Perfluoroalkyl and polyfluoroalkyl substances (PFAS) and their effects on the ovary.

BACKGROUND: Perfluoroalkyl and polyfluoroalkyl substances (PFAS) are found widespread in drinking water, foods, food packaging materials and other consumer products. Several PFAS have been identified as endocrine-disrupting chemicals based on their ability to interfere with normal reproductive function and hormonal signalling. Experimental models and epidemiologic studies suggest that PFAS exposures target the ovary and represent major risks for women's health. OBJECTIVE AND RATIONALE: This review summarises human population and toxicological studies on the association between PFAS exposure a

Lipid metabolism disorders contribute to hepatotoxicity of triclosan in mice.

Previous in vivo exposure studies focused mainly on nuclear receptors involved in hepatotoxicity of triclosan (TCS). As liver plays a vital role in metabolic processes, dysregulations in lipid metabolism have been identified as potential drivers of pathogenesis. Investigation of changes in lipid metabolism might widen our understanding of toxicological effects as well as the underlying mechanism occurring in the liver. In this study, we comprehensively assessed the effect of TCS exposure on hepatic lipid metabolism in mice. Our results showed that TCS induced significant changes in hepatic fre

Outcomes from massive paracetamol overdose: a retrospective observational study.

LINKED ARTICLE: This article is commented on by Bateman DN and Dear JW. Should we treat very large paracetamol overdose differently? Br J Clin Pharmacol 2017; 83: 1163-5. https://doi.org/10.1111/bcp.13279 AIMS: Treatment of paracetamol (acetaminophen) overdose with acetylcysteine is standardized, with dose determined only by patient weight. The validity of this approach for massive overdoses has been questioned. We systematically compared outcomes in massive and non-massive overdoses, to guide whether alternative treatment strategies should be considered, and whether the ratio between measured

Retrorsine in breast milk influences copper handling in suckling rat pups.

AIMS: To explore the hypothesis that a second xenobiotic agent is required with excess copper to produce Indian Childhood Cirrhosis, this study investigated the effect of the pyrrolizidine alkaloid retrorsine fed to the mother during the suckling period upon the serial changes in neonatal copper status. METHODS: Female Wistar rats with new-born litters were fed either a control or a retrorsine (50 mg/kg) diet. At 0, 4, 8, 11, 15, 18 and 21 days, pups from each litter were weighed, sacrificed and their livers removed for copper, DNA and metallothionein analysis. Serum samples were assayed for

Medical application of laser-induced breakdown spectroscopy (LIBS) for assessment of trace element and mineral in biosamples: Laboratory and clinical validity of the method.

BACKGROUND: Biomedical application is based on the use of LIBS-derived data on chemical contents of tissues in diagnosis of diseases, forensic investigation, as well as a mechanism for providing online feedback for laser surgery. Although LIBS has certain advantages, the issue of correlation of LIBS-derived data on chemical element content in different human and animal tissues with other methods, and especially ICP-MS, remains pertinent. The objective of the present review was to discuss the application of laser-induced breakdown spectroscopy (LIBS) for elemental analysis of human biosamples o

Chemical characteristics and oxidative potential of indoor and outdoor PM2.5 in densely populated urban slums.

A significant proportion of population in metropolitan cities in India live in slums which are highly dense and crowded informal housing settlements with poor environmental conditions including high exposure to air pollution. Recent studies report that toxicity is induced by oxidative processes, mediated by the water-soluble PM chemical components leading to reactive oxygen species production thereby causing inflammatory disorders. Hence, for the first time, this study assessed the chemical characteristics and oxidative potential (OP) of indoor and outdoor PM2.5 in two slums in Mumbai, India.

Systems-level molecular and immunological evidence identifies Th17/Treg modulation as a key mechanism of CRSJ's neuroprotection in Parkinson's disease.

BACKGROUND: Parkinson's disease (PD) is a progressive neurodegenerative disorder in which neuroinflammation plays a central role. Congrong Shujing Granules (CRSJ), a traditional Chinese medicine formula, have shown clinical benefits in PD, yet their immunomodulatory mechanisms remain unclear. METHODS: We investigated the effects of CRSJ on Th17/Treg immune balance. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used to identify representative chemical constituents of CRSJ. Representative CRSJ compounds were characterized, and their binding affinities were evaluated by molecular

Effect of permethrin insecticide on TRβ-mediated cone photoreceptor differentiation and spectral sensitivity in zebrafish larvae (Danio rerio).

Permethrin (PM), a pyrethroid insecticide, is used extensively in both domestic and agricultural settings. Recent data suggest that PM exposure poses substantial physiological hazards, particularly to eyes. However, the mechanisms underlying retinal dysfunction following PM exposure have not been explored comprehensively. In this study, both thyroid hormone receptor β (TRβ)- and SWS1-labeled transgenic zebrafish (cone model) as well as TRβ mutant (TRβ-/-) zebrafish were exposed to PM in the initial phases of retinogenesis, to elucidate the mechanisms of PM retinal cone toxicity. PM exposure su

TLC-Derived High-Polar Fractions of Celastrus paniculatus Seeds Attenuate Astrocyte-Driven Microglial Activation Through Suppression of CD40/iNOS Signaling and Pro-Inflammatory Cytokines.

Neuroinflammation mediated by astrocyte-microglia interactions plays a critical role in the progression of neurodegenerative disorders. Celastrus paniculatus (CP) seeds have long been associated with cognitive benefits; however, the chemical composition and anti-inflammatory potential of their high-polarity fractions remain poorly characterized. In this study, thin-layer chromatography (TLC)-derived high-polarity fractions (F6 and F7) from CP seeds were analyzed using untargeted LC-MS/MS metabolite profiling. After quality filtering, 99 metabolites were retained for classification, with enrich

The role of muscle fascia in heterotopic ossification and maintenance of skeletal muscle integrity in fibrodysplasia ossificans progressiva.

The rare genetic disorder fibrodysplasia ossificans progressiva (FOP) is characterized by progressive heterotopic ossification (HO) of skeletal muscles and associated soft tissues. FOP is caused by a gain-of-function mutation in the type l BMP receptor ACVR1 (also known as ALK2) that renders the receptor inappropriately responsive to activin ligands. HO is associated with muscle destruction and compromised muscle regeneration, although little is known of the mechanistic relationship between these pathophysiological disease manifestations. In mouse FOP models, HO is experimentally induced by di
