Telomere-protecting protein 1 promotes gastric cancer cell metastasis via enhancing endoplasmic reticulum stress.

Gastric cancer (GC) is one of major global cancers that is highly heterogeneous and has a poor prognosis, especially in cases associated with Helicobacter pylori (H. pylori) infection. H. pylori promote metastasis via mechanisms like endoplasmic reticulum stress (ERS). Telomere-protecting protein 1 (TPP1), a telomere-protecting protein, is overexpressed in GC and linked to poor outcomes. This study investigated TPP1's role in H. pylori-induced ERS and its implications for GC metastasis. We analyzed TPP1 expression using both Starbase data and clinical samples. Functional assays (e.g., migration…

Intestinal Epithelial-Derived USP13 Alleviates Colonic Inflammation by Suppressing GRP78-mediated Endoplasmic Reticulum Stress.

Inflammatory bowel disease (IBD) is a chronic condition in which endoplasmic reticulum (ER) stress and subsequent apoptosis of intestinal epithelial cells play significant roles. E3 ubiquitin ligases and deubiquitinases are crucial in IBD pathophysiology by modulating protein ubiquitination. This study investigated the regulatory role of a deubiquitinase ubiquitin-specific peptidase 13 (USP13) in a DSS-induced colitis mouse model and explored the underlying mechanism. Intestinal epithelial-specific Usp13 knockout (USP13IEKO) mice were treated with DSS to induce colitis. Adeno-associated virus …

UFM1 at the endoplasmic reticulum: linking ER stress, ribosome quality control, and ER-phagy.

Ubiquitin-fold modifier 1 (UFM1) is a small protein that functions as a ubiquitin-like modifier attached to other proteins to alter their behavior. Although less famous than ubiquitin, UFM1 has gained attention as a key regulator of proteostasis (protein homeostasis) in the cell. Notably, the endoplasmic reticulum (ER) has emerged as the central stage for UFM1's activity. UFM1 was initially recognized for its role in the ER stress response, and we now know it orchestrates two critical quality-control processes at the ER: ribosome-associated quality control and selective autophagy of the ER. To…

2-Chloroethanol induces hepatic toxicity by disrupting endoplasmic reticulum homeostasis ameliorated by dimethyl sulfoxide.

2-Chloroethanol (2CE), a metabolite of ethylene oxide, vinyl chloride, and 1,2-dichloroethene, induces acute liver injury, but its mechanisms remain poorly defined. In this study, we demonstrate that 2CE disrupts endoplasmic reticulum (ER) homeostasis and activates unfolded protein response (UPR) signaling in both mouse liver and H4IIEC3 hepatoma cells. While in vivo eIF2α-CHOP signaling predominated, all three canonical UPR branches-IRE1α-XBP1s, PERK-eIF2α-CHOP, and ATF6-were activated in vitro. Notably, 2CE-induced XBP1s expression was transient, whereas pro-apoptotic UPR signaling persisted…

Anti-prion drugs reduce endoplasmic reticulum stress and protect human dopaminergic neurons from death.

Parkinson's disease (PD) is characterized by the pathologic aggregation of α-synuclein, which induces endoplasmic reticulum (ER) stress and activates the unfolded protein response (UPR), ultimately leading to the death of dopaminergic neurons. This study investigated whether drugs known to mitigate protein aggregation in prion disease models would attenuate the stress response and cell death in PD models. Flunarizine and ten of its structural analogues, previously identified through a drug repositioning approach to reduce the aggregation of PrPSc prion protein, were evaluated in a Chinese hams…

Adaptive and terminal endoplasmic reticulum stress genes methylation levels in Parkinson patients' peripheral blood.

BACKGROUND AND PURPOSE: Misfolded protein stress has come to the fore among the molecular mechanisms that can cause degeneration. Whereas one of the most important protein of adaptive Endoplasmic Reticulum stress (ERS) is XBP1, CHOP and ASK proteins are associated with apoptosis and terminal ERS. To the best of our knowledge, methylation levels of adaptive and terminal ERS genes in Parkinson's Disease (PD) patients' blood are unknown. We aimed to evaluate if there is a difference in the DNA methylation levels of the ERS related protein-coding genes in peripheral blood of PD patients compared w…

RCN1 downregulation-driven endoplasmic reticulum stress impairs endothelial function and diabetic foot ulcer healing.

Diabetic foot ulcers (DFUs) are a leading cause of disability and mortality, with endothelial dysfunction playing a key role in the development of non-healing ulcers. A primary driver of endothelial cell impairment in this context is endoplasmic reticulum (ER) stress, triggered by glycolipotoxicity, though the underlying mechanisms are not fully understood. In this study, we observed that diabetic mice displayed poor ulcer healing associated with reduced angiogenesis and downregulated Reticulocalbin 1 (RCN1) expression. Proteomic analysis in human umbilical vein endothelial cells (HUVECs) iden…

Endoplasmic reticulum-targeted strategies for programmed cell death in cancer therapy: Approaches and prospects.

