Sex hormone-binding globulin in osteoporosis.

Sex hormone-binding globulin (SHBG) is a plasma glycoprotein that binds with high affinity to sex steroids, most notably 5alpha-dihydrotestosterone, testosterone, and 17beta-estradiol, thereby regulating their bioavailability and access to target cells. SHBG modulates the sex-steroid signaling system by binding to a specific membrane receptor (SHBG-R). Plasma SHBG levels vary in health and disease due to the effects of multiple regulation factors (age, body weight, sex steroids, insulin, and others). SHBG is involved in a number of diseases, including osteoporosis. Several studies found an inv

Mechanical properties of L929 cells measured by atomic force microscopy: effects of anticytoskeletal drugs and membrane crosslinking.

To shed light on the architecture of the cytoskeleton, we used the atomic force microscope (AFM) to measure the elasticity, viscoelasticity, and plasticity of L929 cells. The initial elastic response (Young's modulus approximately 4,000 Pa) of the cells to an applied force was followed by a slow compression of the cytoskeleton (tau 1/2 approximately equal to 10 s). When force application was terminated, the cytoskeleton underwent a sudden partial decompression and a subsequent slow, incomplete recovery. The role of the cytoskeletal elements in cell mechanics was accessed in AFM measurements ca

Pulmonary immune cells in health and disease: polymorphonuclear neutrophils.

The pulmonary vasculature represents the largest reservoir of polymorphonuclear neutrophils (PMNs) in the human body. This is in striking contrast with the paucity of PMNs present in the normal airways and alveoli. However, the respiratory tract constitutes an easy access for microorganisms and particles present in inhaled air and, therefore, efficacious defence mechanisms are required. When the mucociliary clearance and the alveolar macrophages are over-whelmed, the rapid recruitment of PMNs from the lung vasculature appears to be a crucial response of the host against the pathogens. The regu

Pathogenesis versus symbiosis: a dual perspective on plant-microbe interactions.

Plants function as integral components of soil ecosystems, embedded within diverse microbial consortia that engage in continuous chemical and informational exchange. This molecular dialog governs how plants distinguish harmful, neutral, and beneficial microbes, transforming the rhizosphere into a dynamic signaling interface. Microbial interactions operate along a chemical continuum in which metabolites, volatiles, peptides, and other small molecules act across aqueous, lipidic, and chemically heterogeneous microenvironments to shape recognition and response. The physicochemical organization of

Remote-Controlled Genetic Circuits: Light, Ultrasound, and Magnetic Fields for Programmable Gene Regulation.

Remote-controlled genetic circuits use externally applied physical signals to regulate gene expression and cellular functions with spatial and temporal precision. Light, ultrasound, and magnetic fields provide complementary advantages in controllability, tissue accessibility, and biological compatibility, but their performance depends on both physical-field propagation and efficient conversion of external energy into intracellular signals and predictable gene-regulatory outputs. This review presents a unified framework of physical transduction, biological sensors, signal processors, and geneti

Understanding environmental adaptation in intermediate wheatgrass.

Intermediate wheatgrass (IWG; Thinopyrum intermedium) is a perennial grain crop that offers many environmental benefits through extensive root systems in addition to food production. IWG's distribution across broad environmental gradients in Eurasia provides a model system for studying climate adaptation genetics essential for developing climate-resilient varieties, and exploring this can help provide opportunities for allele mining and population improvement for breeding programs. Genome-environment association methods were used to explore 296 georeferenced accessions genotyped with 17,797 si

Amino-functionalized MIL-101(Fe)-confined Au nanostructures for advanced electrochemical detection of H2O2 in Alzheimer's disease research.

Oxidative stress is a fundamental pathological contributor to Alzheimer's disease and is closely linked to amyloid-beta-mediated toxicity, tau dysregulation, mitochondrial impairment, and progressive neuronal degeneration. Among the associated reactive oxygen species, hydrogen peroxide (H2O2) is a mechanistically relevant redox intermediate and an important oxidative stress biomarker. Herein, an Au@NH2-MIL-101(Fe) hybrid was rationally engineered as an electrocatalytically active sensing interface for H2O2 determination. The hybrid was constructed by integrating conductive Au nanodomains with

Cell dynamics on energy landscape: Comparing attractor detection in Boolean network and diffusion-based models under in silico gene perturbations.

