Attenuating the DNA damage response to double-strand breaks restores function in models of CNS neurodegeneration.

DNA double-strand breaks are a feature of many acute and long-term neurological disorders, including neurodegeneration, following neurotrauma and after stroke. Persistent activation of the DNA damage response in response to double-strand breaks contributes to neural dysfunction and pathology as it can force post-mitotic neurons to re-enter the cell cycle leading to senescence or apoptosis. Mature, non-dividing neurons may tolerate low levels of DNA damage, in which case muting the DNA damage response might be neuroprotective. Here, we show that attenuating the DNA damage response by targeting

Oxygen vacancy-amplified photoelectrochemical sensing of 8-oxo-guanine DNA glycosylase using Bi6S2O15 nanowires.

8-Oxo-guanine DNA glycosylase (OGG) repairs oxidatively damaged DNA and correlates with various diseases, and sensitive OGG detection is vital for clinical diagnosis. Traditional detection methods have inherent drawbacks, including radiation risk and complicated preparation. Herein, a new photoelectrochemical (PEC) biosensor based on hydrothermally synthesized Bi6S2O15 nanowires was fabricated for OGG quantification via dopamine (DA)-activated cathodic photocurrent amplification. Specifically, OGG specifically cleaved 8-oxoG-containing hairpin DNA to release the DA aptamer, which was degraded

Damaged DNA-binding protein DDB stimulates the excision of cyclobutane pyrimidine dimers in vitro in concert with XPA and replication protein A.

Human cells contain a protein that binds to UV-irradiated DNA with high affinity. This protein, damaged DNA-binding protein (DDB), is a heterodimer of two polypeptides, p127 and p48. Recent in vivo studies suggested that DDB is involved in global genome repair of cyclobutane pyrimidine dimers (CPDs), but the mechanism remains unclear. Here, we show that in vitro DDB directly stimulates the excision of CPDs but not (6-4)photoproducts. The excision activity of cell-free extracts from Chinese hamster AA8 cell line that lacks DDB activity was increased 3-4-fold by recombinant DDB heterodimer but n

Recent advances in targeting histone H3 lysine 36 methyltransferases for cancer therapy.

Histone H3 lysine 36 (H3K36) methylation is a typical epigenetic histone modification that is involved in various biological processes such as DNA transcription, repair and recombination in vivo. Mutations, translocations, and aberrant gene expression associated with H3K36 methyltransferases have been implicated in different malignancies such as acute myeloid leukemia, lung cancer, multiple myeloma, and others. Herein, we provided a comprehensive overview of the latest advances in small molecule inhibitors targeting H3K36 methyltransferases. We analyzed the structures and biological functions

Niraparib in the treatment of previously treated advanced ovarian, fallopian tube or primary peritoneal cancer.

Homologous recombination deficiency is a critical biologic feature of ovarian cancer. This weakness in DNA damage repair relies on functional poly(ADP-ribose) polymerase. Niraparib is a poly(ADP-ribose) polymerase inhibitor, orally available and initially approved for maintenance therapy in women with ovarian cancer by the US FDA in 2017 and by the EMA in 2017 for the same indication. Ovarian cancer represents the most lethal of gynecologic malignancies. The efficacy of niraparib has changed the landscape of ovarian cancer treatment, but overall survival data is still to come. This review summ

The genesis and evolution of high-grade serous ovarian cancer.

Germline mutation in either BRCA1 or BRCA2 is associated with an increased risk of ovarian cancer, particularly the most common invasive histotype - serous carcinoma. In addition, serous ovarian cancers have an unusually high frequency of other molecular events involving BRCA pathway dysfunction. Recent findings show a high frequency of TP53 mutation, chromosomal instability, distinct molecular subtypes and DNA copy number-driven changes in gene expression. These findings suggest a model in which homologous recombination repair deficiency initiates a cascade of molecular events that sculpt the

Heat Shock Protein Family A Member 1 Promotes Intracellular Amplification of Hepatitis B Virus Covalently Closed Circular DNA.

Hepatitis B virus (HBV) contains a partially double-stranded relaxed circular DNA (rcDNA) genome that is converted into a covalently closed circular DNA (cccDNA) in the nucleus of the infected hepatocyte by cellular DNA repair machinery. cccDNA associates with nucleosomes to form a minichromosome that transcribes RNA to support the expression of viral proteins and reverse transcriptional replication of viral DNA. In addition to the de novo synthesis from incoming virion rcDNA, cccDNA can also be synthesized from rcDNA in the progeny nucleocapsids within the cytoplasm of infected hepatocytes vi

Similarities between exogenously- and endogenously-induced envelope stress: the effects of a new antibacterial molecule, TPI1609-10.

