Familial amyotrophic lateral sclerosis with posterior column degeneration and basophilic inclusion bodies: a clinical, genetic and pathological study.

We report an autopsy case of familial amyotrophic lateral sclerosis (FALS). The patient was a Japanese woman with hereditary burden. Family history revealed 12 patients with FALS over four generations. She developed muscle weakness of the proximal part of the upper extremities at age 42, followed by dysarthria, dysphagia, muscle weakness and atrophy in the lower extremities, spasticity, hyperreflexia and Babinski's sign. At age 44, she needed ventilatory support. At age 45, she died of bronchopneumonia. The total duration of the disease was three years and one month. Genetic study showed the a

Clinical Obesity Beyond BMI Identifies a Large High-Risk Subgroup in UK Adults.

INTRODUCTION: BMI is widely used to define obesity but does not capture adiposity distribution and related functional impairment. This study assessed obesity prevalence, disease burden, and incident cardiometabolic risk using the new criteria proposed by The Lancet Commission and compared disease burden across BMI-based and functional classifications. METHODS: A population-based cohort study included 436,751 UK Biobank participants cross-classified according to ethnicity-specific BMI thresholds and the new criteria for preclinical and clinical obesity. Obesity was defined using integrated ant

Global burden of atrial fibrillation/atrial flutter and its attributable risk factors from 1990 to 2019.

AIMS: The aim of this study was to estimate the global burden of atrial fibrillation (AF)/atrial flutter (AFL) and its attributable risk factors from 1990 to 2019. METHODS AND RESULTS: The data on AF/AFL were retrieved from the Global Burden of Disease Study (GBD) 2019. Incidence, disability-adjusted life years (DALYs), and deaths were metrics used to measure AF/AFL burden. The population attributable fractions (PAFs) were used to calculate the percentage contributions of major potential risk factors to age-standardized AF/AFL death. The analysis was performed between 1990 and 2019. Globally,

Schistosomicidal, hepatoprotective and antioxidant activities of the methanolic fraction from Clerodendrum umbellatum Poir leaves aqueous extract in Schistosoma mansoni infection in mice.

BACKGROUND: The intensive use of Praziquantel for the treatment of schistosomiasis has raised concerns about the possible emergence of drug-resistant schistosomes. As drug treatment is an important feature of schistosome control programs, the search for alternative drugs is therefore a priority. The aim of this study was to assess the schistosomicidal, hepatoprotective and antioxidant activities of the methanolic fraction from Clerodendrum umbellatum Poir leaves aqueous extract. METHODS: A phytochemical screening of the fraction of C. umbellatum was conducted. The fraction was administered or

Mitochondrial dysfunction and therapeutic approaches in respiratory and limb muscles of cancer cachectic mice.

What is the central question of this study? We explored whether experimental cancer-induced cachexia may alter mitochondrial respiratory chain (MRC) complexes and oxygen uptake in respiratory and peripheral muscles, and whether signalling pathways, proteasome and oxidative stress influence that process. What is the main finding and what is its importance? In cancer cachectic mice, MRC complexes and oxygen consumption were decreased in the diaphragm and gastrocnemius. Blockade of nuclear factor-κB and mitogen-activated protein kinase actions partly restored the muscle mass and force and correcte

Metabolic Syndrome in Middle Eastern Patients with Atherosclerotic Cardiovascular Disease: A High Burden Driven by Cumulative Risk Factors.

BACKGROUND: Metabolic syndrome (MS), characterized by a constellation of interrelated cardiometabolic abnormalities, markedly amplifies cardiovascular risk. Despite the high prevalence of atherosclerotic cardiovascular disease (ASCVD) in the Middle East, evidence regarding the burden and determinants of MS in this high-risk population remains limited. This study aimed to estimate the prevalence of MS and identify its independent predictors among Middle Eastern patients with established ASCVD. METHODS: This comprehensive analysis integrated data from two complementary sources: a prospective co

Quantification of Lewy body pathology by cerebrospinal fluid endpoint dilution RT-QuIC in a neuropathological autopsy cohort of clinically heterogeneous participants.

The identification of biomarkers predicting the burden of brain alpha-synuclein (α-syn) pathology in vivo represents a research priority in Lewy body disease (LBD). Recently, some kinetic parameters of seed amplification assays (SAAs) for α-syn showed associations with measures of clinical progression. However, preanalytical and analytical factors significantly affect these parameters, reducing reproducibility. The Endpoint Dilution (ED) SAA Real-time Quaking-induced Conversion (RT-QuIC) is emerging as an alternative, more accurate tool for seed quantification. Still, the approach needs valida

An Unusual Lawn Mower Injury: A Case Report.

Penetrating abdominal injuries can be life-threatening and require prompt recognition and surgical intervention. We present the case of a 48-year-old male patient who presented to the emergency department with abdominal pain that began while mowing his lawn just before arrival but without any external signs of trauma. Point-of-care ultrasound revealed a possible linear hyperechoic foreign body just beneath the abdominal wall, a finding confirmed by computed tomography. The foreign body was subsequently retrieved in the operating room, and the patient had an excellent recovery. This type of abd

Closing the loop for Deep Brain Stimulation implants enables personalized healthcare for Parkinson's disease patients.

DEEP brain stimulation implants have improved life quality for more than 70,000 patients world-wide with diseases like Parkinson's, essential tremor, or obsessive-compulsive disorder where pharmaceutical therapies alone could not offer sufficient relief. Still, optimization and monitoring relies heavily on regular clinical visits, putting a burden on patient's comfort and clinicians. Permanent monitoring and combination with other patient health signals could ultimately lead to a personalized closed-loop therapy with remote quality monitoring. This requires technological improvements on the DB

Unequal patterns of tuberculosis in women of reproductive age: Insights from the Global burden of disease study 2021.

BACKGROUND: Tuberculosis (TB) remains a major global cause of mortality and is a leading cause of death among women of reproductive age (WRA). Untreated TB in this population endangers both maternal and neonatal health, yet the age-stratified burden and the shifting distribution of drug-resistant tuberculosis remain poorly defined in global health agendas. This study aims to employ data from the 2021 Global Burden of Disease Study to systematically evaluate the global, regional, and age-specific burden of TB among WRA, to delineate temporal trends in drug-resistant TB, and to assess associated

Global Burden, Risk Factor Analysis, and Prediction Study of Ischemic Stroke, 1990-2030.

BACKGROUND AND OBJECTIVES: Ischemic stroke (IS), 1 of the 2 main subtypes of stroke, occurs because of brain ischemia caused by thrombosis of a cerebral blood vessel. IS is one of the most important neurovascular causes of death and disability. It is affected by many risk factors, such as smoking and a high body mass index (BMI), which are also critical in the preventive control of other cardiovascular and cerebrovascular diseases. However, there are still few systematic analyses of the current and predicted disease burden and the attributable risk factors of IS. METHODS: Based on the Global

Postconcussive Symptoms, PTSD, and Medical Disease Burden in Treatment-Seeking OEF/OIF/OND Veterans.

INTRODUCTION: The most common psychological and cognitive sequelae associated with deployments to Afghanistan (Operation Enduring Freedom) and Iraq (Operation Iraqi Freedom) are mild traumatic brain injury (mTBI) and post-traumatic stress disorder (PTSD). High rates of PTSD are often observed among Veterans with a history of mTBI, and persistent postconcussive symptoms commonly endorsed after mTBI are known to be associated with PTSD. Therefore, this study examined whether PTSD mediates relations between postconcussive symptoms and 2 indices of medical disease burden: 1) the number of disease
