Targeted senotherapy improves electrographic and behavioral outcomes in a mouse model of temporal lobe epilepsy.

OBJECTIVE: Current pharmacotherapy for temporal lobe epilepsy (TLE) is limited to symptomatic treatment and leaves approximately one third of patients with inadequate seizure control. Discovering disease-modifying targets is an unmet clinical need. We have previously identified senescent cells (SCs) as one such target. Many drugs that eliminate SCs (senolytics) interfere with apoptotic resistance proteins, potentially resulting in broad cytotoxicity and numerous side effects. Newer, more targeted therapies, like selective senescence killing compound 1 (SSK1), a gemcitabine prodrug that is sele

Contribution of the Brown Location Test to the pre-surgical neuropsychological assessment of temporal lobe epilepsy.

The nature of memory deficits in mesial temporal lobe epilepsy (mTLE) can depend on the side of the epileptogenic zone (EZ). Although left mTLE has been consistently associated with impairments in verbal memory, the association between right mTLE and non-verbal memory is less clear, which may be attributable to methodological limitations of existing visuospatial memory tests (e.g., possible use of verbalization strategies, involvement of psychomotor or visuospatial skills, task structure which differs between verbal and non-verbal memory tests). The Brown Location Test (BLT) was developed to e

Language and Memory Network Alterations in Temporal Lobe Epilepsy: A Functional and Structural Connectivity Study.

BACKGROUND AND PURPOSE: This study evaluated preoperative alterations and postoperative reorganization of the joint language-memory network (LMN) from the perspective of resting-state functional and structural connectivity in temporal lobe epilepsy (TLE). Graph theory and machine learning approaches were used to explore automatic lateralization. MATERIALS AND METHODS: Resting-state fMRI and DTI data were obtained from 20 healthy subjects and 35 patients with TLE. Functional and structural connectivity were calculated within the LMN before and after temporal lobectomy. ANOVA was performed to i

The possible role of neurogenesis activators in temporal lobe epilepsy: State of art and future perspective.

Neurogenesis is a complex process by which the neurons and supporting cells of the central nervous system (CNS) are generated by neural stem cells. Adult hippocampal neurogenesis (AHN) in the human brain is an active process during life and plays a critical role in the regulation of memory, cognition, and mood. It has been shown that epilepsy is linked with dysregulation of AHN. Of note, AHN is very sensitive to the pathological electrical stimuli during epileptic seizures, which result in the induction of neurogenesis in acute epilepsy and inhibition of neurogenesis in chronic epilepsy. Epile

Investigating the protective effect of hydroxylated fullerenes on cognitive function in rats with temporal lobe epilepsy.

The objective of this study was to explore the protective effects of hydroxy fullerenes (HFs) on cognitive function in rats with temporal lobe epilepsy (TLE) and to elucidate the underlying mechanisms. Eighteen Sprague-Dawley (SD) rats were randomly selected and administered pilocarpine (50 mg/kg) intraperitoneally to establish a TLE model, and were then randomly assigned to the TLE group and the TLE + HFs group. An additional nine SD rats were served as a normal control group (CON group). The Morris water maze (MWM) test was utilized to assess the spatial learning and memory capabilities of t

Demonstration of impaired facial emotion perception in temporal lobe epilepsy by theta responses in EEG.

OBJECTIVE: Temporale lobe and occipito-temporal cortical areas play an important role in facial emotion perception (FEP). FEP might be represented by event-related brain oscillations. In patients with temporal lobe epilepsy (TLE), impairment of FEP was shown with behavioral and functional neuroimaging (FMRI, PET, MEG) but not event-related oscillations, which is a well known method for cognitive research studies. The present study aims to explore FEP by analyzing EEG event-related theta oscillations in patients with TLE. METHODS: 21 patients with TLE and 19 healthy volunteers were included. D

The Effects of Neuronal Fyn Knockdown in the Hippocampus in the Rat Kainate Model of Temporal Lobe Epilepsy.

Previous studies have demonstrated neuronal and microglial Fyn, a Src family kinase (SFK), and how its interactions with tau contribute to epileptogenesis. Saracatinib, a Fyn/SFK inhibitor, modifies disease progression in rat kainate (KA) epilepsy models. In this study, we investigated neuronal-specific fyn knockdown effects on Fyn-tau signaling, neurodegeneration, and gliosis using a calcium/calmodulin-dependent protein kinase II (CaMKII)-promoter-driven adeno-associated viral vector (AAV9)-mediated fyn-shRNA injection in the rat hippocampus. Eight days following AAV administration, rats rece

Identification of proteins with variable levels of post-translational modifications in human temporal lobe epilepsy.

Comparative mass spectrometry analysis of hippocampal tissue samples from patients with sclerotic and non-sclerotic temporal lobe epilepsy and nonepileptic patients was undertaken to identify differences in the levels of protein post-translational modifications (PTMs). The original proteomic data obtained by Mathoux et al. [DOI: 10.1172/jci.insight.188612] and deposited in the PRIDE repository (PXD064519) were used in this work. Our reanalysis of the comparative proteomic data identified 53 proteins with PTMs (phosphorylation, methylation, acetylation, and citrullination) that exhibited signif

The impact of resective epilepsy surgery on cognitive estimation in patients with mesial temporal lobe lesions.

