Persistent organic pollutants and metals in the freshwater biota of the Canadian Subarctic and Arctic: an overview.

Over 1999-2002, an extensive series of contaminant studies was conducted on freshwater biota of Canada's Arctic and Subarctic regions. The majority of inorganic contaminant studies focused on mercury and fish. While mercury concentrations were low in benthic feeding fish such as whitefish, predatory fish such as lake trout, pike, and walleye frequently had mercury levels which exceeded 0.2 mug/g, the consumption guideline for frequent consumers of fish, and 0.5 microg/g, the guideline for the commercial use of fish. Numerous consumption advisories were issued for lakes along the Mackenzie Rive…

Natural toxic impact and thyroid signalling interplay orchestrates riverine adaptive divergence of salmonid fish.

Adaptive radiation in fishes has been actively investigated over the last decades. Along with numerous well-studied cases of lacustrine radiation, some examples of riverine sympatric divergence have been recently discovered. In contrast to the lakes, the riverine conditions do not provide evident stability in the ecological gradients. Consequently, external factors triggering the radiation, as well as developmental mechanisms underpinning it, remain unclear. Herein, we present the comprehensive study of external and internal drivers of the riverine adaptive divergence of the salmonid fish Salv

Aquatic ecological risks due to cyanide releases from biomass burning.

Aquatic toxicity due to the creation and mobilization of chemical constituents by fire has been little studied, despite reports of post-fire fish kills attributed to unspecified pyrogenic toxicants. We examined releases of cyanides from biomass burning and their effect on surface runoff water. In laboratory test burns, available cyanide concentrations in leachate from residual ash were much higher than in leachate from partially burned and unburned fuel and were similar to or higher than the 96-h median lethal concentration (LC50) for rainbow trout (45 microg/l). Free cyanide concentrations in…

[Epidemiology of infectious hematopoietic necrosis (IHN) of salmonid fish in France: study of the course of natural infection by combined use of viral and seroneutralization test and eradication attempts].

Infectious haematopoietic necrosis (IHN), a rhabdoviral infection of salmonid fish, was considered to be an exotic disease in Europe until i … minations and 1,782 serum neutralization tests. The presence of the IHN virus was detected in the 7 trout farm fish populations that were located in 5 regions, one of which was situated 600 km from the place where the first isolation of IHN virus was made. Moreover, 6 out of 7 rainbow trout populations reared in these farms also harboured viral haemorrh …

Secretory IgM regulates gut microbiota homeostasis and metabolism.

The coating of microbiota by secretory immunoglobulins (sIgs) determines which bacteria colonize the gut and influences bacterial metabolism. Previous work has identified sIgA and sIgT as mediators of gut homeostasis. However, sIgM coats a large proportion of the gut microbiota in humans and teleost fish, thus suggesting a conserved role of sIgM in microbiota homeostasis. Here, to investigate this hypothesis, we used the teleost rainbow trout as a model system. Depletion of IgM from trout resulted in severe microbiota-dependent gut tissue damage, body weight loss, bacterial translocation and g…

Is there a plasma membrane-located anion-sensitive ATPase?

A study of the intracellular localization of HCO-3-stimulated, SCN--inhibited magnesium-dependent ATPase was performed in gill tissue of the rainbow trout (Salmo irideus), rabbit kidney and rabbit gastric mucosa. Tissue homogenates were subjected to centrifugal fractionation, and the microsomal (60 min 100 000 X g) and light mitochondrial (20 min 20 000 X g) fractions were further fractionated by density gradient centrifugation. Subfractions were characterized by marker enzyme assays and electron microscopic observation. In trout gill indications for an exclusively mitochondrial localization w…

Limited species differences in estrogen receptor alpha-medicated reporter gene transactivation by xenoestrogens.

Estrogen receptors (ERs) play an important role in estrogen function. However, it is well known that there are species differences in amino acid sequences of the ligand binding domains. Here, we report on the analysis of species differences in ER-dependent transactivation with some chemicals using reporter gene assays. Full-length ER cDNAs from human, rat, chicken, alligator (Caiman), whiptail lizard, African clawed frog and rainbow trout were prepared from hepatic mRNA by the RT-PCR method and inserted into expression plasmids. Both expression and reporter plasmids were transiently transfecte…

Action of gamma-aminobutyric acid (GABA) in the isolated photosensory pineal organ.

The effect of GABA (gamma-aminobutyric acid), its agonists (muscimol, baclofen) and antagonist (bicuculline) on pineal ganglion cells of the luminosity type were studied in the isolated, superfused pineal organ of the rainbow trout, Oncorhynchus mykiss. Extracellular recordings revealed that GABA added through the superfusion medium caused a clear alteration of the activity of projecting neurons, which transmit luminosity responses to the brain. Spontaneous discharges of ganglion cells were predominantly suppressed by GABA (33 neurons out of 48), but 10 neurons were clearly excited. Similar ef…

New salmonid hosts for Tenacibaculum species: Expansion of tenacibaculosis in Chilean aquaculture.