The endoplasmic reticulum (ER) plays a dual role in cancer biology, functioning both to preserve cellular homeostasis and to facilitate pathological progression. Rapidly proliferating cancer cells, which exhibit heightened metabolic activity, frequently experience ER stress that activates the unfolded protein response (UPR), a mechanism that promotes cellular adaptation and survival. However, when ER stress is prolonged or excessive, it can shift the balance toward programmed cell death (PCD), including apoptosis, autophagy, ferroptosis, pyroptosis, and necroptosis. Given the ER's central role…

EMP1 safeguards hematopoietic stem cells by suppressing sphingolipid metabolism and alleviating endoplasmic reticulum stress.

The long-term maintenance of hematopoietic stem cells (HSCs) relies on the regulation of endoplasmic reticulum (ER) stress at a low level, but the underlying mechanism remains poorly understood. Here, we demonstrate that suppression of ER stress improves the functions of HSCs and protects HSCs against ionizing radiation (IR)-induced injury. We identify epithelial membrane protein 1 (EMP1) as a key regulator that mitigates ER stress in HSCs. Emp1 deficiency leads to the accumulation of protein aggregates and elevated ER stress, ultimately resulting in impaired HSC maintenance and self-renewal. …

RAB25/GCN1 signaling promotes endoplasmic reticulum stress to mediate alcohol-associated liver disease progression.

BACKGROUND/AIMS: Endoplasmic reticulum (ER) stress in hepatocytes plays a causative role in alcohol-associated liver disease (ALD). The incomplete inhibition of ER stress by targeting canonical ER stress sensor proteins suggests the existence of noncanonical ER stress pathways in ALD pathology. This study aimed to delineate the role of RAB25 in ALD and its regulatory mechanism in noncanonical ER stress pathways. METHODS: RAB25 activation was examined in liver samples from ALD patients and ethanol-fed mice. The interaction between RAB25 and GCN1 was confirmed through mass spectrometry and co-i…

Inhibition of Endoplasmic Reticulum Oxidoreductin 1 Modulates Neuronal Excitability and Nociceptive Sensitivity in Mice.

BACKGROUND: In the peripheral nervous system, nociceptors become hyperexcitable in both acute and chronic pain conditions. This phenotype can be mediated by dysregulated calcium, which occurs if the endoplasmic reticulum and mitochondria fail to buffer it appropriately. The redox enzyme endoplasmic reticulum oxidoreductin 1 (ERO1) regulates calcium transfer at endoplasmic reticulum-mitochondria contact sites (ERMCSs). This study hypothesized that inhibiting ERO1 and thereby dampening ERMCS calcium transfer might lower nociceptor hyperexcitability in sensory neurons and pain-like behaviors in m…

Obesity concurrent with gestational diabetes mellitus dysregulates mitochondria-endoplasmic reticulum contacts in human placenta.

Obesity concurrent with gestational diabetes mellitus (GDM) markedly increases the risk of adverse pregnancy outcomes, wherein placental dysfunction acts as a key mediating factor, however, the underlying mechanisms involved remain elusive. This pilot study was designed to focus on the role of mitochondria-endoplasmic reticulum contacts (MERCs) in obesity with GDM-induced placental dysfunction. Term placental tissues from pregnant women with obesity and GDM or healthy control were analyzed. Reduced cell count of placental trophoblasts, disorganized cell arrangement, and higher apoptosis rate w…

Regulation of calcium homeostasis in endoplasmic reticulum-mitochondria crosstalk: implications for skeletal muscle atrophy.

This review comprehensively explores the critical role of calcium as an essential small-molecule biomessenger in skeletal muscle function. Calcium is vital for both regulating muscle excitation-contraction coupling and for the development, maintenance, and regeneration of muscle cells. The orchestrated release of calcium from the endoplasmic reticulum (ER) is mediated by receptors such as the ryanodine receptor (RYR) and inositol 1,4,5-trisphosphate receptor (IP3R), which is crucial for skeletal muscle contraction. The sarcoendoplasmic reticulum calcium ATPase (SERCA) pump plays a key role in …

Targeting the unfolded protein response in cancer: exploiting endoplasmic reticulum stress for therapeutic intervention.

Targeting the unfolded protein response (UPR) has become an attractive cancer therapeutic approach. It takes advantage of the distinct ways tumor cells adapt to stress for their growth and survival. Due to rapid cell division, low oxygen levels, and limited nutrients, cancer cells often undergo endoplasmic reticulum (ER) stress, which activates the UPR. The UPR response is controlled by major sensors-inositol-requiring enzyme 1 (IRE1), protein kinase RNA-like endoplasmic reticulum kinase (PERK), and activating transcription factor 6 (ATF6), which help cancer cells adapt and avoid cell death, b…

Toxoplasma gondii VIP1 mediates parasitophorous vacuole-host endoplasmic reticulum interactions to facilitate parasite development.

Membrane contact sites (MCS) are areas of close apposition between organelles without membrane fusion, allowing for exchange of biomolecules. The endoplasmic reticulum (ER) forms many MCS via two proteins, vesicle-associated membrane protein-associated proteins A and B (VAPA and VAPB). The obligate intracellular parasite Toxoplasma gondii resides within mammalian cells in a parasitophorous vacuole (PV), which closely contacts the host ER at distances compatible with MCS. However, the proteins mediating this interaction remain largely unknown. Here, using molecular and microscopy approaches, we…
