Generally, chemical tissue clearing is performed by a solution consisting of two parts benzyl benzoate and one part benzyl alcohol. However, prolonged exposure to this mixture markedly reduces the fluorescence of GFP expressing specimens, so that one has to compromise between clearing quality and fluorescence preservation. This can be a severe drawback when working with specimens exhibiting low GFP expression rates. Sugarcane bagasse is a potential lignocellulosic feedstock for ethanol production, since it is cheap, readily available, and has a high carbohydrate content. We decipher functions of three monooxygenases (PasO1, PasO3, and PasO4) in early biosynthetic steps in spirotetronate PA-46101 and demonstrate a PasO4/PasO3 cascade for forging the signature macrocyclic lactone. The P450 PasO4 oxidizes a methyl group to a carboxylate, enabling a regiospecific Baeyer-Villiger oxidation by PasO3. Structural analysis of a PasO3 homologue identifies a carboxylate-binding pocket essential for this strict substrate specificity. This two-enzyme cascade can serve as a portable biocatalytic tool for diversifying spirotetronates.

The organosolv lignin of palm kernel shell (PKS) was prepared by three different organic solvents (polyethylene glycol (PEG), tetrahydrofuran (THF), and formic acid (FA)) with a 70% mixture of solvents in water, using 2% w/w H2SO4 as a catalyst at 190 °C for 1 h. To elucidate the characteristics and evolution of eating quality in tofu balls during preprocessing, cooking, and frozen reheating, volatile compounds and textural properties were systematically analyzed at each stage.