Steady-state attractor detection differs fundamentally between discrete Boolean network (BN) and continuous diffusion-based models because they operate on different state spaces and exhibit distinct dynamic behaviors. Here, we propose cell dynamics on energy landscape (CellLand) to systematically compare BN and diffusion-based approaches for quantifying cellular dynamics on the energy landscape, with a particular focus on attractor identification under in silico gene perturbations. Although the energy landscape provides a unifying conceptual framework, BN-based models face specific challenges,

Oxygen-driven ethanolamine pretreatment for one-pot functional recovery of in situ aminated lignin and efficient enzymatic glucose production.

The efficient pretreatment of waste biomass to achieve effective carbohydrate conversion and high-value lignin valorization has consistently been regarded as one of the most critical challenges in the field of biomass refining. In this study, an oxygen-driven ethanolamine (OE) pretreatment strategy was developed using reed as the feedstock, enabling efficient carbohydrate recovery while simultaneously driving lignin structural transformation and in situ nitrogen functionalization via a coupled oxidative ammonolysis pathway. Under the selected pretreatment conditions (85 wt% ETA, 1 MPa O2, 170

Time to check up: Retirement and cancer screening in Australia.

Cancer screening is central to early detection and healthy ageing, yet evidence on how major life transitions shape screening uptake remains limited. Using four waves of the Household, Income and Labour Dynamics in Australia (HILDA) Survey covering 2009, 2013, 2017, and 2021, with 18,875 person-wave observations from 8343 individuals aged 50-75, we estimate the causal effect of retirement on cancer screening using an instrumental-variable design that exploits discontinuities in Age Pension eligibility. Retirement significantly increases participation in organised cancer screening, with the cle

Monitoring Autonomic Tone During Spinal Cord Neuromodulation Using Wearable AURIS Sensor.

The transition of bioelectronic medicine to clinical use is currently limited by a lack of non-invasive sensors capable of measuring autonomic tone during active neuromodulation. Conventional monitoring modalities, such as mean arterial pressure (MAP) and Ag/AgCl chest electrodes, are often invasive, cumbersome, or susceptible to motion artifacts. Here, we present a novel framework employing an in-ear sensor (AURIS) to continuously monitor heart rate variability (HRV) during therapeutic neuromodulation. These sensors utilize a polydimethylsiloxane (PDMS) substrate to ensure biocompatibility an

Decoding cardiac homeostasis and injury: the evolving landscape of spatial transcriptomics.

The heart is a structurally complex organ where function is intimately tied to the precise spatial organization of diverse cell types. While single-cell RNA sequencing (scRNA-seq) has revolutionized cardiovascular research by providing a high-resolution "parts list" of the heart, the requisite tissue dissociation destroys the critical spatial context of intercellular communication and microenvironmental niches. Spatial transcriptomics (ST) has emerged as a transformative technology that bridges this gap, enabling the mapping of gene expression back to its histological coordinates. This review

Genetic control of morphological transitions in a coacervating protein template.

Nature routinely exploits liquid-liquid phase separation (LLPS) of proteins to control the assembly and mineralization of hybrid materials. Here, we show that fusion of the Car9 silica-binding peptide to an elastin-like polypeptide (ELP) yields temperature- and sequence-programmable soft matter templates for the synthesis of silicified architectures ranging in size from nanometers to micrometers. Specifically, we demonstrate unprecedented control over the diameter of silica nanoparticles (SiNP) in the 30-60 nm range with 4 nm precision, show that a single arginine residue (R4) in the Car9 sequ

Thickness-directed oxide-templated conversion of sputtered tungsten films into WS2 nanotubes.

Controlling the topology of curved transition-metal dichalcogenides remains challenging because shell formation, precursor conversion and hollowing occur simultaneously during growth. Here, we show that the thickness of sputtered tungsten (W) films provides a direct handle for selecting WS2 nanotube topology during charcoal-assisted oxide-template sulfurization. Varying the initial W thickness from 1 to 100 nm drives a transition from laterally extended WS2-like domains to hollow nanotubes and, finally, to retained-core nanotubes exhibiting shell-front mismatch and spiral-like shell wrapping.

