Modulation of SIRT1/PPARγ pathways and tight junction proteins by nicotinamide riboside under chronic variable stress.

Chronic stress disrupts homeostasis, leading to major health problems such as liver damage, intestinal barrier dysfunction, and impaired glucose metabolism. Although current treatments, including anxiolytics, sedatives, antidepressants, and beta blockers, are effective, their adverse effects emphasize the need for safer alternatives. Nicotinamide riboside (NR), a precursor of nicotinamide adenine dinucleotide (NAD+), plays a central role in energy metabolism and oxidative stress regulation; elevated NAD + levels have been associated with reduced risk of chronic diseases such as obesity and typ

Oxidative Stress and SIRT1-Nrf2 Anti-Ferroptotic Pathways in Granulosa Cells: A Molecular Key to Follicular Atresia and Ovarian Aging.

The functional deterioration of granulosa cells (GCs), essential for follicular growth, steroidogenesis, and oocyte competence, indicates ovarian aging and reduced fertility. An expanding corpus of research indicates that oxidative stress is a primary molecular contributor to granulosa cell dysfunction, culminating in mitochondrial impairment, reduced metabolic support for oocytes, and the activation of regulated apoptotic pathways that end in follicular atresia. Ferroptosis, an emergent type of iron-dependent lipid peroxidation, has been identified as a crucial mechanism contributing to chemo

The activation of SIRT1-Nrf2 axis exerts beneficial effects against rotenone-induced cognitive deficits in mice through inhibition of neuroinflammation and ferroptosis.

Rotenone, a widely used agricultural pesticide has been linked to an increased risk of Parkinson's disease (PD) following chronic exposure. Our previous study found that rotenone not only caused motor deficits in mice, but was also able to cause cognitive deficits, a common non-motor symptom in PD. This study aimed to explore whether activation of the SIRT1-Nrf2 axis confers neuroprotection against rotenone-induced cognitive impairments. We found that rotenone significantly decreased the expression and activation of SIRT1 and Nrf2 in the hippocampus of mice. Pharmacological activation of SIRT1

Tregs are key mediators of renal dysfunction induced by low-dose fluoride exposure.

Against the backdrop of generally reduced fluoride exposure levels in the current environment, the immune barrier function, as the core of the body's defense system, has attracted significant attention for its role in immune regulation under fluoride exposure. The kidneys, serving as a key target organ for Tregs-mediated immune homeostasis, rely on Foxp3 acetylation as a core regulatory mechanism. However, the role of Tregs in fluoride-induced renal injury remains unclear. In this cross-sectional study, we collected data from 336 residents in areas with low-to-moderate fluoride exposure (water

The Role of SIRT1 in Mediating Bone Marrow Edema and Its Interface With Bone Remodeling.

BACKGROUND: Bone marrow edema (BME), a common magnetic resonance imaging (MRI) finding characterized by increased fluid signal, is associated with pain and structural deterioration in conditions such as osteoarthritis and avascular necrosis. The pathophysiology of BME involves a complex interplay of vascular leakage, inflammation, microdamage, disrupted bone remodeling, and increased marrow adiposity. Sirtuin 1 (SIRT1), an NAD+-dependent deacetylase, is a critical regulator of bone homeostasis, promoting osteoblastogenesis, inhibiting osteoclastogenesis, and suppressing bone marrow adipose tis…

Diosmetin's impact on epileptic seizures: a study on inflammatory pathways and neuronal health.

This study investigated the anti-seizure potential of diosmetin (DM), a naturally occurring active compound, and explored its molecular mechanisms. A pentetrazole (PTZ)-induced chemical kindling mouse model was employed to assess seizure severity, cognitive function, motor coordination, and anxiety-like behaviors. Hippocampal neuronal injury was examined by hematoxylin-eosin and Nissl staining. In addition, an in vitro seizure model was established in PC12 cells using Mg2+-free extracellular fluid to evaluate the effects of DM on cell viability and apoptosis. Levels of inflammatory mediators w

Nanoparticle-mediated SIRT1 inhibition suppresses M2 macrophage polarization and hepatocarcinogenesis in chronic hepatitis B.

Hepatocellular carcinoma (HCC) is a major complication of chronic hepatitis B (CHB), with macrophage M2 polarization playing a critical role in shaping the tumor-promoting hepatic immune microenvironment. Sirtuin 1 (SIRT1) has been implicated in immune modulation and liver carcinogenesis. This study investigates the potential of Mimetic Nanoparticles (MNPs) for delivering SIRT1 inhibitors to regulate macrophage polarization and remodel the hepatic immune microenvironment, aiming to prevent HCC development post-CHB. A transgenic mouse model of CHB was established, and RNA sequencing (RNA-seq) a…

Melatonin Attenuates the Inflammatory Response of Astrocyte Following Ischemia Stroke through SIRT1/NF-κΒ Signaling Pathway.

Ischemic stroke (IS) remains a significant health burden with high rates of disability and mortality. Rapid reperfusion is critical for treating IS, yet paradoxically, it induces reperfusion injury. Astrocytes, the most abundant glial cells in the CNS, are increasingly recognized for their role in neuroinflammation and tissue repair post-ischemia. Silent information regulator 2 homolog 1 (SIRT1), a NAD + -dependent deacetylase, has emerged as a key regulator of cell survival, metabolism, and neuroprotection. Melatonin, an endogenous hormone with potent anti-inflammatory properties, has demonst

Regulation of macrophage pyroptosis by SIRT1 during Pseudomonas aeruginosa-induced pneumonia.

