TaRGET II Data Portal: a multi-omics resource for environmental toxicant studies in mice.

The Toxicant Exposures and Responses by Genomic and Epigenomic Regulators of Transcription (TaRGET) program is a multiphase program that aims to understand how environmental factors contribute to disease susceptibility using toxicant-exposed mouse models. Here, we present the TaRGET II Data Portal ( https://data.targetepigenomics.org/ ), a comprehensive repository of multi-omics datasets generated from mice exposed to a range of environmental toxicants, including arsenic (As), lead (Pb), bisphenol A (BPA), tributyltin (TBT), di(2-ethylhexyl) phthalate (DEHP), tetrachlorodibenzo-p-dioxin (TCDD)

Don't judge a sperm by its cover.

Male fertility is primarily assessed using descriptive semen parameters, such as sperm concentration, motility and morphology. These conventional parameters provide an initial assessment of male reproductive health and may explain why a couple is struggling to conceive; however, they are not a direct measure of fertility potential. Growing evidence suggests that spermatozoa transmit far more than paternal DNA. Alongside the genome, spermatozoa deliver proteins, RNA and epigenetic information that are responsive to environmental and lifestyle exposures, and can influence embryo development, pre

Rapid multiplex detection of Echinococcus granulosus and Echinococcus multilocularis using a one-pot RPA-assisted CRISPR-Cas12a/Cas13a assay in a portable multi-tube device.

Echinococcosis, caused by Echinococcus granulosus and Echinococcus multilocularis, remains a significant zoonotic threat, particularly in pastoral regions where rapid environmental surveillance is essential yet technically constrained. Here, we report a rapid and integrated one-pot recombinase polymerase amplification -assisted, orthogonal CRISPR-Cas12a/Cas13a platform for rapid and specific discrimination of these two species in environmental samples. Coupled with a simplified NaOH-based DNA extraction method, the assay enables a streamlined workflow completed within 60 min, achieving a detec

Epigenetic Mechanisms in Autoimmune Thyroid Diseases: Bridging Research and Clinical Applications.

Autoimmune thyroid disease (AITD) exemplifies an organ-specific autoimmune disorder, including Hashimoto's thyroiditis (HT) and Graves' disease (GD). HT is characterized by hypothyroidism, whereas GD primarily presents as hyperthyroidism. Immunological evidence indicates that AITD pathogenesis requires both a permissive genetic background and environmental triggers to initiate and sustain disease progression. However, the exact molecular and cellular pathways through which these elements synergize to trigger and sustain autoimmune responses remain unclear. Emerging evidence suggests that epige

The mitoepigenome responds to stress, suggesting novel mito-nuclear interactions in vertebrates.

The mitochondria are central in the cellular response to changing environmental conditions resulting from disease states, environmental exposures or normal physiological processes. Although the influences of environmental stressors upon the nuclear epigenome are well characterized, the existence and role of the mitochondrial epigenome remains contentious. Here, by quantifying the mitochondrial epigenomic response of pineal gland cells to circadian stress, we confirm the presence of extensive cytosine methylation within the mitochondrial genome. Furthermore, we identify distinct epigenetically

Detection of Coccidioides immitis in Archived Soils from Madera County, California.

In the 1970s, archaeologists excavating sites in Madera County, California, which is now submerged by Hensley Reservoir, contracted coccidioidomycosis (Valley fever), a fungal disease caused by Coccidioides immitis. Soil samples from these sites were archived but not processed due to concerns about infectious arthroconidia. This study investigates whether these decades-old samples, stored at room temperature, still contain Coccidioides and pose a risk to researchers and collections managers. We analyzed 176 subsamples using molecular and culture-based methods, with DNA extractions performed in

Epidemiological investigation of a clinical case of Q fever in the Brazilian Amazon.

The bacterium Coxiella burnetii, responsible for causing Q fever in humans and coxiellosis in animals, is globally distributed, with the exception of New Zealand. In Brazil, is a neglected zoonosis, with scarce information, and with the number of reported cases in humans and animals is certainly underestimated, being considered an important neglected zoonosis. The present study aimed to evaluate, through an epidemiological investigation, a farm located in Belterra, Pará, Brazil, where a recent human case of Q fever occurred, considered the first recognized in the Amazon region of Brazil. Serology and PCR for Apicomplexa and SARS-CoV-2 in feral cats from a Brazilian island.

