Multidrug Resistance and Virulence Profiles of Salmonella Isolated from Swine Lymph Nodes.

Salmonella spp. is a foodborne pathogen present in the pork production chain, leading to potential contamination of end products and causing salmonellosis cases and outbreaks worldwide. The emergence of multidrug-resistant (MDR) Salmonella spp., especially isolates obtained from animal origin food, is a global concern. This study aimed to isolate Salmonella from swine mesenteric lymph nodes (MLN) and to characterize the virulence and antibiotic resistance profiles. MLN samples were obtained from a swine slaughterhouse and subjected to Salmonella spp. isolation. Ten MLN samples were positive an

Vital and dispensable roles of Plasmodium multidrug resistance transporters during blood- and mosquito-stage development.

Multidrug resistance (MDR) proteins belong to the B subfamily of the ATP Binding Cassette (ABC) transporters, which export a wide range of compounds including pharmaceuticals. In this study, we used reverse genetics to study the role of all seven Plasmodium MDR proteins during the life cycle of malaria parasites. Four P. berghei genes (encoding MDR1, 4, 6 and 7) were refractory to deletion, indicating a vital role during blood stage multiplication and validating them as potential targets for antimalarial drugs. Mutants lacking expression of MDR2, MDR3 and MDR5 were generated in both P. berghei

Characterization of a lytic Staphylococcus aureus phage JAsa55 and its biocontrol potential in milk.

Staphylococcus aureus (S. aureus) is a prominent foodborne pathogen that frequently causes food poisoning by contaminating various foods, particularly dairy products, and poses severe threats to food safety and public health. The increasing prevalence of multidrug-resistant S. aureus strains has further limited the efficacy of conventional interventions. In this study, we isolated and characterized a novel lytic phage JAsa55 using an MDR S. aureus strain as host. Transmission electron microscopy showed that JAsa55 possesses an icosahedral head and a long non-contractile tail, consistent with a

Wastewater treatment configuration shapes ESBL- and KPC-producing gram-negative bacteria in effluents from a Colombian city: a one health perspective.

Wastewater-based surveillance is increasingly used to track antimicrobial resistance (AMR), yet evidence from treatment configurations common in small and mid-sized cities, particularly upflow anaerobic sludge blanket (UASB) reactors and stabilization ponds, remains limited. We conducted culture-based surveillance in two full-scale municipal wastewater treatment systems in Colombia: a UASB-trickling filter line and a stabilization pond system. Influent, biological-stage, and effluent samples were analyzed for extended-spectrum β-lactamase (ESBL)-associated and carbapenem-resistant Gram-negativ

Clonal spread of blaCTX-M-65 producing Salmonella enterica serovars detected in poultry retail meat in North Carolina, USA.

The rise of antimicrobial resistance (AMR) in Salmonella enterica poses a significant public health threat, particularly through the dissemination of extended-spectrum β-lactamase (ESBL) genes such as blaCTX-M-65. This study investigated the prevalence, resistance profiles, and genomic characteristics of S. enterica isolates from retail poultry products in North Carolina, collected between 2020 and 2024. Among 132 isolates representing 25 serovars, 14 were identified as multidrug-resistant (MDR) strains harboring blaCTX-M-65. Whole-genome sequencing revealed that these isolates belonged to thr

Phage Host Range Expansion Through Directed Evolution on Highly Phage-Resistant Strains of Klebsiella pneumoniae.

Multidrug-resistant (MDR) strains of Klebsiella pneumoniae present an acute threat as they continue to disseminate globally. Phage therapy has shown promise as a powerful approach to combat MDR infections, but narrow phage host ranges make development of broad acting therapeutics more challenging. The goal of this effort was to use in vitro directed evolution (the "Appelmans protocol") to isolate K. pneumoniae phages with broader host ranges for improved therapeutic cocktails. Five myophages in the genus Jiaodavirus (family Straboviridae) with complementary activity were mixed and passaged aga

Screening of Klebsiella pneumoniae subsp. pneumoniae Strains with Multi-Drug Resistance and Virulence Profiles Isolated from an Italian Hospital between 2020 and 2023.

Klebsiella pneumoniae strains that are resistant to multiple drugs (KPMDRs), which are often acquired in hospital settings and lead to healthcare-associated infections, pose a serious public health threat, as does hypervirulent K. pneumoniae (hvKp), which can also cause serious infections in otherwise healthy individuals. The widespread and often unnecessary use of antibiotics seen during the recent COVID-19 pandemic has exacerbated the challenges posed by antibiotic resistance in clinical settings. There is growing concern that hypervirulent (hvKp) strains may acquire genes that confer antimi

Global colistin use: a review of the emergence of resistant Enterobacterales and the impact on their genetic basis.

