Individual-Level Prediction of Exposure Therapy Outcome Using Structural and Functional MRI Data in Spider Phobia: A Machine-Learning Study.

Machine-learning prediction studies have shown potential to inform treatment stratification, but recent efforts to predict psychotherapy outcomes with clinical routine data have only resulted in moderate prediction accuracies. Neuroimaging data showed promise to predict treatment outcome, but previous prediction attempts have been exploratory and reported small clinical sample sizes. Herein, we aimed to examine the incremental predictive value of neuroimaging data in contrast to clinical and demographic data alone (for which results were previously published), using a two-level multimodal ense

Neuroimaging Detectable Differences between Parkinson's Disease Motor Subtypes: A Systematic Review.

BACKGROUND: The neuroanatomical substrates of Parkinson's disease (PD) with tremor-dominance (TD) and those with non-tremor dominance (nTD), postural instability and gait difficulty (PIGD), and akinetic-rigid (AR) are not fully differentiated. A better understanding of symptom specific pathoanatomical markers of PD subtypes may result in earlier diagnosis and more tailored treatment. Here, we aim to give an overview of the neuroimaging literature that compared PD motor subtypes. METHODS: A systematic literature review on neuroimaging studies of PD subtypes was conducted according to the Prefe

Analyzing heterogeneity in biomarker discriminative performance through partial time-dependent receiver operating characteristic curve modeling.

This study investigates the heterogeneity of a biomarker's discriminative performance for predicting subsequent time-to-event outcomes across different patient subgroups. While the area under the curve (AUC) for the time-dependent receiver operating characteristic curve is commonly used to assess biomarker performance, the partial time-dependent AUC (PAUC) provides insights that are often more pertinent for population screening and diagnostic testing. To achieve this objective, we propose a regression model tailored for PAUC and develop two distinct estimation procedures for discrete and conti

Multimodal Brain Age Prediction with Feature Selection and Comparison.

Brain age, an estimated biological age from anatomical and/or functional brain imaging data, and its deviation from the chronological age (brain age gap) have shown the potential to serve as biomarkers for characterizing typical brain development, the abnormal aging process, and early indicators of clinical neuropsychiatric problems. In this study, we leverage multimodal brain imaging data for brain age prediction. We studied and compared the performance of individual data modalities (gray matter density in components and regions of interest, cortical and subcortical anatomical features, resti

Electroencephalographic spectral power as a marker of cortical function and disease severity in girls with Rett syndrome.

BACKGROUND: Rett syndrome is a neurodevelopmental disorder caused by a mutation in the X-linked MECP2 gene. Individuals with Rett syndrome typically develop normally until around 18 months of age before undergoing a developmental regression, and the disorder can lead to cognitive, motor, sensory, and autonomic dysfunction. Understanding the mechanism of developmental regression represents a unique challenge when viewed through a neuroscience lens. Are circuits that were previously established erased, and are new ones built to supplant old ones? One way to examine circuit-level changes is with

Functional lesional neurosurgery for tremor-a protocol for a systematic review and meta-analysis.

INTRODUCTION: The recent introduction of incision-less lesional neurosurgery using Gamma Knife and MRI-guided focused ultrasound has revived interest in lesional treatment options for tremor disorders. Preliminary literature researches reveal that the consistency of treatment effects after lesional neurosurgery for tremor has not formally been assessed yet. Similarly, the efficacy of different targets for lesional treatment and incidence of persistent side effects of lesional neurosurgical interventions has not been comprehensively assessed. This work therefore aims to describe a suitable proc

Imaging of the Functional and Dysfunctional Visual System.

Functional magnetic resonance imaging (fMRI) is used clinically to map the visual cortex before brain surgery or other invasive treatments to achieve an optimal balance between therapeutic effect and the avoidance of postoperative vision deficits. Clinically optimized stimuli, analyses, and displays permit identification of cortical subregions supporting high-acuity central vision that is critical for reading and other essential visual functions. A novel data display permits instant appreciation of the functional relationship between the pattern of fMRI brain activation and the pattern of visi

Main effect and interactions of brain regions and gender in the calculation of volumetric asymmetry indices in healthy human brains: ANCOVA analyses of in vivo 3T MRI data.

