Dopamine (DA) is one of the putative neurotransmitters of the lateral efferent olivocochlear fibers. The cochlear DA content after noise exposure was analyzed using high-performance liquid chromatography coupled with electrochemical detection. Animals were exposed for 1 h to white noise at 70, 90 or 110 dB SPL or were kept in conditions of silence. Half of the animals were pretreated with piribedil, a D2 agonist, and the other half served as controls. In control (untreated) animals, noise stimulation resulted in a progressive decrease of cochlear DA concentration. This decrease was scarcely de…

The extracellular levels of dopamine (DA) and DA metabolites in the caudate-putamen (CPu) and the nucleus accumbens (NA) of rats following administration of haloperidol (HAL) decanoate and fluphenazine (FLU) decanoate for 8 months were assessed using intracranial microdialysis 1 month after final injection. Both HAL- and FLU-treated animals showed persisting plasma neuroleptic levels at time of sacrifice. Extracellular basal levels of homovanillic acid (HVA) in the CPu were significantly elevated in the FLU-treated animals, while basal levels of 3,4-dihydroxyphenylacetic acid (DOPAC) in the CP…

Dopamine beta-hydroxylase (DBH) deficiency is a genetic disorder in which affected patients cannot synthesize norepinephrine, epinephrine, and octopamine in either the central nervous system or the peripheral autonomic neurons. Dopamine acts as a false neurotransmitter in their noradrenergic neurons. Neonates with DBH deficiency have had episodic hypothermia, hypoglycemia, and hypotension, but survivors sometimes cope relatively well until late childhood when overwhelming orthostatic hypotension profoundly limits their activities. The hypotension may be so severe that clonic seizures supervene…

Dopamine receptors have been identified in many tissues including the kidney. To establish an in vitro system as a model for dopamine action, we studied the effect of dopamine (DA) receptor agonists and antagonists on adenosine 3',5'-cyclic monophosphate (cAMP) formation in opossum kidney (OK) cells. The stimulation of cAMP production in these cells by dopamine was dose dependent, and markedly higher levels were observed in the presence of dopamine plus a phosphodiesterase inhibitor, 3-isobutyl-1-methylxanthine. Half-maximal stimulation was found with 1.15 +/- 0.22 microM dopamine. A DA1-recep…

The benefits of dopamine resuscitation in neonatal shock are in doubt. To evaluate dopamine's effectiveness on regional perfusion and survival, neonatal pigs were subjected to fecal Escherichia coli peritonitis-induced septic shock and were randomly divided into equal groups. Control animals (Group I) were not resuscitated with dopamine. Dopamine resuscitation, as a continuous infusion of 5 micrograms/kg/min (Group II), 50 micrograms/kg/min (Group III), or 200 micrograms/kg/min (Group IV), began when cardiac output dropped 20% below baseline. Radiolabeled microspheres were used to assess regio…

D2 dopamine receptor densities were measured in postmortem samples of the caudate nucleus and putamen from 36 parkinsonian patients. The relationship between the age of the patient, duration of the disease, and duration of L-dopa therapy versus density of brain D2 dopamine receptors was examined using [3H]spiperone. Receptor density in parkinsonian tissues was constant over the age range of 56 to 90 years, as was the case for control tissues. Density did not change with duration of disease up to 24 years. Treatment with L-dopa did not cause progressive reduction in receptor density. The dimini…

Separation of dopamine (DA) agonist and antagonist receptors was attempted by means of a covalently-bound fluphenazine-agarose (Flu-agarose). Incubation of striatal membranes with Flu-agarose resulted in loss of 3H-spiroperidol (3H-SPI) binding sites, while incubation with non-coupled agarose did not cause any change. The loss of 3H-SPI binding to the Flu-agarose treated membranes was not attributed to the release of fluphenazine from Flu-agarose as justified by several criteria. Flu-agarose adsorbed more effectively striatal membranes with 3H-SPI binding sites than those with 3H-DA binding si…

The binding of novel dibenzodioxazocine derivatives to rat striatal dopamine receptors was studied in vitro, using 3H-spiperone as radioligand. The biochemical-pharmacological characteristics of the effect of a selected representative, EGYT-2509 are discussed in details. The parameters of specific spiperone binding to rat striatal membrane preparation (KD = 0.550 nM, Bmax = 465 fmole/mg protein) as well as the displacing potencies of known dopamine receptor ligands matched closely the corresponding values in the literature. Using 0.4 nM radioligand, a Ki value of 404 nM was obtained for EGYT-2…