Antibiotics with novel and/or multiple targets are highly desirable in the face of the steady rise of clinical antibiotic resistance. We have screened and identified small molecules, typified by the compound TPI1609-10 (aka SM10), with antibiotic activity against both gram-positive and gram-negative bacteria. SM10 was screened in vitro to bind branched Holliday junction intermediates of homologous recombination and tyrosine recombinase-mediated recombination; thus, the cellular targets of the small molecules were expected to include the RuvABC Holliday junction resolvasome and the XerCD comple

BRCA1 and BRCA2 protein expressions in an ovotestis of a 46, XX true hermaphrodite.

BRCA1 and BRCA2 breast cancer susceptibility genes encode proteins, the normal cellular functions of which are complex and multiple, and germ-line mutations in individuals predispose both to breast and to ovarian cancer. There is nevertheless substantial evidence linking BRCA1 and BRCA2 to homologous recombination and DNA repair, to transcriptional control and to tissue proliferation. There is controversy regarding the localization of BRCA1 and BRCA2 proteins to either nucleus or cytoplasm and whether the expression is present in premeiotic germ cells or can still be expressed in mitotic sperm

RAD51 promotes osteogenic differentiation and inhibits DNA damage in osteoporosis though regulating cGAS-STING signaling pathway.

INTRODUCTION: Osteoporosis (OP) represents a metabolic bone disorder characterized by reduced bone density and increased fracture susceptibility, primarily caused by an imbalance between bone resorption and formation. Osteogenic differentiation plays a critical role in OP, as it promotes bone formation processes. RAD51, a crucial gene encoding a protein essential for homologous recombination repair of DNA double-strand breaks, is central to maintaining genomic stability and ensuring accurate DNA repair. Previous investigations conducted by our research team have suggested the involvement of RA

Nonhomologous end joining is essential for cellular resistance to the novel antitumor agent, beta-lapachone.

Commonly used antitumor agents, such as DNA topoisomerase I/II poisons, kill cancer cells by creating nonrepairable DNA double-strand breaks (DSBs). To repair DSBs, error-free homologous recombination (HR), and/or error-prone nonhomologous end joining (NHEJ) are activated. These processes involve the phosphatidylinositol 3'-kinase-related kinase family of serine/threonine enzymes: ataxia telangiectasia mutated (ATM), ATM- and Rad3-related for HR, and DNA-dependent protein kinase catalytic subunit (DNA-PKcs) for NHEJ. Alterations in these repair processes can cause drug/radiation resistance and

Biochemical and functional characterization of Plasmodium falciparum DNA polymerase δ.

BACKGROUND: Emergence of drug-resistant Plasmodium falciparum has created an urgent need for new drug targets. DNA polymerase δ is an essential enzyme required for chromosomal DNA replication and repair, and therefore may be a potential target for anti-malarial drug development. However, little is known of the characteristics and function of this P. falciparum enzyme. METHODS: The coding sequences of DNA polymerase δ catalytic subunit (PfPolδ-cat), DNA polymerase δ small subunit (PfPolδS) and proliferating cell nuclear antigen (PfPCNA) from chloroquine- and pyrimethamine-resistant P. falcipar

Prognostic implications of tissue-based homologous recombination deficiency in metastatic gastric cancer treated with immune checkpoint inhibitor plus chemotherapy.

BACKGROUND: Homologous recombination deficiency (HRD), a genomic instability phenotype resulting from impaired DNA repair, has been associated with increased tumor immunogenicity in several solid tumors. However, its clinical relevance in metastatic gastric cancer (mGC), particularly in the context of immunotherapy-containing regimens, remains unclear. METHODS: This study included 139 GC patients with nivolumab plus chemotherapy as first line between May 2022 and May 2024 and underwent tissue-based NGS (n = 116); a smaller subset additionally underwent ctDNA-based NGS (n = 24). ATP Hydrolysis Gates an Ultra-Rapid Unwinding Mode of AfXPB Revealed by DNA Electrochemistry.