INTRODUCTION: Macroanatomical right-left hemispheric differences in the brain are termed asymmetries, although there is no clear information on the global influence of gender and brain-regions. The aim of this study was to evaluate the main effects and interactions of these variables on the measurement of volumetric asymmetry indices (VAIs). MATERIALS AND METHODS: Forty-seven healthy young-adult volunteers (23 males, 24 females) agreed to undergo brain magnetic resonance imaging in a 3T scanner. Image post processing using voxel-based volumetry allowed the calculation of 54 VAIs from the fron

Brain imaging in fibromyalgia.

Fibromyalgia is a primary brain disorder or a result of peripheral dysfunctions inducing brain alterations, with underlying mechanisms that partially overlap with other painful conditions. Although there are methodologic variations, neuroimaging studies propose neural correlations to clinical findings of abnormal pain modulation in fibromyalgia. Growing evidences of specific differences of brain activations in resting states and pain-evoked conditions confirm clinical hyperalgesia and impaired inhibitory descending systems, and also demonstrate cognitive-affective influences on painful experie

Integration of "what" and "where" in frontal cortex during visual imagery of scenes.

Imagination is a key function for many human activities, such as reminiscing, learning, or planning. Unravelling its neuro-biological basis is paramount to grasp the essence of our thoughts. Previous neuroimaging studies have identified brain regions subserving the visualisation of "what?" (e.g. faces or objects) and "where?" (e.g. spatial layout) content of mental images. However, the functional role of a common set of involved regions - the frontal regions - and their interplay with the "what" and "where" regions, has remained largely unspecified. This study combines functional MRI and elect

NeuroBridge ontology: computable provenance metadata to give the long tail of neuroimaging data a FAIR chance for secondary use.

BACKGROUND: Despite the efforts of the neuroscience community, there are many published neuroimaging studies with data that are still not findable or accessible. Users face significant challenges in reusing neuroimaging data due to the lack of provenance metadata, such as experimental protocols, study instruments, and details about the study participants, which is also required for interoperability. To implement the FAIR guidelines for neuroimaging data, we have developed an iterative ontology engineering process and used it to create the NeuroBridge ontology. The NeuroBridge ontology is a com

Lessons from human neuroimaging on the developing cerebellum in health and disease.

The cerebellum is a critical structure for both motor and cognitive behaviors. Although the majority of human neuroimaging research has examined the cerebellum in adults, this "little brain" may be especially pivotal during development. Indeed, the cerebellum has a unique developmental timecourse relative to the cerebral cortex, suggesting that it may play a unique role in the development of motor and cognitive functions. Here, we review the literature on cerebellar development with a focus on neuroimaging studies in humans. We discuss several structural and functional MRI studies that examine

Neuroimaging of the ictal-interictal continuum: Where do we stand? A systematic review and meta-analysis of current evidence.

OBJECTIVE: Patterns within the ictal-interictal continuum (IIC) are frequently encountered in critically ill patients undergoing continuous electroencephalographic studies and may reflect ongoing ictal activity or secondary brain injury. Peri-ictal neuroimaging abnormalities (PNA) have been described in association with status epilepticus (SE), but their occurrence in patients with IIC across different neuroimaging modalities remains incompletely characterized. We aimed to assess the occurrence of PNA in adults with IIC across structural and functional neuroimaging techniques. METHODS: We per…

Neuroimaging in lesioning therapy for obsessive-compulsive disorder: region-based and network analysis of preoperative outcome predictors and postoperative effects.

INTRODUCTION: Lesioning procedures such as capsulotomy and cingulotomy offer therapeutic options for patients with severe, treatment-resistant obsessive-compulsive disorder (OCD). However, treatment response remains variable, and mechanisms underlying clinical improvement are poorly understood. Neuroimaging may help identify response predictors and elucidate mechanisms of action. METHODS: We systematically reviewed neuroimaging findings from studies of lesioning therapy for OCD, focusing on preoperative predictors of response and postoperative imaging changes. To synthesize findings, we perfo

An overview of the first 5 years of the ENIGMA obsessive-compulsive disorder working group: The power of worldwide collaboration.

Neuroimaging has played an important part in advancing our understanding of the neurobiology of obsessive-compulsive disorder (OCD). At the same time, neuroimaging studies of OCD have had notable limitations, including reliance on relatively small samples. International collaborative efforts to increase statistical power by combining samples from across sites have been bolstered by the ENIGMA consortium; this provides specific technical expertise for conducting multi-site analyses, as well as access to a collaborative community of neuroimaging scientists. In this article, we outline the backgr
