Proteins from Modern and Ancient Wheat Cultivars: Impact on Immune Cells of Healthy Individuals and Patients with NCGS.

In non-celiac gluten sensitivity (NCGS), the elimination of wheat results in a clear symptom improvement, but gluten has still not been proven as (the sole) trigger. Due to the increase in the prevalence of gluten-related diseases, the breeding of high-performance wheat cultivars is discussed as a trigger. To analyze the immune stimulation and signal pathways, the immune cells of healthy subjects and patients with NCGS were stimulated with gliadins from wheat, and the expression and secretion of interleukin 1ß (IL1ß) and interleukin 6 (IL6) were studied. To determine the impact of wheat breedi

Exogenous expression of barley HvWRKY6 in wheat improves broad-spectrum resistance to leaf rust, Fusarium crown rot, and sharp eyespot.

Systemic acquired resistance (SAR) is a broad-spectrum plant defense phenomena controlled by the salicylic acid receptor NPR1. Key regulators of the SAR signaling pathway showed great potentials to improve crop resistance to various diseases. In our previous investigation, a barley transcription factor gene HvWRKY6 was identified as downstream of NPR1 during SAR. However, the broad-spectrum resistance features and molecular mechanisms of HvWRKY6 remain to be explored. In this study, a transgenic wheat line exogenously expressing HvWRKY6 showed improved resistance to leaf rust, Fusarium crown r

Efficacy of citric acid chelate and Bacillus sp. in amelioration of cadmium and chromium toxicity in wheat.

Heavy metals contamination in agricultural soil is a major issue having drastic effects on plants and human health. To solve this issue, we have formulated and tested a new approach of fusion of inorganic (citric acid chelate) and organic (Bacillus sp.) amelioration methods for heavy metals. The Bacillus sp. was heavy metal tolerant and showed plant growth-promoting characteristics including phosphate solubilization, siderophore production, hydrogen cyanide production, indole acetic acid production, and 1-Aminocyclopropane-1-carboxylate deaminase production. The analysis of data showed that pl

Proteomic modelling of gluten digestion from a physiologically relevant food system: A focus on the digestion of immunogenic peptides from wheat implicated in celiac disease.

Celiac disease is an autoimmune illness activated by gluten peptides produced during gastrointestinal digestion. A simulated in vitro digestion of gluten was conducted to define the profile and kinetic release pattern of immunogenic gluten peptides in a physiologically relevant food matrix. White bread was digested using the INFOGEST in vitro standardised digestion protocol from 0 to 240 min and subsequently analysed by SDS-PAGE, quantitative LC-MS/MS, untargeted LC-MS/MS and ELISA. The release profile of six gluten peptides was defined by quantitative LC-MS/MS; none were detected in the gastr

Comparison of the antifungal effect of undissociated lactic and acetic acid in sourdough bread and in chemically acidified wheat bread.

Sourdough is a very interesting natural preservation system to prolong mould free shelf-life of bread. Numerous studies have reported that the antifungal activity of sourdough is mainly correlated with the presence of lactic (LA) and acetic acid (AA), but very few information is available on the effect of undissociated acid concentrations in the aqueous phase of bread (CHA; mmole/L). This study was conducted to provide additional information about the mode of action of the acids in sourdough bread, enabling a better shelf-life prediction. This study was divided into two parts. In part 1, three

How microwave treatment of gluten affects its toxicity for celiac patients? A study on the effect of microwaves on the structure, conformation, functionality and immunogenicity of gluten.

The microwave heating of wheat kernels, flour, and gluten, has attracted attention lately because it has been claimed to abolish gluten toxicity for celiac patients. Nevertheless, contradictory results have been reported regarding the effect on gluten celiac-immunotoxicity. In order to better understand the effect of the microwave treatment on gluten structure, conformation, functionality and celiac-immunotoxicity, a central composite design with two factors, power level, and treatment time, was used to investigate a possible quadratic and interaction effects between both factors. Coeliac disease.