Feral cats in insular environments represent a relevant interface between wildlife, environmental contamination, and zoonotic risk under a One Health perspective, as feralization increases exposure to infections and facilitates pathogen transmission. Herein, we surveyed Toxoplasma gondii, Sarcocystis spp., Neospora caninum, and SARS-CoV-2 in Furtada Island, Mangaratiba, Rio de Janeiro, Brazil, where there has been a feral cat population since the 1950s. This study aimed to investigate serological, molecular, and morphological indicators of infection in feral cats by assessing antibodies agains

Machine learning-enhanced surface-enhanced spectroscopic detection of polycyclic aromatic hydrocarbons in the human placenta.

The detection and identification of polycyclic aromatic hydrocarbons (PAHs) and their derivatives, polycyclic aromatic compounds (PACs), are essential for environmental and health monitoring, for assessing toxicological exposure and their associated health risks. PAHs/PACs are the most dangerous chemicals found in tobacco smoke, and cigarette use during pregnancy can convey these molecules to the developing fetus through the placenta. This exposure is associated with many negative health outcomes, from premature birth to sudden infant death syndrome and adverse neurodevelopmental disorders. Th

Tooth Decay: Genetic and Epigenetic Insights Driving the Development of Anti-Caries Vaccines.

Dental caries is now recognised as a multifactorial disease shaped by complex interactions among genetic, epigenetic, microbiological, environmental, and social factors. This narrative review synthesises recent findings on the influence of genetic and epigenetic factors on caries susceptibility, exploring implications for personalised prevention strategies, including novel vaccine approaches. Numerous gene polymorphisms in pathways related to enamel formation, saliva composition, immune response, and taste perception have been linked to increased caries risk, with some effects modulated by sex

Pesticide-induced transgenerational alterations of genome-wide DNA methylation patterns in the pancreas of Xenopus tropicalis correlate with metabolic phenotypes.

The unsustainable use of manmade chemicals poses significant threats to biodiversity and human health. Emerging evidence highlights the potential of certain chemicals to cause transgenerational impacts on metabolic health. Here, we investigate male transmitted epigenetic transgenerational effects of the anti-androgenic herbicide linuron in the pancreas of Xenopus tropicalis frogs, and their association with metabolic phenotypes. Reduced representation bisulfite sequencing (RRBS) was used to assess genome-wide DNA methylation patterns in the pancreas of adult male F2 generation ancestrally expo

A crosstalk between epigenetic modulations and non-alcoholic fatty liver disease progression.

Non-alcoholic fatty liver disease (NAFLD) has recently emerged as a major public health concern worldwide due to its rapidly rising prevalence and its potential to progress into end-stage liver disease. While the precise pathophysiology underlying NAFLD remains incompletely understood, it is strongly associated with various environmental triggers and other metabolic disorders. Epigenetics examines changes in gene expression that are not caused by alterations in the DNA sequence itself. There is accumulating evidence that epigenetics plays a key role in linking environmental cues to the onset a

DNA damage and oxidative stress in gill cells of Cnesterodon decemmaculatus exposed to pesticides by runoff source in an agricultural basin.

In our country, great concern exists about diffuse pollution cause by the great use of pesticides in rural environments. A thorough analysis is needed to generate information, know the real situation and thus, be able to make decisions with the purpose of reducing environmental pollution. In situ bioassays have been carried out using Cnesterodon decemmaculatus within limnocorrals located in a surface natural water system that receives rainfall excess flowing from an agricultural basin with a typical crop rotation, including corn, wheat and soy. Specimens were taken from the limnocorrals 72 h a

Years of Schooling Could Reduce Epigenetic Aging: A Study of a Mexican Cohort.

