BACKGROUND: Cellular senescence influences cancer progression through irreversible cell-cycle arrest, immune surveillance, and paracrine signaling mediated by the senescence-associated secretory phenotype (SASP). SASP comprises inflammatory cytokines, chemokines, growth factors, and matrix-remodeling enzymes that regulate the tumor microenvironment (TME).
OBJECTIVE: This review describes the current evidence on the mechanisms of SASP induction, its condition-dependent roles in cancer, TME remodeling, and emerging strategies targeting senescent cells and SASP signaling.
METHODS: Relevant lite…

BACKGROUND: Age-related hearing loss (ARHL) is a progressive, bilaterally symmetric sensorineural hearing disorder characterized by predominant high-frequency threshold elevation, and it represents the most prevalent chronic sensory impairment in the geriatric population worldwide. Circular RNA PVT1 (circPVT1) has been identified as an endogenous inhibitor of cellular senescence in multiple cell types, but its expression profile and functional role in auditory cell senescence remain unknown.
METHODS AND RESULTS: In this study, we established a D-galactose-induced senescence model in HEI-OC1 a…

BACKGROUND: Cellular senescence plays a critical role in regulating angiogenesis. While UTX has been implicated in vascular regeneration following spinal cord injury (SCI), its underlying mechanisms remain incompletely understood. This study investigates how senescence mediates UTX-regulated vascular responses after SCI.
METHODS: We assessed p16INK4a (a senescence marker) and UTX expression in spinal cord endothelial cells at 3, 5, 7 and 14 days post-SCI. By breeding UTXflox/flox mice with heterozygous Tek-Cre mice, we generated endothelial-specific UTX knockout mice. Cellular senescence and …

Radiation-induced lung injury (RILI) not only limits the therapeutic dose that can be administered during chest and thorax radiotherapy (RT), but also significantly impairs patients' health and quality of life. RT-induced senescence and the associated altered secretory profile, the senescence-associated secretory phenotype (SASP), have emerged as a central process for the development and progression of pneumonitis and pulmonary fibrosis. Among various lung cell types, the phenomenon of permanent cell cycle arrest, which is also accompanied by significant morphological changes, has been observe

Serum amyloid A (SAA) is an apolipoprotein produced mainly by the liver. SAA1 is upregulated during inflammation, whereas SAA4 is constitutively expressed and is generally considered to be unresponsive to proinflammatory cytokines. However, SAA4 has been identified as a gene whose expression increases in senescent hepatocytes. Cellular senescence has been implicated in the onset and progression of hepatic steatosis. This study aimed to clarify how cellular senescence influences SAA expression and cytokine responsiveness under steatotic conditions. An in vitro senescence-like model was establis…

BACKGROUND & AIMS: Accumulating evidence suggest that Hippo-yes-associated protein (YAP) pathway plays important roles in development and repair after injuries in biliary system. We disclosed that senescent biliary epithelial cells (BECs) participate in the pathogenesis of primary biliary cholangitis (PBC). We hypothesized that dysregulation of Hippo-YAP pathway may be associated with biliary epithelial senescence in pathogenesis of PBC.
APPROACH & RESULTS: Cellular senescence was induced in cultured BECs by treatment with serum depletion or glycochenodeoxycholic acid. The expression and acti…

Cellular senescence is characterized as a stable proliferation arrest that can be triggered by multiple stresses. Most knowledge about senescent cells is obtained from studies in primary cells. However, senescence features may be different in cancer cells, since the pathways that are involved in senescence induction are often deregulated in cancer. We report here a comprehensive analysis of the transcriptome and senolytic responses in a panel of 13 cancer cell lines rendered senescent by two distinct compounds. We show that in cancer cells, the response to senolytic agents and the composition …

Cellular senescence caused by oncogenic stimuli is associated with the development of various age-related pathologies through the senescence-associated secretory phenotype (SASP). SASP is mediated by the activation of cytoplasmic nucleic acid sensors. However, the molecular mechanism underlying the accumulation of nucleotide ligands in senescent cells is unclear. In this study, we revealed that the expression of RNaseH2A, which removes ribonucleoside monophosphates (rNMPs) from the genome, is regulated by E2F transcription factors, and it decreases during cellular senescence. Residual rNMPs ca…

Cellular senescence-an age-associated state characterized by irreversible cell cycle arrest and proinflammatory signaling-contributes to vascular dysfunction and cardiovascular disease. However, the molecular mechanisms linking senescence to vascular impairment remain incompletely defined. Progranulin (PGRN) is a multifunctional protein involved in inflammation, lysosomal function, and cellular homeostasis, but its role in vascular aging is not well understood. We assessed PGRN expression in human and mouse arteries and in senescent vascular smooth muscle cells (VSMCs). Vascular function was e…

Cellular senescence is implicated in age-related pathologies, and identifying circulating biomarkers of senescence holds great diagnostic potential. Circulating senescence signatures are predictive of age-related traits and diseases, though cell type senescence signatures have not been comprehensively explored. In this study, senescence signatures from the Senescence Catalog (SenCat), including 14 human cell types are examined in circulation for clinical relevance in two longitudinal studies-1,275 participants of the Baltimore Longitudinal Study of Aging (BLSA) and 997 participants of the Inve…

Diabetic nephropathy (DN) is a common and severe microvascular complication of diabetes. The stress-induced senescence of renal tubular cells (RTCs) promotes the persistent progression of DN. Fatty-acid β‑oxidation (FAO) is the main energy source of RTCs; factors such as high glucose can trigger mitochondrial injury and abnormalities in renal tubular lipid metabolism, thereby constituting key contributors to stress-induced senescence of RTCs. Despite their ability to undergo cellular senescence, these cells maintain metabolic activity and continuously secrete pro-inflammatory, pro-fibrotic, an…

Aging is associated with impaired lung repair and increased susceptibility to injury, yet the mechanisms underlying age-associated dysfunction of alveolar type II (AT2) cells remain poorly understood. Although cellular senescence is a hallmark of aging, emerging evidence suggests that aging is also characterized by progressive loss of cellular resilience, rendering epithelial cells increasingly vulnerable to stress. Here, we show that AT2 cells in aged lungs remain predominantly non-senescent under basal conditions but exhibit increased susceptibility to stress-induced senescence. This stress-…

IDH1/2 mutations are prevalent genetic alterations in gliomas that facilitate metabolic reprogramming and epigenetic modifications, which are essential for glioma progression. However, their exact contributions to tumorigenesis remain to be fully elucidated. Cellular senescence is a known precursor to tumorigenesis, and multiple oncogenes can initiate this senescence program. Our study demonstrated that the IDH1 mutation inhibits the proliferation of astrocytes and glioma cells, inducing cellular senescence through mechanisms involving DNA damage and increased production of reactive oxygen spe…

Aging-related bone diseases are often interpreted through hormonal decline, defective remodeling, and chronic low-grade inflammation, yet these accounts do not fully capture the cellular, spatial, and temporal heterogeneity of skeletal aging. Bone is an immune-active organ sustained by coordinated signaling among skeletal, hematopoietic, immune, adipose, vascular, and neural compartments. Cellular senescence perturbs this network through stable cell-cycle arrest, metabolic remodeling, and a heterogeneous senescence-associated secretory phenotype (SASP). Whereas recent osteoimmunology syntheses…
