To evaluate the merit of antigen detection assays as a tool to monitor the efficacy of chemotherapy for lymphatic filariasis, we serially measured antigen levels in sera from jirds infected with Brugia malayi and from humans with bancroftian filariasis. Antigenemia was detected in all animals with parasitologically proven infection and was present in jirds with prepatent or occult filariasis. Antigen levels correlated with worm burdens, and progressively declined in drug-cured animals. Treatment with diethylcarbamazine (DEC) triggered a transient increase in serum levels of filarial antigens b

Lymphocyte proliferative responses to homologous and heterologous filarial antigens, mitogens and purified protein derivatives (PPD) were analysed in a group of 37 subjects from an area endemic for bancroftian filarial infection. The majority of the subjects without any clinical or parasitological evidence of filariasis (endemic normals) reacted with homologous microfilarial antigens only. Non-treated patients with patent microfilaraemia, did not respond to homologous of heterologous microfilarial antigens. In contrast, diethylcarbamazine (DEC)-treated microfilaraemic patients, reacted with ho

Two patients with chyluria who were successfully treated by surgical disconnection of the lymphatics from the renal pelvis are reported. One patient also had an associated glomerulonephritis, possibly secondary to filariasis. Although glomerulonephritis associated with filariasis has been reported previously the details of renal biopsies in man have included only light microscopic findings. In this patient immunofluorescent and electron microscopic studies showed mesangial deposits of immunoglobulins and complement which suggests that glomerulonephritis in patients with filariasis may be an im

For epidemiological studies of infectious diseases, pathogen-specific antibody levels in an area give us essential and appropriate informati … , and these are important infections included in NTDs, caused, for example, by Leishmania donovani, Wuchereria bancrofti, Schistosoma japonicum, Paragonimus westermani, Echinococcus granulosus, Echinococcus multilocularis, Strongyloides stercoralis, and Opisthorchis viverrini. The easy and safe urine surveillance system might be an admirable tool for future …

Lymphatic filariasis is an infection transmitted by blood-sucking mosquitoes with filarial nematodes of the species Wuchereria bancrofti, Brugia malayi und B. timori . It is prevalent in tropical countries throughout the world, with more than 60 million people infected and more than 1 billion living in areas with the risk of transmission. Worm larvae with a length of less than 1 mm are transmitted by mosquitoes, develop in human lymphatic tissue to adult worms with a length of 7-10 cm, live in the human body for up to 10 years and produce millions of microfilariae, which can be transmitted fur…

BACKGROUND: There is compelling evidence that not only do anti-filarials significantly reduce larval forms, but that host immune responses also contribute to the clearance of filarial parasites; however, the underlying mechanisms have not been fully elucidated.
MAIN TEXT: Filarial infections caused by Wuchereria bancrofti and Brugia species (lymphatic filariasis) and Onchocerca volvulus (onchocerciasis) affect almost 200 million individuals worldwide and pose major public health challenges in endemic regions. Indeed, the collective disability-adjusted life years for both infections is 3.3 mil…

Lymphatic filariasis is a global health problem endemic in Nigeria. Peripheral blood smears are frequently negative for microfilariae in a s … maturia. However, fine needle aspiration cytology performed on the swelling showed microfilariae of Wuchereria bancrofti for which appropriate treatment was instituted with excellent outcome. This case demonstrates that lymphatic filariasis should be considered in the differential diagnosis of lymph node swellings in the groin and that aspiration cytology ma …

Lymphatic filariasis is a parasitic infection that causes a devastating public health and socioeconomic burden with an estimated infection of over 120 million individuals worldwide. The infection is caused by three closely related nematode parasites, namely, Wuchereria bancrofti, Brugia malayi, and B. timori, which are transmitted to human through mosquitoes of Anopheles, Culex, and Aedes genera. The species have many ecological variants and are diversified in terms of their genetic fingerprint. The rapid spread of the disease and the genetic diversification cause the lymphatic filarial parasi…

Filarial infections remain a major public health and socio-economic problem across the tropics, despite considerable effort to reduce diseas … parasites including onchocerciasis (river blindness, Onchocerca volvulus) and lymphatic filariasis (Wuchereria bancrofti or Brugia malayi) is founded on evidence from both humans and animal models for the development of protective immunity. Nonetheless, enormous challenges need to be faced in terms of overcoming parasite-induced suppression without inducing …

Polymerase chain reaction based methods are promising tools for the monitoring and evaluation of the Global Program for the Elimination of Lymphatic Filariasis. The currently available PCR methods do not differentiate the DNA of Wuchereria bancrofti or Brugia malayi by a single PCR and hence are cumbersome. Therefore, we designed a single step PCR strategy for differentiating Bancroftian infection from Brugian infection based on a newly identified gene from the W. bancrofti genome, abundant larval transcript-2 (alt-2), which is abundantly expressed. The difference in PCR product sizes generate…

Lymphocele is a well-known complication of renal transplantation. Presenting symptoms are nonspecific; most patients are entirely asymptomatic. Herein, we have reported a case of lymphocele due to an asymptomatic lymphatic Wuchereria bancrofti filariasis with deterioration of graft function. A 53-year-old man with end-stage renal disease secondary to vascular disease was admitted 40 days after transplantation with vague, isolated abdominal pain. An abdomen and pelvis ultrasound examination demonstrated a cystic structure in the renal hilus. Graft function deteriorated, so the patient underwent…

The relationship between HIV, lymphatic filariasis, malaria (Plasmodium falciparum) and intestinal helminths (Ascaris lumbricoides, Trichuris trichiura and hookworm) was assessed in a cross-sectional study conducted in 2002 among 907 adults in Tanga Region, Tanzania. Overall prevalences were 7.9% for HIV, 43.5% for Wuchereria bancrofti-specific circulating filarial antigen (CFA), 12.3% for P. falciparum, 1.2% for A. lumbricoides, 7.1% for T. trichiura and 75.7% for hookworm. Anaemia was assessed separately for males and females and was found to be more prevalent among females (58.8%) than male…

BACKGROUND & OBJECTIVES: Hydrocele is the most common clinical manifestation in males infected with Wuchereria bancrofti. IgE response to a filarial allergen Sd30 was evaluated in hydrocele patients living in a W. bancrofti endemic region of Orissa, India.
METHODS: IgE levels to Sd30, an antigenic fraction isolated from Setaria digitata, were determined by ELISA in sera from patients of filariasis (n = 93). IgG and IgM levels were also determined.
RESULTS: It was observed that microfilaraemic patients with hydrocele (n = 16) exhibited considerably reduced (P < 0.01) IgE levels in comparison …

Infections with Dracunculus medinensis frequently occur in the same geographical area as infections with Onchocerca volvulus and Wuchereria bancrofti. This study analysed the significance of D. medinensis infections for the specificity and sensitivity of available tests for antibody-based diagnosis of onchocerciasis (using individual recombinant clones OV-10, OV-11 and OV-16, and the OV-7/OV-10/OV-16 tri-cocktail, in an enzyme-linked immunosorbent assay) and for circulating antigen-based diagnosis of bancroftian filariasis (using the TropBio and the ICT card tests). Some immunological cross-re…
