Dual mechanisms of morin in bioavailability potentiation and hepatoprotection: Enhancing enrofloxacin safety in poultry through ABC transporter inhibition and antioxidant pathway activation.

Antimicrobial resistance (AMR) poses a major global public health threat. The low oral bioavailability of antibiotics in poultry, mediated by ATP-binding cassette (ABC) transporters, predisposes the host to subinhibitory conditions that exacerbate AMR transmission. However, earlier-generation ABC transporter inhibitors, such as verapamil and cyclosporine A, are limited by inherent hepatotoxicity. This study aimed to elucidate whether morin enhances the bioavailability of substrate drugs by inhibiting avian ABC transporters and to evaluate its potential in mitigating drug-induced liver injury (

Jaceosidin, a natural polymethoxyflavone, suppresses leukemia cell growth and impairs lipid metabolism-dependent survival in AML and cytarabine-resistant cells.

Acute myeloid leukemia (AML) is an aggressive hematologic malignancy characterized by uncontrolled proliferation of myeloid precursors and frequent chemotherapy resistance. Jaceosidin (Jace), a polymethoxyflavone with antioxidant and anticancer properties, has not yet been investigated for its therapeutic potential in AML. In this study, we aimed to assess the anti-leukemic potential of Jace using AML cell lines, cytarabine-resistant sublines, patient-derived samples, and zebrafish xenograft models. Jace significantly reduced the viability of AML cells, selectively targeting leukemic cells whi

Quality by design (QbD) development of a Soluplus/TPGS-based ternary amorphous solid dispersion for enhanced oral bioavailability of acacetin.

Acacetin, a natural flavonoid with high-melting-point (> 200 °C) and poorly aqueous solubility, presents significant challenges in oral bioavailability due to its low dissolution rate. To address this limitation, acacetin-loaded amorphous solid dispersion (ASD) were developed via a quality-by-design (QbD) framework and manufactured using solvent-free hot-melt extrusion (HME). The formulation incorporated Soluplus as the polymeric matrix and d-a-tocopherol polyethylene glycol succinate (TPGS) as multifunctional plasticizer. The amorphization of acacetin was confirmed by differential scanning ca

Bioguided Identification of Polymethoxyflavones as Novel Vascular CaV1.2 Channel Blockers from Citrus Peel.

The huge amount of citrus peel produced worldwide represents an economic burden for society. However, this agricultural by-product is a rich source of natural molecules, potentially endowed with interesting pharmacological activities. In this regard, we decided to investigate if the polymethoxyflavones contained in citrus peel waste could be exploited as novel vasorelaxant agents. A hydroalcoholic blond orange (Citrus sinensis) peel extract, obtained by ultrasonication, was partitioned in dichloromethane. Column chromatography allowed for the isolation of four polymethoxyflavones, namely, scut…

Protective effects of myricetin and morin on neurological damage in Aβ1-42/Al3+ -induced Alzheimer's disease model of rats.

Alzheimer's disease (AD) is a degenerative neurological disorder with unclear pathogenesis. Single-target drugs have very limited efficacy in treating AD, but synthetic multi-target drugs have poor efficacy and safety. Therefore, finding suitable natural multi-target drugs against AD is of great interest for research studies. We chose two flavonols, myricetin and morin, for the relevant study. In this study, we used microinjection of Aβ1-42 oligomers into the CA1 region of rat hippocampus, combined with gavage of Aluminum chloride hexahydrate (AlCl3·6H2O) solution to establish AD rat models, a

Tangeretin alleviates Tunicamycin-induced endoplasmic reticulum stress and associated complications in skeletal muscle cells.

Endoplasmic reticulum (ER) stress and associated oxidative stress are involved in the genesis and progression of skeletal muscle diseases such as myositis and atrophy or muscle wasting. Targeting the ER stress and associated downstream pathways can aid in the development of better treatment strategies for these diseases with limited therapeutic approaches. There is a growing interest in identifying natural products against ER stress due to the lower toxicity and cost effectiveness. In the present study, we investigated the protective effect of Tangeretin, a citrus methoxyflavone found in citru

HIMH0021 attenuates ethanol-induced liver injury and steatosis in mice.

Chronic alcohol consumption causes alcohol-induced lipogenesis and promotes hepatic injury by preventing the oxidation of hepatocellular fatty acids through the suppression of the activation of AMP-activated protein kinase (AMPK). HIMH0021, an active flavonoid compound, which is a component of the Acer tegmentosum extract, has been shown to protect against liver damage caused by alcohol consumption. Therefore, in this study, we aimed to determine whether HIMH0021 could regulate alcoholic fatty liver and liver injury in mice. Oral administration of 10 days of Lieber-DeCarli ethanol plus a singl

A citrus polymethoxy flavonoid, nobiletin inhibits sebum production and sebocyte proliferation, and augments sebum excretion in hamsters.