The success and sustainability of Chilean aquaculture largely depends on the control of endemic and emerging pathogens, including several species of the genus Tenacibaculum. Tenacibaculum dicentrarchi and "Tenacibaculum finnmarkense" have been detected and confirmed in Chilean Atlantic salmon (Salmo salar). However, no outbreaks of tenacibaculosis in rainbow trout (Oncorhynchus mykiss) or coho salmon (Oncorhynchus kisutch) have been reported, either in Chile or globally. The aims of this study were to determine whether the mortalities recorded for rainbow trout and coho salmon from five marine…

Molecular characterization and pathogenicity of Klebsiella pneumoniae species complex associated with mass mortality of Oncorhynchus mykiss in the Ladakh trans-himalayas.

Bacterial diseases pose a significant threat to the entire aquaculture system, both in cold and warm-water environments. The Himalayan region has been widely utilised for trout farming. However, this region is presently under tremendous stress due to the outbreak of several diseases, which are responsible for a lot of mortality and have resulted in significant economic loss. In the present study, Klebsiella pneumoniae belonging to the K. pneumoniae species complex was isolated from diseased rainbow trout Oncorhynchus mykiss cultured in the trans-Himalayan region of Ladakh. Infected fish were i…

Antigen Uptake during Different Life Stages of Zebrafish (Danio rerio) Using a GFP-Tagged Yersinia ruckeri.

Immersion-vaccines (bacterins) are routinely used for aquacultured rainbow trout to protect against Yersinia ruckeri (Yr). During immersion vaccination, rainbow trout take up and process the antigens, which induce protection. The zebrafish was used as a model organism to study uptake mechanisms and subsequent antigen transport in fish. A genetically modified Yr was developed to constitutively express green fluorescent protein (GFP) and was used for bacterin production. Larval, juvenile and adult transparent zebrafish (tra:nac mutant) received a bath in the bacterin for up to 30 minutes. Sample…

Co-occurrence of heterogeneous Flavobacterium psychrophilum isolates within the same Chilean farm and during the same infectious outbreak.

Flavobacterium psychrophilum is a pathogenic bacterium affecting Chilean salmonid farms. High antigenic and genetic diversity exists among Chilean F. psychrophilum isolates, but the distribution thereof among farms is poorly understood. These epidemiological data are key for developing isolate-specific vaccines. The present study isolated F. psychrophilum in diseased Atlantic salmon (Salmo salar) and rainbow trout (Oncorhynchus mykiss) from five freshwater farms between 2018 and 2019. Each farm only raised one salmonid species and was geographically separated from and did not share culturing w…

Ambient Developmental Temperatures and Maternal Thyroid Supply Promote Early Life Diversification in Salmonids.

The interplay between ambient temperature and thyroid signaling controls the morphogenesis of poikilotherms in the early stages of life. These two metabolic conditioning factors are crucial for the divergence of fish morphs undergoing adaptive diversification. Using Salvelinus malma, a model salmonid species with high phenotypic plasticity and a capacity for adaptive radiation, we conducted a series of experiments by rearing sympatric large predatory, mid-sized generalized and dwarf insectivorous morphs from both lacustrine and riverine environments. First, we compared the developmental rates

Phylogenomic analysis of the Lake Kronotskoe species flock of Dolly Varden charr reveals genetic and developmental signatures of sympatric radiation.

Recent adaptive radiations provide experimental opportunities to parse the relationship between genomic variation and the origins of distinct phenotypes. Sympatric radiations of the charr complex (genus Salvelinus) present a trove for phylogenetic analyses as charrs have repeatedly diversified into multiple morphs with distinct feeding specializations. However, charr species flocks normally comprise only two to three lineages. Dolly Varden charr inhabiting Lake Kronotskoe represent the most extensive radiation described for the genus, containing at least seven lineages, each with defining morp

Darkfield illumination improves microscopic detection of metals in Timm's stained tissue.

Deposits of trace or toxic metals can be quickly identified by light microscopical surveys of tissue sections stained for metals by variants of Timm's silver enhancement method. The present work shows that the small, isolated silver grains that label isolated deposits of metal in tissue are undetectable in brightfield light microscopy but are easily detected in darkfield microscopy. Darkfield illumination is therefore recommended for improving the detection of trace or toxic metals in tissue.

Lectin identification of olfactory receptor neuron subclasses with segregated central projections.

Our previous studies have demonstrated that the primary olfactory projection in rainbow trout is organized nontopographically; the pattern of termination of olfactory axons in the olfactory bulb is unrelated to the distribution of their cell bodies in the olfactory mucosa. In the present research we have further characterized the organization of this projection by examining the lectin-binding properties of olfactory receptor neurons. The results indicate that in trout, as in mammals, populations of olfactory receptor neurons differ significantly from one another in their carbohydrate "signatur…

[Control of respiration in teleostean fishes: existence of chemoreceptors, physiologically analogous to those of higher vertebrates].

Injection of NaCN or lobeline into the ventral aorta of the trout or carp provokes hyperventilation and bradycardia within 5 seconds, responses which are the weaker the higher the oxygen tension of the ambient water. The reactions, precisely similar to the chemoreflexes of higher vertebrates, suggest that these fish also possess arterial chemoreceptors.