The dramatic global rise of MDR and XDR Enterobacterales in human medicine forced clinicians to the reintroduction of colistin as last-resort drug. Meanwhile, colistin is used in the veterinary medicine since its discovery, leading to a steadily increasing prevalence of resistant isolates in the livestock and meat-based food sector. Consequently, transmission of resistant isolates from animals to humans, acquisition via food and exposure to colistin in the clinic are reasons for the increased prevalence of colistin-resistant Enterobacterales in humans in the last decades. Initially, resistance

Antimicrobial Susceptibility and Frequency of bla and qnr Genes in Salmonella enterica Isolated from Slaughtered Pigs.

Salmonella enterica is known as one of the most common foodborne pathogens worldwide. While salmonellosis is usually self-limiting, severe infections may require antimicrobial therapy. However, increasing resistance of Salmonella to antimicrobials, particularly fluoroquinolones and cephalosporins, is of utmost concern. The present study aimed to investigate the antimicrobial susceptibility of S. enterica isolated from pork, the major product in Philippine livestock production. Our results show that both the qnrS and the blaTEM antimicrobial resistance genes were present in 61.2% of the isolate

The culturable soil antibiotic resistome: a community of multi-drug resistant bacteria.

Understanding the soil bacterial resistome is essential to understanding the evolution and development of antibiotic resistance, and its spread between species and biomes. We have identified and characterized multi-drug resistance (MDR) mechanisms in the culturable soil antibiotic resistome and linked the resistance profiles to bacterial species. We isolated 412 antibiotic resistant bacteria from agricultural, urban and pristine soils. All isolates were multi-drug resistant, of which greater than 80% were resistant to 16-23 antibiotics, comprising almost all classes of antibiotic. The mobile r

High prevalence of class 1 integron, biofilm formation, and antimicrobial resistance profiles of Escherichia coli isolated from cattle, water, and soil in Bangladesh.

BACKGROUND: Antimicrobial resistance (AMR) is a global threat. Escherichia coli is an important reservoir of resistance genes, and the coexistence of mcr with class 1 integron (intI1) is alarming, mcr confers resistance to colistin, a last-resort antibiotic, while intI1 often carries multiple transferable resistance determinants. Although colistin use in livestock is banned in Bangladesh, data on colistin-resistant E. coli in food animals, particularly cattle, remain scarce. This study investigated multidrug-resistance patterns, biofilm formation, and the prevalence of intI1 and its associatio

Genetic characterization, mechanisms and dissemination risk of antibiotic resistance of multidrug-resistant Rothia nasimurium.

Rothia nasimurium is part of the commensal flora of humans and other animals and has recently received increasing attention for its multidrug-resistance (MDR) and pathogenicity. Currently, no systematic reports characterize the genetics, mechanisms, and dissemination risks of antibiotic resistance in MDR R. nasimurium. Here, we present the first report outlining a MDR strain of R. nasimurium, E1706032a, isolated from ducks exhibiting clinical sickness. Phylogenetic analysis indicates that E1706032a mostly likely originated in the commensal bacteria of Amazona aestiva in Florida. E1706032a is r

Consecutive salinomycin treatment reduces doxorubicin resistance of breast tumor cells by diminishing drug efflux pump expression and activity.

Chemoresistance is a major challenge for the successful therapy of breast cancer. The discovery of salinomycin as an anticancer stem cell drug provides progress in overcoming chemoresistance. However, it remains to be elucidated whether salinomycin treatment is able to sensitize cancer cells to chemotherapeutic drugs. In the present study, we consecutively treated epithelial MCF-7 and BT-474 breast cancer cells as well as mesenchymal MDA-MB 231 and MDA-MB 436 cells with salinomycin, and analyzed the gene expression of the two prominent multiple drug resistance (MDR) genes, MDR1 and BCRP1. Defective mitochondrial disulfide relay system, altered mitochondrial morphology and function in Huntington's disease.

A number of studies have been conducted that link mitochondrial dysfunction (MD) to Huntington's disease (HD); however, contradicting results had resulted in a lack of a clear mechanism that links expression of mutant Huntingtin protein and MD. Mouse homozygous (HM) and heterozygous (HT) mutant striatal cells with two or one allele encoding for a mutant huntingtin protein with 111 polyGln repeats showed a significant impairment of the mitochondrial disulfide relay system (MDRS). This system (consisting of two proteins, Gfer and Mia40) is involved in the mitochondrial import of Cys-rich protein