Acne vulgaris is characterized by excess sebum production, and apart from all-trans retinoic acid (atRA) or 13-cis retinoic acid (13-cisRA), there are few effective agents for acne therapy that directly suppresses sebaceous lipogenesis. In this study, we demonstrated that topical application of a citrus polymethoxy flavonoid, nobiletin, to hamster auricles decreased skin surface triacylglycerols (TG) level and the size of sebaceous glands along with inhibition of diacylglycerol acyltransferase (DGAT)-dependent TG synthesis and sebocyte proliferation. The inhibitory actions were similar to that

High-contiguity genome assembly of moth bean (Vigna aconitifolia) reveals genes for disease resistance and flavonoid biosynthesis.

Moth bean (Vigna aconitifolia), a protein-rich, arid-adapted legume of considerable agronomic importance, lacks a whole genome assembly, limiting genome-enabled trait discovery and genetic improvement. In this study, we estimated its genome size using 31.28 Gb of Illumina short-read sequencing data, and a k-mer analysis (k = 21) predicted a genome size of 448.98 Mb. We generated a highly contiguous 420.3 Mb genome assembly using 37.04 Gb of PacBio HiFi long-read sequencing data, corresponding to 82.5× genome coverage. The assembly achieved a contig N50 of 22.30 Mb, with a maximum contig length

Comparison of the Metabolites in Fingered Citron Fruit (Citrus medica L. var. sarcodactylis Swingle) and Chayote (Sechium edule) Based on UPLC-Q-Orbitrap MS/MS.

Fingered citron (Citrus medica L. var. sarcodactylis Swingle) is a medicinal and edible citrus fruit cultivated in several major production regions of China. However, comprehensive information on its regional metabolite variation and its chemical differentiation from chayote (Sechium edule), a botanically unrelated commodity that may be confused with fingered citron because of partially overlapping Chinese vernacular names, remains limited. In this study, untargeted UPLC-Q-Orbitrap MS/MS was used to profile fingered citron samples from Zhejiang (CF1), Sichuan (CF2), Guangdong/Guangxi (CF3), an

Hyperoside modulates bile acid and fatty acid metabolism, presenting a potentially promising treatment for non-alcoholic fatty liver disease.

INTRODUCTION: Non-alcoholic fatty liver disease (NAFLD) is a multifactorial chronic condition that requires a systematic approach for effective management. Multi-effect therapeutic drugs derived from traditional Chinese medicine are increasingly being recognized as promising alternatives for NAFLD intervention. Hyperoside, a natural flavone glycoside found in Cuscuta chinensis Lam, Forsythia suspensa, and Crataegus pinnatifida Bge, has been shown to effectively mitigate NAFLD in rats. However, the underlying mechanism through which hyperoside alleviates NAFLD remains unclear. OBJECTIVE: This

Stepwise co-fermented traditional Chinese medicine byproducts improve antioxidant and anti-inflammatory effects in a piglet model.

BACKGROUND: Lianhua Qingwen capsule is a traditional Chinese medicine (TCM) formula having antiviral and anti-inflammatory activities. During capsule production, a large amount of byproducts will be yielded and disposed of as waste by burying. Resourceful utilization of these kinds of TCM byproducts as feed additives through stage-based co-fermentation using enzyme and probiotics could reduce environmental stress and resource shortage. The in vitro characterization and the supplementary effects of fermented TCM byproducts (FTCM) for weaned piglets (initial body weight: 7.23 ± 0.33 kg; dose: ba

Formation of eicosanoids and other oxylipins in human macrophages.

In this review it is attempted to summarize current studies about formation of eicosanoids and other oxylipins in different human macrophages. There are several reports on M1 and M2 cells, also other phenotypes have been described. The eicosanoids formed in the largest amounts are the COX products TxB2 and PGE2. Thus shortlived bioactive TxA2 is a dominating product both in M1- and in M2-lineages, one exception seems to be MGM-CSF, TGFβ cells. 5-LOX products are produced in both M1 and M2 macrophages, as well as in not fully polarized cells of both lineages. MM-CSF as well as M2 macrophages pr

The protective effect of puerarin-loaded mesoporous silicon nanoparticles on alcoholic hepatitis through mTOR-mediated autophagy pathway.

Puerarin, a bioactive flavone compound isolated from Pueraria (Wild.), provides hepatoprotection by anti-inflammatory, anti-alcoholism, and regulating mechanistic target of rapamycin (mTOR). Building evidence suggests that the activation of mTOR reduces liver injuries associated with alcohol consumption and metabolism. However, the poor water solubility, low bioavailability, and short half-life of puerarin hinder its clinical application. The utility of mesoporous silicon nanoparticles (MSNs) can improve traditional Chinese medicine limitations. Stober methods were used to fabricate MSNs@Pue,
