The complexity of coffee and its impact on metabolism.

Coffee is one of the three most consumed beverages in the world. It is made by first roasting coffee beans, and then grinding and boiling or steeping the roasted beans in water (brewing). The process of roasting and brewing produces a complex mix of bioactive compounds, including methylxanthines (caffeine, theobromine, theophylline), diterpenes, chlorogenic acid, trigonelline, flavonoids, and hydroxycinnamic acid. In the body, these compounds may be metabolized to produce other bioactive compounds. For example, caffeine is primarily (80%) broken down by demethylation to produce paraxanthine. I…

Toxicological safety, antioxidant activity and phytochemical characterization of leaf and bark aqueous extracts of Commiphora leptophloeos (Mart.) J.B. Gillett.

This study aimed to characterize the phytochemical profile of bark and leaves aqueous extract Commiphora leptophloeos, and conduct in vivo a … ned utilizing blood samples. Gallic acid, catechin, and epicatechin were identified in the bark and chlorogenic acid in leaves. Data demonstrated a high content of phenolic compounds and flavonoids associated with significant antioxidant potential. No significant signs in damage or symptoms of toxicity were detected. No marked reduction in cell viability was found …

Influence of Extraction Solvent on the Phenolic Profile and Bioactivity of Two Achillea Species.

The phenolic composition, as well as the antioxidant and antimicrobial activities of two poorly investigated Achillea species, Achillea lingulata Waldst. and the endemic Achillea abrotanoides Vis., were studied. To obtain a more detailed phytochemical profile, four solvents with different polarities were used for the preparation of the plant extracts whose phenolic composition was analyzed using UHPLC-MS/MS (ultra-high performance liquid chromatography-tandem mass spectrometry). The results indicate that both of the investigated Achillea species are very rich in both phenolic acids and flavono

Phytochemical Variations in Spiny and Spineless Safflower (Carthamus tinctorius L.) Phenotypes.

Currently, documentation in the scientific literature regarding the specific phytochemical variations among regionally adapted spiny and spineless safflower (Carthamus tinctorius L.) germplasm remains limited. This study evaluated the biochemical profiles of petals from four Romanian local populations representing spiny (var. typicus) and spineless (var. inermis) phenotypes. Screening via spectrophotometric methods and HPLC-DAD targeted total phenolic content (TPC), total flavonoid content (TFC), individual phenolic compounds, fat-soluble vitamins, total dye yield, and antioxidant capacity (DP

Bioherbicidal Evaluation of Methanol Extract of Sorghum halepense L. Rhizome and Its Bioactive Components Against Selected Weed Species.

Sorghum halepense (L.) Pers. (common name Johnson grass) is a perennial invasive weed that causes great harm worldwide, and its allelopathy has been demonstrated in a series of experiments. The present study offers new insights into its organ-specific phytochemical profiles using state-of-the-art metabolomic technology and explores the effects of a methanol extract of S. halepense rhizomes (ShER) and its major bioactive compounds (p-hydroxybenzoic acid and chlorogenic acid) on three noxious weed species. The phytotoxic effects of ShER are reflected through the inhibition of seed germination an…

Chemical Composition and Antioxidant Profile of Sorghum (Sorghumbicolor (L.) Moench) and Pearl Millet (Pennisetumglaucum (L.) R.Br.) Grains Cultivated in the Far-North Region of Cameroon.

Sorghum and pearl millet are grain crops that can grow in semi-arid climates, with nutritional and bioactive properties superior to those of major cereals such as rice, wheat, and maize. However, these properties vary a lot, depending on the genetic factors, growing conditions, and place of cultivation. Four sorghum and two pearl millet grains cultivars grown in the Far-North region of Cameroon were screened for their chemical composition and antioxidant profile. The proximate and mineral analyses were performed using AOAC standard methods. The antioxidant profile was assayed spectrophotometri

Estimation of the Chelating Ability of an Extract from Aronia melanocarpa L. Berries and Its Main Polyphenolic Ingredients Towards Ions of Zinc and Copper.

Previously, we have revealed that prolonged administration of a polyphenol-rich 0.1% extract from the berries of Aronia melanocarpa L. (chokeberries) alone and under chronic exposure to cadmium influences the body status of zinc (Zn) and copper (Cu). The aim of this study was to evaluate, in an in vitro model, the chelating properties of the extract (0.05% and 0.1%) and its main polyphenolic ingredients (cyanidin 3-O-β-galactoside, chlorogenic acid, neochlorogenic acid, (+)-catechin, (-)-epicatechin, quercetin, and kaempferol) regarding divalent ions of Zn (Zn2+) and Cu (Cu2+) at pH reflecting…

Digestion behavior and antidepressant-like effect promoted by acute administration of blueberry extract on mice.

The aim of this study was to evaluate the antidepressant-like effect of the acute administration of blueberry extract in mice. In addition, the digestion behavior of individual phenolic compounds using an in vitro digestion model was also investigated and the main bioaccessible compounds were determined. During digestion, important changes were observed in the polyphenols concentrations and antioxidant capacity upon the passage through the gastric and enteric phases. Bioactive compounds such as chlorogenic and ferulic acids, catechin, epicatechin, quercetin and malvidin were highly bioaccessib

Human gastrointestinal metabolism of the anti-rheumatic fraction of Dianbaizhu (Gaultheria leucocarpa var. yunnanensis) in vitro: Elucidation of the metabolic analysis in gastric juice, intestinal juice and human intestinal bacteria by UPLC-LTQ-Orbitrap-MSn and HPLC-DAD.

The metabolic conversion effect of the four index compounds, MSTG-B, MSTG-A, gaultherin, and chlorogenic acid by human intestinal bacteria exhibited much stronger. Those markers' variation in content-time curve in volunteer A gut flora were faster than that in volunteer B's. These results indicate that ARF is relatively stable in the gastrointestinal tract.

Dietary fiber modification by cellulase-AP3 supplementation tailors oat digestion under elderly gastrointestinal conditions with attenuated starch hydrolysis and enhanced polyphenol bioaccessibility.

The resilient fiber matrix of oats limits nutrient release and moderates glycemic response. This study employed a dynamic in vitro stomach-intestine model to investigate how Cellulase-AP3 enzyme supplementation alters oat digestion under simulated elderly conditions. Cellulase-AP3 supplementation caused significant oat matrix disintegration by enzymatically converting insoluble fiber (54.84% to 32.18%) into soluble fiber (50.56% to 59.63%), which in turn elevated digestive fluid viscosity and delayed gastric emptying. This modified physiological environment hindered amylase action, thereby sub

Bioavailability of dietary polyphenols and gut microbiota metabolism: antimicrobial properties.

Polyphenolic compounds are plant nutraceuticals showing a huge structural diversity, including chlorogenic acids, hydrolyzable tannins, and flavonoids (flavonols, flavanones, flavan-3-ols, anthocyanidins, isoflavones, and flavones). Most of them occur as glycosylated derivatives in plants and foods. In order to become bioactive at human body, these polyphenols must undergo diverse intestinal transformations, due to the action of digestive enzymes, but also by the action of microbiota metabolism. After elimination of sugar tailoring (generating the corresponding aglycons) and diverse hydroxyl m…