DNA helicases play a central role in DNA metabolism, enabling recombination, replication, transcription, translation, and repair. Recently, an unexpectedly rapid, exponential ATP-stimulated DNA unwinding rate was observed with an Archaeoglobus fulgidus helicase (AfXPB), markedly faster than that of conventional helicases, yet the mechanistic basis remains unresolved. Here, we distinguish the roles of ATP-helicase binding and ATP hydrolysis on the dynamic DNA unwinding activity of AfXPB via an electrochemical assay with redox-active DNA monolayers. To distinguish ATP-helicase binding from ATP h

RAD52 inactivation is synthetically lethal with deficiencies in BRCA1 and PALB2 in addition to BRCA2 through RAD51-mediated homologous recombination.

Synthetic lethality is an approach to study selective cell killing based on genotype. Previous work in our laboratory has shown that loss of RAD52 is synthetically lethal with BRCA2 deficiency, while exhibiting no impact on cell growth and viability in BRCA2-proficient cells. We now show that this same synthetically lethal relationship is evident in cells with deficiencies in BRCA1 or PALB2, which implicates BRCA1, PALB2 and BRCA2 in an epistatic relationship with one another. When RAD52 was depleted in BRCA1- or PALB2-deficient cells, a severe reduction in plating efficiency was observed, wit

Targeting MMR-deficient colorectal cancer with a potent small molecule UNI110.

Mismatch repair (MMR) deficiency is a hallmark of microsatellite instability (MSI) in hereditary non-polyposis colorectal cancer, Lynch syndrome, contributing to resistance against conventional chemotherapy and posing a significant therapeutic challenge. In this study, we introduce UNI110, a novel small molecule derived from Baicalein, engineered for enhanced selectivity against MMR-deficient cancer cells. UNI110 exhibits a remarkable sevenfold increase in potency over Baicalein, demonstrating significantly lower IC50 values and heightened cytotoxic effects in MMR-deficient cell lines. Mechani

Comprehensive analysis of BRCA1/2 mutations, "BRCAness" and PARP inhibitors in melanoma.

The BRCA1 and BRCA2 genes encode critical proteins in the homologous recombination (HR) DNA repair pathway. While their role in hereditary breast and ovarian cancer syndromes is well established, recent evidence suggests a more nuanced role in other malignancies, including melanoma, where homologous recombination deficiency (HRD) occurs in 18-57 % of cases. This review is the first to comprehensively synthesize current findings on epidemiology and molecular mechanisms, while investigating potential predictive biomarkers, diagnostic implications, and therapeutic relevance of BRCA1/2 mutations i

Targeting RAD51 phosphotyrosine-315 to prevent unfaithful recombination repair in BCR-ABL1 leukemia.

Chronic myeloid leukemia chronic phase (CML-CP) CD34(+) cells contain numerous DNA double-strand breaks whose unfaithful repair may contribute to chromosomal instability and disease progression to blast phase (CML-BP). These phenomena are often associated with the appearance of imatinib-resistant BCR-ABL1 kinase mutants (eg, T315I) and overexpression of BCR-ABL1. Here we show that BCR-ABL1 (nonmutated and T315I mutant) promoted RAD51 recombinase-mediated unfaithful homeologous recombination repair (HomeoRR) in a dosage-dependent manner. BCR-ABL1 SH3 domain interacts with RAD51 proline-rich reg

Targeting the ATM-TRMT10A-BRCA1 axis confers synthetic lethality to PARP inhibition in metastatic castration-resistant prostate cancer.

Metastatic castration-resistant prostate cancer (mCRPC) progresses aggressively and resists existing therapies. Although poly(ADP-ribose) polymerase inhibitors (PARPis) benefit a subset of patients with mCRPC and BRCA1/2 deficiencies, therapeutic options remain limited for those without such mutations. Here, we uncover a critical role for the ATM-TRMT10A-BRCA1 signaling axis in regulating homologous recombination (HR) repair and PARPi sensitivity. We demonstrate that ATM phosphorylates TRMT10A at serine-28 after DNA damage, promoting BRCA1 recruitment and efficient HR repair. TRMT10A deletion

Mutational signature dynamics shaping the evolution of oesophageal adenocarcinoma.

A variety of mutational processes drive cancer development, but their dynamics across the entire disease spectrum from pre-cancerous to advanced neoplasia are poorly understood. We explore the mutagenic processes shaping oesophageal adenocarcinoma tumorigenesis in 997 instances comprising distinct stages of this malignancy, from Barrett Oesophagus to primary tumours and advanced metastatic disease. The mutational landscape is dominated by the C[T > C/G]T substitution enriched signatures SBS17a/b, which are linked with TP53 mutations, increased proliferation, genomic instability and disease pro