Adverse conditions in early life, including environmental, biological and social influences, are risk factors for ill-health during aging and the onset of age-related disorders. In this context, the recent field of social epigenetics offers a valuable method for establishing the relationships among them However, current clinical studies on environmental changes and lifespan disorders are limited. In this sense, the Tlaltizapan (Mexico) cohort, who 52 years ago was exposed to infant malnutrition, low income and poor hygiene conditions, represents a vital source for exploring such factors. Role of Epigenomics in Bone and Cartilage Disease.

Phenotypic variation in skeletal traits and diseases is the product of genetic and environmental factors. Epigenetic mechanisms include information-containing factors, other than DNA sequence, that cause stable changes in gene expression and are maintained during cell divisions. They represent a link between environmental influences, genome features, and the resulting phenotype. The main epigenetic factors are DNA methylation, posttranslational changes of histones, and higher-order chromatin structure. Sometimes non-coding RNAs, such as microRNAs (miRNAs) and long non-coding RNAs (lncRNAs), ar

Helicobacter pylori as an oncogenic pathogen, revisited.

Gastric cancer is an inflammation-associated malignancy aetiologically related to infection with the bacterium, Helicobacter pylori, which is considered a necessary but insufficient cause. Unless treated, H. pylori causes life-long acute and chronic gastric inflammation resulting in progressive gastric mucosal damage that may result in gastric cancer. The rate of progression from superficial gastritis, to an atrophic metaplastic mucosa, and ultimately to cancer relates to the virulence of the infecting H. pylori as well as host and environmental factors. H. pylori virulence is a reflection of

Genotyping and Axenic Growth of Coxiella burnetii Isolates Found in the United States Environment.

Coxiella burnetii is a gram-negative bacterium that is the etiologic agent of the zoonotic disease Q fever. Common reservoirs of C. burnetii include sheep, goats, and cattle. These animals shed C. burnetii into the environment, and humans are infected by inhalation of aerosols. A survey of 1622 environmental samples taken across the United States in 2006-2008 found that 23.8% of the samples contained C. burnetii DNA. To identify the strains circulating in the U.S. environment, DNA from these environmental samples was genotyped using an SNP-based approach to derive sequence types (ST) that are

Manganese superoxide dismutase Ala-9Val polymorphism and risk of breast cancer in a population-based case-control study of African Americans and whites.

INTRODUCTION: A polymorphism in the manganese superoxide dismutase (MnSOD) gene, Ala-9Val, has been examined in association with breast cancer risk in several epidemiologic studies. Results suggest that the Ala allele increases the risk of breast cancer and modifies the effects of environmental exposures that produce oxidative damage to DNA. METHODS: We examined the role of the MnSOD Ala-9Val polymorphism in a population-based case-control study of invasive and in situ breast cancer in North Carolina. Genotypes were evaluated for 2025 cases (760 African Americans and 1265 whites) and for 1812

Evaluation of mold exposure in cystic fibrosis patients' dwellings and allergic bronchopulmonary risk.

Very few studies have been conducted on cystic fibrosis (CF) patients' exposure to the indoor environment and, to our knowledge, there are no studies dealing with the link between specific fungal environmental exposure at home and fungal colonization resulting in allergic bronchopulmonary aspergillosis (ABPA). Fungal exposure of CF adult patients with ABPA (n=4) with fungal sensitization (n=7) and with no ABPA (n=5) was assessed in 16 homes by dust sampling with electrostatic dust fall collectors (EDCs). Aspergillus fumigatus was specifically quantified by real-time quantitative polymerase cha

The multiplex interactions and molecular mechanism on genotoxicity induced by formaldehyde and acrolein mixtures on human bronchial epithelial BEAS-2B cells.

Aldehydes are common air pollutants with carcinogenicity. Genotoxicity of single aldehyde has been studied well, but the combined genotoxicity is rarely known. Here, we evaluated the combined genotoxicity of formaldehyde and acrolein on BEAS-2B cells in terms of DNA strands breakage, chromosome damage and gene mutation below subcytotoxic concentrations covering smoking-related concentrations. Meanwhile, the molecular mechanism was investigated further based on oxidative stress, DNA-protein crosslinks (DPCs), cell cycle and DNA damage-repair pathway. Co-exposure to formaldehyde and acrolein mix
