From molecules to field: Integrated insights into cuticle-mediated drought tolerance in plants.

Drought stress exacerbates non-stomatal water loss, which hinders agricultural growth and global food security. The cuticle, a layer of cutin, waxes, and other polymers, protects plants from the negative impact of drought. One crucial adaptation that has enabled terrestrial plants to survive and flourish in drought-prone regions is the development of cuticle structures. In this review, we (1) provide an overview of the molecular and enzymatic pathways involved in the biosynthesis of cutin and wax; (2) analyze important regulatory networks that control the formation of cuticles, such as abscisi

Aspartic Acid Catalyzed by Asparaginase Provides Rice with Broad-Spectrum Resistance.

Amino acid metabolism plays a key role in regulating interactions between plants and pathogens. Asparaginase hydrolyzes asparagine, a major nitrogen transport and storage amino acid, into aspartate. However, the immune function of plants asparaginase remains largely unknown. Rice possesses two asparaginase genes, OsASNase1 and OsASNase2, the function of which is unknown. In this study, we revealed that both OsASNase1 and OsASNase2 are cytoplasmic asparaginases that hydrolyze asparagine into aspartic acid and ammonia. Overexpression of OsASNase1 and OsASNase2 enhanced rice resistance to bacteri

SlJMJ12 overexpression confers improved cadmium tolerance in Arabidopsis and tomato.

Cadmium (Cd) is a toxic heavy metal threatening ecosystems and crop production. The functions of histone demethylases in plant Cd tolerance remain poorly understood. We functionally characterized the tomato histone demethylase SlJMJ12 using Arabidopsis and tomato. Cd rapidly induces leaf-expressed SlJMJ12, with peak expression at 3 h post-treatment. Compared with the wild type, SlJMJ12 loss-of-function mutants show Cd sensitivity in both Arabidopsis and tomato, with reduced biomass, higher plant Cd and ROS levels, and decreased antioxidant enzyme activities, whereas overexpression improves Cd

A mutant allele of the long-day floral activator NtFT5 is associated with the short-day-specific flowering of tobacco cultivar Maryland Mammoth.

Flowering in day-neutral tobacco (Nicotiana tabacum) cultivars requires the photoperiod-dependent expression of FLOWERING LOCUS T (FT)-like floral activators, which compete with FT-like inhibitors to interact with FD proteins and induce the switch from vegetative to reproductive growth. In the short-day (SD) cultivar Maryland Mammoth (MM), vegetative growth persists under long-day (LD) conditions, generating unusually tall plants. We found that overexpression of the major LD floral inducer (NtFT5) from the day-neutral cultivar Hicks induces flowering in MM plants under LD conditions. However,

Pan-Genome Analysis of the Tubulin Gene Family Reveals Candidates for Fiber Strength in Gossypium barbadense.

Background/Objectives: Tubulins (Tub) are central components of microtubules, but intraspecific variation and developmental expression of the Tub family in Gossypium barbadense remain poorly characterized. This study aimed to characterize the GbTub family using a pan-genome framework and identify candidates associated with fiber development and strength. Methods: A total of 50 GbTub genes were identified in the G. barbadense 3-79 reference genome, and their orthologous presence-absence patterns were subsequently assessed across 12 additional G. barbadense accessions. Phylogenetic, presence-abs

A regulatory module integrates lipid metabolism and cell wall remodeling to establish the seed coat palisade barrier.

Seed physical dormancy is a crucial adaptive trait conferred by the water-impermeable seed coat, particularly its specialized palisade layer. Although the structural components of this barrier, including the cuticle and cell wall polysaccharides, are well characterized, the molecular mechanisms coordinating their formation and remodeling remain poorly understood. Here, using single-nucleus RNA sequencing of Medicago truncatula, we identify a regulatory module essential for palisade layer formation and the establishment of physical dormancy. We show that MtKNOX4 interacts with cytoplasmic MtSAW

AUXIN RESPONSE FACTORs: from transcription factors to auxin effectors.

Signalling pathways have originated and diversified extensively during evolution. Using AUXIN RESPONSE FACTORs (ARFs) as a model, we examine how a plant signalling system evolved through the co-option of pre-existing transcriptional regulators, a principle well established in evolutionary biology in organisms, such as animals. ARFs are an ancient family of DNA-binding proteins that predate the canonical nuclear auxin pathway and likely acted as auxin-independent transcriptional repressors. The emergence of auxin signalling involved coupling this ancestral transcriptional module to a regulatory

The 3R-MYB transcription factor VvMYB3R5 regulates Colletotrichum viniferum resistance through affecting proanthocyanidin synthesis in grapevine.

Grape ripe rot (Colletotrichum spp.), a significant fungal disease, causes substantial economic losses. Although multiple MYB transcription factors have been reported to be involved in plant disease resistance against ripe rot, the specific functions and mechanisms of 3R-MYB subfamily members in grape responses to fungal pathogen infection remain unknown. Here, we identified and functionally characterized a 3R-MYB transcription factor, VvMYB3R5, for the first time, revealing its crucial positive regulatory role in grape resistance to C. viniferum and in proanthocyanidin (PA) accumulation. We d

StCYP90C1 modulates plant architecture via regulating brassinosteroid biosynthesis in potato.