OBJECTIVE: In recent research, cognitive estimation has been shown to be impaired in patients with right-sided mesial temporal lobe epilepsy due to hippocampal sclerosis (mTLE-HS), an often drug-resistant epilepsy subtype. Despite the impact of cognitive estimation on patients' daily life, its neural basis and outcome in epilepsy surgery are relatively unknown. This study examines the significance of the right temporal lobe regarding cognitive estimation before and after epilepsy surgery. METHODS: Two hypotheses were tested: Right mTLE-HS patients show worse presurgical cognitive estimation p

EEG-Based Seizure Onset Detection of Frontal and Temporal Lobe Epilepsies Using 1DCNN.

OBJECTIVE: The manual interpretation of electroencephalogram (EEG) signals for detecting epileptic seizures is time-consuming and labor-intensive, highlighting the critical importance of exploring automated seizure detection methods. Given this, this work concentrates on seizure detection using scalp EEG signals collected from people with frontal lobe epilepsy (FLE) and temporal lobe epilepsy (TLE). METHOD: 20 FLE patients and 20 TLE patients are utilized in our work, and a parallel one-dimensional convolutional neural network (1DCNN) model is built for classification. Our work explores two s

Ondansetron reduces seizures and improves cognitive impairment in a rat model of temporal lobe epilepsy.

Temporal lobe epilepsy (TLE) remains an intractable chronic neurological disorder. Emerging evidence suggests a crucial modulatory role for the 5-hydroxytryptamine 3 receptor (5-HT₃R) in epilepsy. Ondansetron, a selective 5-HT3 receptor antagonist widely used as an antiemetic, has demonstrated neuroprotective effects in preclinical models of several neurological disorders. However, its potential therapeutic efficacy in chronic TLE has not yet been investigated. In this study, we evaluated ondansetron's effects on spontaneous recurrent seizures (SRS), cognitive function, and hippocampal neurode

[Neurorehabilitation after epilepsy surgery in the temporal lobe in patients with specific neurocognitive risk profiles].

INTRODUCTION: In case of pharmacoresistant focal epilepsy, surgery is often the only way to achieve seizure freedom. These operations may result in cognitive deficits, especially with surgery in the left temporal lobe. The aim of this study was to determine whether patients operated in the left or right temporal lobe, respectively, have different cognitive outcomes. Furthermore, it was investigated if there was a positive influence of occupational therapy on neurorehabilitation in epilepsy patients. METHODS: In all patients cognitive performance was assessed preoperatively and six months post

Serum MicroRNAs as Predictors of Diagnosis and Drug-resistance in Temporal Lobe Epilepsy: A Preliminary Study.

OBJECTIVE: Temporal lobe epilepsy (TLE) is the most common form of refractory focal epilepsy, and the current clinical diagnosis is based on EEG, clinical neurological history and neuroimaging findings. METHODS: So far, there are no blood-based molecular biomarkers of TLE to support clinical diagnosis, despite the pathogenic mechanisms underlying TLE involving defects in the regulation of gene expression. MicroRNAs (miRNAs) have emerged as important post-transcriptional regulators of gene expression. RESULTS: Recent studies show the feasibility of detecting miRNAs in body fluids; circulating

Valproic acid attenuates the severity of astrogliosis in the hippocampus of animal models of temporal lobe epilepsy.

Reactive astrogliosis is one of the most frequency neuropathological alterations in the hippocampus of animal models and patients with temporal lobe epilepsy (TLE). Valproic acid (VPA), a widely used antiepileptic drug (AED), acts by blocking ion channels and enhancing GABAergic activity. This study investigated the effects of VPA on hippocampal astrogliosis in a rat model of TLE. The results demonstrated that chronic administration of VPA at a dose of 200 mg/kg significantly reduced the severity of astrogliosis and ameliorated neuronal loss in the hippocampus at the early and middle stages po

Disease-modifying effects of sodium selenate in a model of drug-resistant, temporal lobe epilepsy.

There are no pharmacological disease-modifying treatments with an enduring effect to mitigate the seizures and comorbidities of established chronic temporal lobe epilepsy (TLE). This study aimed to evaluate for disease modifying effects of sodium selenate treatment in the chronically epileptic rat post-status epilepticus (SE) model of drug-resistant TLE. Wistar rats underwent kainic acid-induced SE or sham. Ten-weeks post-SE, animals received sodium selenate, levetiracetam, or vehicle subcutaneousinfusion continuously for 4 weeks. To evaluate the effects of the treatments, one week of continuo

New insights for predicting surgery outcome in patients with temporal lobe epilepsy. A systematic review.

Resective surgery is the treatment of choice for one-third of adult patients with focal, drug-resistant epilepsy. This procedure is associated with substantial clinical and cognitive risks. In clinical practice, there is no validated model for epilepsy surgery outcome prediction (ESOP). Meta-analyses on ESOP studies assessing prognostic factors report discrepancies in terms of study design. Our review aims to systematically investigate methodological and analytical aspects of studies predicting clinical and cognitive outcomes after temporal lobe epilepsy surgery. A systematic review of ESOP st