Coeliac disease is an immune-mediated enteropathy against dietary gluten present in wheat, rye and barley and is one of the most common lifelong food-related disorders worldwide. Coeliac disease is also considered to be a systemic disorder characterized by a variable combination of gluten-related signs and symptoms and disease-specific antibodies in addition to enteropathy. The ingestion of gluten leads to the generation of harmful gluten peptides, which, in predisposed individuals, can induce adaptive and innate immune responses. The clinical presentation is extremely variable; patients may h

Non-Destructive Determination of Alkylresorcinol (ARs) Content on Wheat Seed Surfaces and Prediction of ARs Content in Whole-Grain Flour.

Alkylresorcinols (ARs) are beneficial for human health and can be used as biomarkers for whole-grain cereal intake. In previous studies, ARs content in whole-grain flour was determined by milling the seeds into powder, thus destroying their structure. In this paper, we adopted a non-destructive extraction approach. Chloroform and ethyl acetate extraction methods were carried out to extract lipids from the seed surface and whole-grain flour of 36 wheat varieties in China, respectively. GC-MS analysis identified chemical compounds in which ARs were the major compounds for all the samples. The av

Rht8 gene as an alternate dwarfing gene in elite Indian spring wheat cultivars.

Optimizing wheat height to maximize yield has been an important aspect which is evident from a successful example of green revolution. Dwarfing genes (Rht) are known for yield gains due to lodging resistance and partitioning of assimilates into ear. The available and commercially exploited sources of dwarfism in Indian spring wheat are Rht1 and Rht2 genes inspite of availability of over 20 dwarfing genes. Rht8 a Gibberellic acid sensitive dwarfing gene is another reduced height gene commercially exploited in some Mediterranean countries. Two F2 populations segregating for Rht1 and Rht8 genes w

Gluten and Functional Abdominal Pain Disorders in Children.

In children, functional gastrointestinal disorders (FGIDs) are common at all ages. Consumption of certain foods, particularly gluten, is frequently associated with the development and persistence of FGIDs and functional abdominal pain disorders (FAPDs) in adults and children. However, this association is not well defined. Even without a diagnosis of celiac disease (CD), some people avoid gluten or wheat in their diet since it has been shown to trigger mostly gastrointestinal symptoms in certain individuals, especially in children. The incidence of conditions such as non-celiac gluten sensitivi

An improved method for studying whole sections of late developing wheat grain.

Whole sections are important for investigation in situ of the development of cell morphology and accumulation of storage substance in cereal kernels. We developed an improved, simple method to obtain good quality whole histological sections of wheat grains during mid- and late-stage (28 and 35 days post anthesis) grain filling. The wheat grains were fixed and dehydrated, cleared in chloroform for 3 days and infiltrated in paraffin at 55° C for 3 - 5 days. The infiltrated grains then were embedded in paraffin wax, softened using diethyl pyrocarbonate (DEPC) water before sectioning. After these

[Non-celiac wheat sensitivity - growing evidence for a wheat-dependent immune-mediated disease].

These past years, double-blinded placebo controlled food challenges in subjects with irritable bowel syndrome (IBS) reporting sensitivity to gluten have identified a group of individuals without celiac disease (CD) in whom symptoms were clearly aggravated by wheat. Not only did the exposure to wheat trigger intestinal symptoms, but also frequently fatigue and a sensation of foggy mind. Recent studies in mice and sensitive subjects suggest an immune activation by various wheat proteins, with a rapid increase of intestinal mucosal permeability and recruitment of intraepithelial lymphocytes. Stability of β-carotene during baking of orange-fleshed sweet potato-wheat composite bread and estimated contribution to vitamin A requirements.

Orange-fleshed sweet potato (OFSP) is known to be a rich source of β-carotene, a precursor of vitamin A and a potential tool for fighting vitamin A deficiency (VAD) in developing countries. OFSP flour was incorporated into wheat flour at 10, 20 and 30% (w/w) substitution levels. The stability of β-carotene during baking and the contribution of OFSP-wheat composite breads to vitamin A requirements were evaluated. The retention of all-trans-β-carotene in breads containing 10, 20 and 30% OFSP flour was 62.7, 71.4 and 83% respectively, after baking. Breads containing 20% and 30% OFSP flour could b

The role of tailored biochar in increasing plant growth, and reducing bioavailability, phytotoxicity, and uptake of heavy metals in contaminated soil.