Dopamine receptors were studied in post-mortem brains from control and Huntington's disease patients, using the specific binding of [3H]spiperone to dopamine D-2 receptors and [3H]piflutixol to dopamine D-1 receptors. Both [3H]spiperone binding and [3H]piflutixol binding were reduced by 45-50% in Huntington's disease putamen. The loss of [3H]spiperone and [3H]piflutixol binding sites correlated with decreased GABA concentrations observed in Huntington's disease putamen. A selective loss (48%) of [3H]piflutixol binding was observed in Huntington's disease substantia nigra pars reticulata, [3H]p…

Dopamine was transformed into a redox chemical system for delivery to the brain. The lipoidal form allowed penetration of the blood-brain barrier. Oxidative and hydrolytic processes then transformed the delivery form into a quaternary ammonium precursor of dopamine. The quaternary ammonium precursor was rapidly eliminated from the general circulation, whereas that formed in the brain was locked in, thereby providing a significant and sustained brain-specific dopaminergic activity.

In 20 healthy subjects hydro-saline depletion was achieved by a 4-day natriuretic treatment associated with hypo-saline diet. Dopamine (DA) was infused in subpressor dose (.1 microgram . kg-1 . min-1) during sustained hypotonic polyuria induced by 5% glucose infusion. Four 15-min clearance periods were performed. DA was infused during the 2nd and the 3rd period. The effective renal plasma flow (RPF) as well as the glomerular filtration rate (GFR) were calculated by PAH and creatinine clearances, respectively. In our experimental conditions DA produced: a) an increase in the abnormally isoosmot…

6-OH-DA HBr 5-25 mg injected into one common carotid artery of pentobarbitone-anesthetized cats, induced a transient inhibition of chemosensory discharges recorded from the ipsilateral carotid nerve, but did not change chemosensory frequency during spontaneous ventilation, periodic deep breaths and asphyxia. Chemosensory activities recorded simultaneously from both carotid nerves showed no differences in their inhibitory responses to dopamine and their excitatory responses to NaCN, up to 48 h after the injections of 6-OH-DA. It is concluded that 6-OH-DA acts initially as a dopamine analogue, a…

OBJECTIVE: To ascertain whether vascular endothelial growth factor (VEGF) secretion by luteinized granulosa cells (GCs) is modulated by the dopaminergic system in a dose-dependent fashion and how this is related to the differential efficacy of dopamine receptor 2 (D2)-agonists (D2-ag) in preventing ovarian hyperstimulation syndrome (OHSS).
DESIGN: The relationship between the dopaminergic system and VEGF secretion in luteinized GCs was evaluated. Archived human ovaries were immunostained to characterize D2 expression.
SETTING: University affiliated infertility center.
PATIENT(S): Premenopau…

Dopamine receptor signaling exhibits prominent plasticity that is important for the pathogenesis of both addictive and movement disorders. Psychoactive stimulants that activate the dopamine D(1) receptor (Drd1a) induce the rapid phosphorylation and activation of extracellular signal-regulated kinase 1/2 (ERK1/2) in neurons of the nucleus accumbens and ventral striatum. This response is known to be dependent on the phosphatase inhibitor dopamine- and cAMP-regulated phosphoprotein-32 (DARPP-32) and appears critical for the sensitization of Drd1a responses that contributes to addiction. Loss of d…

The interactions between dopamine and muscarinic receptor subtypes coupled to adenylate cyclase in superfused rat neostriatal slices were investigated using the efflux of cyclic AMP, in the presence of the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine, as a highly sensitive parameter of cyclic AMP production. Cyclic AMP efflux induced by simultaneous activation of (stimulatory) D-1 and (inhibitory) D-2 dopamine receptors by dopamine was reduced profoundly by the muscarinic receptor agonist oxotremorine and by inhibition of acetylcholinesterase with physostigmine, but not by the M-1 m…

Dopamine receptors (DRs) have been implicated in numerous disorders and diseases (e.g., Alzheimer's disease, Parkinson's disease, schizophrenia, and substance use disorders) and have served as attractive drug targets for these ailments. Despite their potential clinical utility, the development of selective DR ligands has been challenging due to difficulties in selectivity among the DR subtypes as well as other biogenic amine receptors and poor pharmacokinetic properties. The realization of their full potential necessitates continued advancements in DR ligands as investigative tools. This revie…
