Lack of effect of the peptide pyro-Glu-His-Gly-OH on food consumption in mice and rats.

1. Single subcutaneous injections (0.5-10 mg/kg) or repeated oral administration (5 mg/kg, twice daily for 3 or 5 days) of the proposed anorexigenic peptide pyro-GLU-HIS-GLY-OH did not affect food intake in the mouse. 2. Single intravenous injections (0.05-1 mg/kg) or daily intravenous injections (0.3-10 mg/kg, for 5 days) of pyro-GLU-HIS-GLY-OH did not affect food intake in the rat. 3. These results, together with results obtained recently by other workers, indicate that pyro-GLU-HIS-GLY-OH does not have anorexigenic properties in rodents.

The effect of viloxazine on drug-induced inhibition of food intake in the rat.

In male Wistar rats trained to eat their normal daily dietary requirement in a restricted 2 h period, dose-dependent decreases in food consumption were produced by fenfluramine, tiflorex, mazindol and amphetamine. The antidepressant drug viloxazine (Vivalan) alone did not alter food intake significantly, nor did the drug prevent the inhibitory effects of either mazindol or amphetamine. However, complete prevention of the inhibitory effect of fenfluramine was achieved with 7.5 mg kg-1 viloxazine, while 40 mg kg-1 viloxazine similarly prevented the anorectic action of tiflorex. An interaction in

[Psychopharmacological profile of a new dopaminergic agonist, RU 24213 (author's transl)].

The psychopharmacological properties of RU 24213 were compared to those of other dopaminergic agonists (apomorphine, dexamphetamine, bromocriptine and L-dopa) in various behavioural tests. In naive mice the drug reduced the locomotor hyperactivity in the primary exploratory phase and produced stimulation in the subsequent stabilized activity period. In rats it provoked dose-related stereotypies, specially gnawing and sniffing. It delayed the cataleptic state induced by prochlorperazine without affecting its intensity. In animals unilaterally lesioned with 6-OHDA in the nigro-striatal pathway,

The roles of brain noradrenaline and dopamine in the anorectic activity of diethylpropion in rats: a comparison with d-amphetamine.

The anorectic activity of diethylpropion and d-amphetamine was studied in rats subjected to various treatments known to affect brain monoamines. The effect of diethylpropion, like that of d-amphetamine, was completely prevented by a lesion of the ventral noradrenergic bundle, which selectively decreases brain noradrenaline, but was not significantly modified in desipramine pretreated rats by an intraventricular injection of 6-hydroxydopamine, a condition decreasing only dopamine. Pretreatment with penfluridol significantly reduced the effect of d-amphetamine but not that of diethylpropion. A n

Nutritional Knowledge, Dietary Habits, and Nutritional Status of Patients with Chronic Kidney Disease According to Disease Stage.

Background/Objectives: Appropriate nutritional management constitutes one of the key elements of conservative treatment and renal replacement therapy in patients with chronic kidney disease (CKD). The level of patients' nutritional knowledge may significantly influence adherence to dietary recommendations, the rate of disease progression, and the frequency of complications. The aim of this study was to assess the level of nutritional knowledge, dietary habits, adherence to dietary recommendations, and nutritional status of patients with CKD according to disease stage. Methods: This cross-secti

Metabolic and psychiatric effects of acyl coenzyme A binding protein (ACBP)/diazepam binding inhibitor (DBI).

Acyl coenzyme A binding protein (ACBP), also known as diazepam binding inhibitor (DBI) is a multifunctional protein with an intracellular action (as ACBP), as well as with an extracellular role (as DBI). The plasma levels of soluble ACBP/DBI are elevated in human obesity and reduced in anorexia nervosa. Accumulating evidence indicates that genetic or antibody-mediated neutralization of ACBP/DBI has anorexigenic effects, thus inhibiting food intake and inducing lipo-catabolic reactions in mice. A number of anorexiants have been withdrawn from clinical development because of their side effects i

[Ten questions on the treatment of obesity: from dieting to surgery].

Advantages and risk of weight reduction achieved by combining diets or unbalanced diets with restriction of carbohydrates or fat are discussed. Nutriments differ according to their density, their flavour, their satiating, and their thermogenic effect. Food is also a source of pleasure and a remedy to frustration. One of the problems of dieting is that most patients regain the weight they have lost due to a hypometabolism secondary to caloric restriction. Only physical exercise together with behavior therapy are able to slow down this weight regain. The indication of drug treatment with orlista

Vagotomy attenuates behavioural effects of interleukin-1 injected peripherally but not centrally.