Pseudomonas aeruginosa (PA), an opportunistic gram-negative bacterial pathogen, is the main cause of lung infections. Pulmonary infection induced by PA results in inflammatory lung injury characterized by macrophage pyroptosis. In our study, a mouse model of pulmonary infection was established by tracheal intubation with PA in vivo, and the MH-S macrophage line was stimulated with PA in vitro. HE staining was performed to observe changes in the lung tissue. The mechanism was further explored through various methods, including flow cytometry, LDH release assays, ELISA, real-time PCR, Western bl

Kallistatin Improves Lipid Metabolism and Alleviates Cardiac Hypertrophy via the SIRT1/PPAR Pathway: An Experimental Study.

Cardiovascular disease is a major health concern, with cardiac hypertrophy (CH) leading to heart failure and increased mortality. Although kallistatin has shown a protective effect against cardiovascular diseases, its role in CH remains unclear. The effects of kallistatin on myocardial lipid metabolism, inflammation, and hypertrophy via the SIRT1/PPARα pathway in both animal model and cell culture were assessed. The rat model of CH was induced by Angiotensin II (Ang II). Plasma kallistatin and inflammatory indicators (IL-6, TNF-α, MCP-1) were measured using ELISA. In Vitro, Ang II-treated NRVM

Groundbreaking Insights Into SIRT1/NRF2-Mediated Ferroptosis Inhibition by Resveratrol in Parkinson's Disease Models.

BACKGROUND: Parkinson's disease (PD) is the second most prevalent neurodegenerative disorder, characterized by the degeneration of dopamine (DA) neurons in the substantia nigra (SN) of the midbrain. Recent studies have highlighted the role of ferroptosis in neuronal death, with elevated peroxide levels being a hallmark of this process. Resveratrol (RSV), a natural compound, has shown promise as a neuroprotective agent. This study explores the potential of RSV in mitigating ferroptosis in PD and elucidates its mechanisms. METHODS: Network pharmacology was employed to predict the interactions b

Niacin enhances hematoma clearance and neurological recovery via the HCAR2/SIRT1/Nrf2 pathway after germinal matrix hemorrhage.

Germinal matrix hemorrhage (GMH) is a common form of neonatal stroke in preterm infants and often results in severe and lasting neurological deficits. Persistent hematoma is a key contributor to secondary brain injury. In this study, we investigated the therapeutic potential of niacin in promoting hematoma clearance and neuroprotection after GMH. We found that intranasal administration of niacin markedly improved neurological function, accelerated hematoma clearance, and attenuated secondary brain damage in neonatal rats. Mechanistically, niacin activated HCAR2(hydroxycarboxylic acid receptor

Tetramethylpyrazine alleviates ventricular remodeling after myocardial infarction by regulating Sirt1/p300/Yy1/sST2 signaling axis.

BACKGROUND: Adverse ventricular remodeling following myocardial infarction (MI) is a critical factor in the progression of heart failure (HF). However, clinical interventions are limited by the lack of reliable biomarkers and targeted therapies. Soluble suppression of tumorigenicity 2 (sST2) is recognized as a prognostic biomarker in HF, with elevated levels associated with poorer outcomes, while its reduction is indicative of successful therapeutic intervention in mitigating remodeling. Tetramethylpyrazine (TMPZ), a bioactive alkaloid derived from Ligusticum chuanxiong Hort., has demonstrated

The long non-coding RNA CRNDE stabilises SIRT1 protein and influences Hedgehog signalling in multiple myeloma.

Multiple myeloma (MM) is a malignancy of immunoglobulin-secreting plasma cells that represents 10% of all hematological cancers and remains essentially incurable, despite recent advances. Long non-coding RNAs (lncRNAs) are an important class of regulatory molecules that have been strongly implicated in the aetiology of MM. Colorectal Neoplasia Differentially Expressed (CRNDE) is one lncRNA that is upregulated in tumor plasma cells of MM patients and contributes to disease progression and outcome. In order to characterise the molecular mechanisms of CRNDE action in MM, here we have carried out

SIRT1 prevents noise-induced hearing loss by enhancing cochlear mitochondrial function.

Exposure to traumatic noise triggers cochlear damage and consequently causes permanent sensorineural hearing loss. However, effective treatment strategies for noise-induced hearing loss (NIHL) are lacking. Sirtuin 1 (SIRT1) is a NAD+-dependent deacetylase that plays a critical role in multiple physiological and pathological events. However, its role in NIHL pathogenesis remains elusive. This study revealed that SIRT1 expression in the cochlea progressively decreases in a mouse model of NIHL. Hair cell-specific knockout of SIRT1 exacerbates the noise-induced loss of outer and inner hair cell sy…

Identification of novel SIRT1 up-regulators using a cell-based high-throughput screening assay.

Sirtuin 1 (SIRT1) has emerged as a promising therapeutic target for many diseases such as neurodegenerative disorders. Resveratrol, a plant polyphenol, is known to induce SIRT1 expression and activity. To excavate active compounds that can increase SIRT1 transcription from FDA-approved drugs, a cell-based SIRT1 up-regulator screening assay was developed. 293A cells were transfected with a SIRT1-promoter-luciferase reporter gene construct, and the stably transfected cell line was selected to establish a specific SIRT1 expression assay in 96-well microplate format using resveratrol as a positive…

Dimethyl fumarate restores Ca2+ dyshomeostasis through activation of the SIRT1 signal to treat nonalcoholic fatty liver disease.

Nonalcoholic fatty liver disease (NAFLD) is characterized by an excessive lipid accumulation in the liver, with a global prevalence of approximately 25 %. While early-stage steatosis is reversible and can be intervened upon, it has the potential to progress to some serious complications, including cirrhosis and even liver cancer. Dimethyl fumarate (DMF), a derivative of fumaric acid shows promise in intervening in certain diseases. However, the precise effect and underlying mechanism of DMF on hepatic steatosis remain unclear. In this study, we demonstrated that DMF mitigates hepatic steatosis
