Computational pharmacology and in silico clinical trials for the development of drugs from natural products: A review of advances, opportunities, and challenges.

Natural products have inspired the discovery of several drug candidates and US Food and Drug Administration-approved drugs. However, the conventional drug development pathway has several limitations, necessitating innovative strategies. Computational pharmacology and in silico clinical trials (ISCTs) have emerged as a potential to optimize the drug discovery process. Current literature rarely discusses these 2 domains together, leaving limited guidance on how computational tools used in natural product research can complement ISCT frameworks. This review addresses this gap by summarizing key c

A mobile intra-abdominal low-grade fibromyxoid sarcoma arising from the jejunal mesentery in an adolescent: a case report and literature review.

Low-grade fibromyxoid sarcoma (LGFMS) is a rare fibroblastic neoplasm that only exceptionally involves the intra-abdominal cavity. We report a 17-year-old girl with a one-year history of a progressively enlarging, painless, freely mobile abdominal mass. Computed tomography and magnetic resonance imaging showed a localized, well-circumscribed, pedunculated lesion with positional variability, initially suggesting a completely resectable gastrointestinal stromal tumor. Preoperative biopsy was not performed because systemic treatment was not anticipated and intact resection was planned while minim

Perceive-Assess-Dose-Safeguard: a safety-gated state-action grammar for psychotherapy micro-decisions in computational psychiatry.

Psychotherapy unfolds as a sequence of rapid micro-decisions under uncertainty. Within seconds, clinicians integrate verbal, paraverbal, embodied, and relational cues, estimate the patient's momentary capacity for affective work, choose an intervention dose, and apply stop rules to prevent overwhelm and rupture. Computational psychiatry offers principled frameworks for sequential decision-making, but progress in computational psychotherapy remains constrained by the lack of clinically grounded, machine-readable grammars that capture therapist micro-decisions in context. I introduce the Perceiv

A hybrid deep learning model for robust and efficient plant leaf disease detection using ResNet50, PCA, and SVM.

Agricultural productivity sustains itself by detecting diseases in the leaves of plants, especially in poor nations where economic growth is greatly affected by delayed or incorrect detection of diseases. Although high classification accuracy is achieved by the application of deep learning techniques like VGG16 and DC-GAN-based architecture, high computational complexity is still an issue. The optimization-oriented hybrid model for the classification of plant leaf diseases developed in this research emphasizes the viability of deployment and computational efficiency over algorithmic innovation

Spin-dependent electron transfer through a ring-wire coupled junction: Role of in-plane electric field.

We study spin-dependent transport in a hybrid magnetic system, where a non-magnetic (NM) wire is coupled to a side-attached antiferromagnetic (AFM) mesoscopic ring, placed between two non-magnetic electrodes subject to an in-plane electric field oriented perpendicular to the NM wire. The system is described within a tight-binding (TB) framework, and transport properties are computed using the non-equilibrium Green's function (NEGF) formalism.
We consider two junction configurations distinguished by the ring-wire coupling: a single-coupled junction and a double-coupled junction. In the sing

Multi-Constraint Three-Dimensional Bin Packing Optimization for Mixed Vehicle Types: A Heuristic Approach.

Aiming at the multi-constraint three-dimensional bin packing problem for mixed vehicle types in urban logistics, where traditional exact algorithms are limited by NP-hard computational complexity and practical engineering constraints, this study proposes a progressive optimization framework for vehicle loading and fleet allocation optimization. First, a heuristic loading algorithm based on the extreme point method and greedy strategy is developed to maximize single-vehicle loading efficiency by balancing space and weight utilization. Second, an NSGA-II based evolutionary framework with sequent

Process Integration and Reliability Challenges of Through-Glass Vias for Glass-Based Advanced Packaging: A Focused Review.

Recent advances in chiplet architectures, heterogeneous integration, 2.5D/3D packaging, high-performance computing, and RF applications have increased the demand for high-density vertical interconnects and low-loss packaging platforms. Glass substrates have attracted considerable attention for next-generation advanced packaging because of their low dielectric loss, high dimensional stability, smooth surface, and compatibility with large-area panel-level processing. Through-glass vias (TGVs) are essential vertical interconnect structures that enable the electrical integration of glass substrate

Research on key components for railway freight vehicles based on improved real-time object detection methods.

The detection of key components in railway freight vehicles is challenged by diverse object categories, large scale variations, blurred structural boundaries, and complex background interference. To address the limitations of existing real-time Transformer-based detectors such as RT-DETR in multi-scale representation and small object modeling, this study proposes an enhanced real-time detection framework. A lightweight dynamic hybrid convolutional network is introduced to replace part of the backbone, employing adaptive dynamic kernel allocation to preserve large receptive fields while signifi

Laparoscopic-first approach for treating a small bowel injury in a patient stabilized after initial hemodynamic shock: a case report and video.

