in vivo with thermoluminescence dosimeters positioned in the esophagus by means of a nasogastric tube that ran centrally through the target volume.