Microwave-assisted catalytic pyrolysis was investigated using K3PO4 and clinoptilolite to enhance biochar sorption affinity for heavy metals. The performance of resulting biochar samples was characterized through their effects on plant growth, bioavailability, phytotoxicity, and uptake of heavy metals in a sandy soil contaminated with Pb, Ni, and Co. The produced biochars have high cation-exchange capacity (CEC) and surface area, and rich in plant nutrients, which not only reduced heavy metals (Pb, Ni, and Co), bioavailability and phytotoxicity, but also increased plant growth rate by up to 14

Wheat straw biochar-supported nanoscale zerovalent iron for removal of trichloroethylene from groundwater.

This study synthesized the wheat straw biochar-supported nanoscale zerovalent iron (BC-nZVI) via in-situ reduction with NaBH4 and biochar pyrolyzed at 600°C. Wheat straw biochar, as a carrier, significantly enhanced the removal of trichloroethylene (TCE) by nZVI. The pseudo-first-order rate constant of TCE removal by BC-nZVI (1.079 h-1) within 260 min was 1.4 times higher and 539.5 times higher than that of biochar and nZVI, respectively. TCE was 79% dechlorinated by BC-nZVI within 15 h, but only 11% dechlorinated by unsupported nZVI, and no TCE dechlorination occurred with unmodified biochar.

An alternative method based on enzymatic fat hydrolysis to quantify volatile compounds in wheat bread crumb.

An alternative method to quantify 40 volatile compounds in wheat bread crumb is proposed. It consists of a Soxhlet extraction with a mixture of dichloromethane and diethyl ether containing lipases and a subsequent concentration with Vigreux column. It is the first time that lipases are added to transform the fat into free fatty acids and glycerol, which elute at the end of the chromatogram after the analytes, avoiding problems in the chromatography due to fat residues, such as dirtiness in the injector, column clogging or overlapping peaks. The extract is most easily analysed by GC/MS, using a

Application of the Prunus spp. Cyanide Seed Defense System onto Wheat: Reduced Insect Feeding and Field Growth Tests.

Many crops are ill-protected against insect pests during storage. To protect cereal grains from herbivores during storage, pesticides are often applied. While pesticides have an undoubtable functionality, increasing concerns are arising about their application. In the present study, we investigated a bioinspired cyanogenic grain coating with amygdalin as cyanogenic precursor mimicking the feeding-triggered release of hydrogen cyanide (HCN) found for example in bitter almonds. The multilayer coating consisted of biodegradable polylactic acid with individual layers containing amygdalin or β-gluc

Label-free SPR detection of gluten peptides in urine for non-invasive celiac disease follow-up.

Motivated by the necessity of new and efficient methods for dietary gluten control of celiac patients, we have developed a simple and highly sensitive SPR biosensor for the detection of gluten peptides in urine. The sensing methodology enables rapid and label-free quantification of the gluten immunogenic peptides (GIP) by using G12 mAb. The overall performance of the biosensor has been in-depth optimized and evaluated in terms of sensitivity, selectivity and reproducibility, reaching a limit of detection of 0.33 ng mL(-1). Besides, the robustness and stability of the methodology permit the con

Quantum dot based rapid tests for zearalenone detection.

Three different kinds of immunosorbent assays with luminescence detection were developed for the determination of zearalenone (ZEN), a secondary toxic metabolite of Fusarium fungi. CdSe/ZnS core/shell quantum dots (QDs) were used as a label in quantitative micro-well plate immunoassays (fluorescent-labeled immunosorbent assay, FLISA) and in qualitative column test methods. As carriers for QD-based column tests, sepharose gel (for covalent binding of antibody) and polyethylene frits (for physical absorption of antibody) were used and compared. The application of QDs as a label resulted in a fou