INTRODUCTION: Laparotomy is the standard treatment for penetrating abdominal trauma; however, the role of laparoscopy is expanding. We report the case of a penetrating jejunal injury in a patient who initially presented with shock but was successfully managed using a laparoscopic-first approach. CASE PRESENTATION: A 61-year-old male with a history of distal gastrectomy presented with a self-inflicted abdominal stab wound and had ingested antihypertensive drugs and cigarettes. He was initially diagnosed with hemorrhagic shock (systolic blood pressure, 60-70 mmHg), which stabilized after fluid

Gillespie-based simulation and inference for non-Markovian stochastic reaction networks.

Discrete stochastic processes are widespread across physics, chemistry, ecology, and beyond. In computational biology and epidemiology, however, most simulators still assume Markovian kinetics with memoryless dynamics, despite growing evidence for history-dependent effects in gene regulation, RNA transcription, cell differentiation, and infection. This reliance on Markovian models limits the routine use and comparison of more realistic, memory-aware descriptions. Here, we develop and benchmark a unified framework for simulating non-Markovian reaction networks using Gillespie-based algorithms.

Integrating artificial intelligence and bioimpedance spectroscopy for enhanced pancreatic disease diagnosis.

The purpose of this study is to develop bioimpedance spectroscopy methods for diagnosing pancreatic diseases. We developed a descriptor approach using impedance spectroscopy results, generating four amplitude-phase-frequency responses from four quasi-orthogonal leads. They create the feature spaces required for our hybrid classifier for diagnosing pancreatic diseases, consisting of five macro levels, the first of which is based on probabilistic neural networks, and the remaining four are based on fuzzy inference. We also presented a device structure for generating informative feature spaces. T

From T-cell defect to tumorigenesis: Epstein-Barr virus-associated smooth muscle tumors in ITK deficiency.

Interleukin-2-inducible T-cell kinase (ITK) is critical for T-cell receptor signaling and antiviral immunity. ITK deficiency is a rare combined immunodeficiency, classically linked to Epstein-Barr virus (EBV)-driven lymphoproliferation, with only two previous reports of EBV-associated smooth muscle tumors (EBV-SMT). We report a 13-year-old boy with recurrent respiratory infections and bilateral adrenal masses. Immunophenotyping, targeted gene panel sequencing, histopathology, immunohistochemistry, and EBV in situ hybridization were performed to investigate underlying immunodeficiency and tumor

Dysphagia Lusoria Revealing Bicuspid Aortic Stenosis With an Aberrant Right Subclavian Artery and Contralateral Subclavian Aneurysm.

Aberrant right subclavian artery is the most common congenital anomaly of the aortic arch and may present with dysphagia lusoria due to esophageal compression. Although bicuspid aortic valve disease is commonly associated with aortopathy, its coexistence with an aberrant right subclavian artery and a contralateral subclavian artery aneurysm is exceedingly rare. We report the case of a 33-year-old male presenting with exertional dyspnea, chest pain, presyncope, and intermittent dysphagia to solids. Echocardiography demonstrated severe bicuspid aortic stenosis with a mean gradient of 95 mmHg and

A class-wise quantum relational calibration network for brain tumor diagnosis.

Accurate and reliable brain tumor diagnosis from magnetic resonance imaging (MRI) remains a challenging task, particularly in multi-class settings where inter-class ambiguity and miscalibration may degrade predictive performance. In this work, we propose a Class-wise Quantum Relational Calibration Network (CQRCNet), a hybrid quantum-classical framework that integrates quantum relational modeling into a classical convolutional neural network backbone. Our approach employs a parameter-shared and low-qubit quantum circuit to explicitly model pairwise interactions between class-specific evidences,

Single-channel EEG-based sleep stage classification via hybrid data distillation.

Objective. With the advancement of deep learning technologies, more and more researchers have begun developing end-to-end automatic sleep stage classification frameworks. However, these frameworks typically require access to large electroencephalogram (EEG) datasets for training, which imposes a significant computational burden. Furthermore, EEG data contains patient privacy information, and using such data for training raises concerns about privacy infringement. To address these issues, we propose a hybrid data distillation method. We aim to enable single-channel EEG sleep stage classification

Design and Optimization of a Hybrid Railcar Structure with Multilayer Composite Panels.

Within contemporary railway engineering, manufacturers of rolling stock are increasingly focused on developing vehicles that combine reduced weight with enhanced reliability. This objective is largely motivated by the need to decrease energy demand and limit environmental impact, encouraging the integration of innovative materials and cut-ting-edge design strategies. The growing use of multilayer composite materials in the railway sector stems from their unique combination of high strength and low weight, making them particularly suitable for structural applications. This study investigates th

Fracture Resistance of 3D-Printed Hybrid Abutment Crowns Made from a Tooth-Colored Ceramic Filled Hybrid Composite: A Pilot Study.

The aim of this pilot in vitro study is to investigate the fracture strength of hybrid abutment crowns (HACs) made of a 3D-printable, tooth-colored, ceramic-reinforced composite (CRC). Based on an upper first premolar, a crown was designed, and specimens were additively fabricated from a composite material (VarseoSmile Crown plus) (N = 32). The crowns were bonded to standard abutments using a universal resin cement. Half (n = 16) of the samples were subjected to artificial aging, during which three samples suffered minor damage. All specimens were mechanically loaded at an angle of 30° to the