Electrochemical Vertical Flow Immunoassay for Fast and Decentralized Determination of Stroke-Related Biomarker Glial Fibrillary Acidic Protein.

Rapid identification of stroke types remains a major clinical challenge, as current diagnostic methods often rely on neuroimaging, which is time-consuming and not always accessible in pre-hospital settings or remote zones. Glial fibrillary acidic protein (GFAP) has emerged as a promising biomarker for distinguishing hemorrhagic from ischemic stroke, paving the way for rapid, quantitative, and portable detection platforms. Here, we report an electrochemical vertical flow immunoassay (eVFIA) as a point-of-care device for the on-site determination of GFAP. The device combines a multilayer paper-b

Deterministic Evolution of Aptamers via a Microfluidic-Integrated Robotic Platform Using Complex Exosomes as Targets.

Exosome (EXO) membrane proteins are attractive biomarkers for liquid biopsy, yet their heterogeneity makes it difficult to develop reliable antibody-based recognition reagents. Aptamers provide high-affinity and highly specific alternatives through the systematic evolution of ligands by exponential enrichment (SELEX), but the nanosized EXOs introduce substantial separation challenges that complicate SELEX workflows. Here, we present DeteRministic Evolution of Aptamers via a Microfluidic-integrated robotic platform (DREAMbot), an automated system engineered to execute multiround EXO-targeted ap

Small bites for big problems: stepwise aggregate degradation by autophagy.

Protein aggregates are a pathological hallmark of diverse disorders, including many neurodegenerative diseases, but also cardiometabolic disease and cancer. While the ubiquitin-proteasome system efficiently removes many soluble misfolded proteins, large or persistent assemblies often require the autophagy-lysosome pathway for their degradation. In the present mini-review, we summarize our knowledge of aggrephagy, the selective clearance of protein aggregates by autophagy, and discuss two recent manuscripts that argue that some aggregates must be primed for autophagosomal degradation, through c

A robotic simulation platform for fluoroscopy-guided catheter navigation in transcatheter tricuspid valve intervention: system development and proof-of-concept applications.

Robot-assisted transcatheter tricuspid valve intervention requires operators to manipulate a flexible catheter through a robotic interface while interpreting two-dimensional fluoroscopic images. However, accessible platforms for robotic catheter training and image-guided control evaluation remain limited. This study presents a virtual-physical robotic simulation platform integrating a physically manipulated catheter, optical shape tracking, patient-specific cardiac anatomy, CT-derived virtual fluoroscopic projection rendering, and image-space anatomical augmentation. An innovative germination-driven LAB fermentation strategy for allergenicity reduction in soymilk gels through spatiotemporal protease-mediated epitope disruption.

This study innovatively integrated seed germination (1-3 days) with LAB fermentation to establish a structure-immunomodulation nexus in fermented soymilk gels. Germination degraded hydrophobic proteins, forming soft gels with substantially reduced hardness and improved water-holding capacity, alongside refined porous networks. Structural loosening enabled LAB proteases to access core allergenic domains, cleaving β-barrel (e.g., Gly m 5.03, residues 134-164) and α-helix clusters (e.g., Gly m 5.02, residues 118-124). This synergistic proteolysis reduced IgE-binding and antigenicity by over 40% c

A Redox-Active Tetrathiafulvalene-Based 3D Covalent Organic Framework with scu Topology for Controllable Charge Transport.

Unlike 2D frameworks where conductivity is largely confined to in-plane transport, the scu topology offers 3D conduction pathways that enhance bulk charge mobility. When integrated with redox-active species like tetrathiafulvalene (TTF), the scu architecture promotes electron transfer across the 3D network, enabling tunable conductivity. This article presents the construction of a 3-periodic (4,8)-c covalent organic framework (COF), TU-48, adopting a twofold interpenetrated scu net, achieved through the integration of a tetratopic D 2h-symmetric rectangular TTF structural motif and an octatopi