Peripheral and central injections of recombinant rat interleukin-1 beta (IL-1 beta) have been shown to decrease social exploration in rats. To test the involvement of vagal afferents in the communication between the immune system and the brain, sham-operated and vagotomized rats were injected peripherally or centrally with physiological saline or IL-1 beta 4 weeks after surgery. Vagotomy attenuated the depression in social exploration induced by i.p. administration of IL-1 beta (15 micrograms) but did not alter the behaviour-depressing effects of an intracerebroventricular (i.c.v.) injection o

Human satietin: rapid development of tolerance and its specificity to feeding behavior in rats.

Semipurified human satietin (SP hSAT) significantly (p less than 0.05) reduced food intake for each of five consecutive days when injected intraperitoneally (2 mg/kg/day). Tolerance developed rapidly during the second and third days of injection but food intake of those animals receiving the highest doses of Sp hSAT (1 and 2 mg/kg/day) remained significantly below (p less than 0.05) that of the saline-injected animals. Food intake of rats receiving doses of 0.5 and 0.25 mg/kg/day Sp hSAT was below that of the saline-treated group for the first two days of injection (p less than 0.05) but not t

Role of thyroid hormones in an abnormal grooming behavior in thyroidectomized cats and cats with pontile lesions.

Thyroidectomized cats and cats with pontile lesions exhibit the same abnormal grooming behavior. Investigations were conducted to determine whether the abnormal grooming behavior in cats with pontile lesions was attributable to a hypothyroid condition. Administration of thyroid hormones abolished the abnormal behavior of thyroidectomized cats but was without effect in cats with pontile lesions. Studies of brain and blood levels of thyroid hormones in cats with pontile lesions failed to detect any differences from normal cats, and normal effects of thyroid hormones on evoked potentials to light

From stress cytokine to drug target: The translational landscape of growth differentiation factor 15.

Growth differentiation factor 15 (GDF15) is a stress-responsive cytokine that translates cellular injury, metabolic imbalance, and disease burden into systemic physiological responses. Discovery of its hindbrain-restricted receptor, glial cell line-derived neurotrophic factor family receptor α-like (GFRAL) and cosignaling partner rearranged during transfection (RET), established a dedicated neuroendocrine axis (GDF15-GFRAL-RET) linking stress to appetite suppression, nausea, and autonomic effects on energy expenditure. Initially regarded as a passive disease biomarker, GDF15 is now recognized

Setmelanotide-mediated MC4R activation improves hypothalamic obesity via CaMKK2/AMPK pathways.

Hypothalamic obesity (HO) is a disabling disease caused by central nervous system (CNS) damage due to neurosurgery, trauma, or tumors, especially in hypothalamus. The pathological mechanism of its neural circuits is still unclear, and there is currently no corresponding drug due to the complex etiology. G protein-coupled receptors (GPCRs) regulate neural function in many CNS diseases. Among them, melanocortin 4 receptor (MC4R) regulate metabolism and appetite in the hypothalamus. Setmelanotide, an MC4R agonist, has demonstrated anti-obesity effects in genetic forms of obesity; however, its eff

Investigating ghrelin and its isoforms in eating disorders: a network meta-analysis.

BACKGROUND: Eating disorders (EDs) are serious psychiatric conditions disorders characterized by impairments in appetite regulation and energy balance. Among the biological factors implicated in their pathophysiology, the orexigenic hormone ghrelin and its isoforms (acyl-ghrelin [AG] and desacyl-ghrelin [DAG]) have been shown to play a key role in appetite regulation and may contribute to the maintenance of disordered eating behaviors. METHODS: A systematic review of existing literature and meta-analysis were conducted, including 80 studies examining fasting blood levels of ghrelin, AG, and D

Serum Levels of Hormones Regulating Appetite in Patients with Fetal Alcohol Spectrum Disorders.

Prenatal alcohol exposure is the cause of impaired growth and a wide range of developmental and behavioral disorders in the child. Improper eating patterns are commonly associated with fetal alcohol spectrum disorders (FASD) and may contribute to poor nutrition and growth restriction. To date, there have been only a few studies investigating the hormonal regulation of appetite in patients with FASD. We analyzed the levels of neuropeptide Y (NPY), Agouti signaling protein (ASP), alpha-melanocyte-stimulating hormone (α-MSH), and kisspeptin (KISS1) in 57 patients with FASD and 23 healthy controls