Brassinosteroid (BR) exert a crucial regulatory role in shaping plant architecture. However, the BR biosynthesis and underlying genetic variations governing potato plant architecture remain poorly understood. Here, we identified a single recessive locus, ap, which controls the abnormal architectural plant (AP) with dark-green curled leaves, shortened internodes, and delayed flowering, using a BC4S1 population. The functional gene, StCYP90C1, encoding 3-epi-6-deoxocathasterone 23-monooxygenase, was identified at this locus. StCYP90C1 expression was significantly lower in the leaves of APs than

MicroRNA156-SPL13B Module Induces Parthenocarpy Through the Gibberellin Pathway.

Fruit development typically follows pollination and fertilisation. However, parthenocarpy, the formation of seedless fruits, represents an alternative pathway. The fruit that grows from the ovary under unpollinated and/or unfertilised conditions is termed parthenocarpic. This trait is not only desirable to consumers but also contributes to yield stability under adverse environmental conditions. In this study, we demonstrate that overexpression of miR156h regulates parthenocarpy under unpollinated conditions via a gibberellic acid (GA)-dependent pathway. MdSPL13B was found to be involved in Mdm

Evolutionarily New Solanaceous DCL2 Family Members Acquire Functions in Tomato.

RNA silencing is one of the major defence activities against viral pathogens in plants. Silencing signals are initiated by Dicer-like proteins (DCLs) to generate viral-derived small RNAs (sRNAs). Viral sRNAs are then loaded into Argonaute proteins to form an RNA-induced silencing complex to guide cleavage of target RNAs based on sequence homology. While the model regarding RNA silencing-mediated defence against viral pathogens is largely established based on extensive studies using the model plant Arabidopsis thaliana, there are diverse sets of silencing components in other plants, especially

A fasciclin-like arabinogalactan protein OsFLA8 is involved in Casparian strip formation and salt tolerance in rice.

FASCICLIN-LIKE ARABINOGALACTAN PROTEINs (FLAs), a subclass of plant arabinogalactan proteins (AGPs), play diverse roles in plant development, including pollen development, secondary cell wall regulation, and root and stem growth. However, the biological functions of the majority of FLA-encoded proteins in rice remain largely unexplored, particularly their roles in root development and cell wall formation. In this study, we identified OsFLA8, a member of the FLA family, as a critical component of Casparian strip (CS) formation and salt stress tolerance in rice. Subcellular localization and expr

Variation in a single allele drives divergent yield responses to elevated CO2 between rice subspecies.

Rising atmospheric CO2 generally increases yield of indica rice, one of the two main Asian cultivated rice subspecies, more strongly than japonica rice, the other main subspecies. The molecular mechanisms driving this difference remain unclear, limiting the potential of future rice yield increases through breeding efforts. Here, we show that between-species variation in the DNR1 (DULL NITROGEN RESPONSE1) allele, a regulator of nitrate-use efficiency in rice plants, explains the divergent response to elevated atmospheric CO2 (eCO2) conditions. eCO2 increased rice yield by 22.8-32.3% in plants c

Phenylalanine metabolism-dependent lignification confers rhizobacterium-induced plant resistance.

Phenylalanine metabolism serves as an important route for the production of diverse secondary metabolites including phenylpropanoids. The phenylpropanoid pathway is involved in plant immunity, but whether it can regulate rhizobacteria-induced resistance is poorly understood. In this study, we confirmed a growth-promoting rhizobacterium strain JR48 could induce resistance, strengthen salicylic acid (SA) signaling, and increase lignin content during Phytophthora capsici infection. We conducted transcriptome sequencing to analyze the effect of JR48 on the expression of pepper (Capsicum annuum L.)

Co-elevated atmospheric CO2 and temperature increase rice yield but degrade nutrient quality in a cold temperature region.

The responses of three Japonica rice cultivars were investigated throughout a full growing season in computer-controlled open-top chambers under four treatments: control (CK), elevated CO2 (700 ppm, eC), elevated temperature (+2 °C, eT), and their combination (eCeT). To assess the impact of climate change factors, the present study performed comprehensive physiological measurements and flag leaf transcriptomic analysis to evaluate rice yield and quality. Both eC and eCeT synergistically upregulated genes related to photosystem II (PsbA) and photosystem I (PsaA/B/C), thereby enhancing photochem

Metabolite and transcriptome reveal the lipid-associated key components and genes regulated by BoORP3a in ornamental kale.

BoORP3a, an oxysterol-binding protein, located in the endoplasmic reticulum (ER), may function in cuticular wax deposition in ornamental kale. In this study, we investigated its regulation of the key components of cuticular wax and lipids, metabolic pathways, and potential target genes. HS-SPME/GC-MS identified 34 and 31 volatile organic compounds in wild-type and the BoORP3a-overexpressing plant OE-ORP3a-7, respectively, primarily including alkane, ketone, ester, and alcohol. Hentriacontane, 15-nonacosanone, and > C20 alkanes were more abundant in OE-ORP3a-7, which may result in more cuticula

Selenate simultaneously alleviated cadmium and arsenic accumulation in rice (Oryza sativa L.) via regulating transport genes.

Cadmium (Cd) and arsenic (As) have contrasting biogeochemical behaviors in paddy soil, which posed an obstacle for reducing their accumulation in rice (Oryza sativa L.) simultaneously. In this study, selenate exhibited a more effective ability than selenite on simultaneous alleviation of Cd and As accumulation in rice under Cd-As co-exposure, and the mechanisms need to be further investigated. The results showed that selenate significantly decreased the root Cd and As contents by 59%-83% and 43%-72% compared to Cd-As compound exposure, respectively. Correspondingly, it significantly down-regul
