A practical, algorithmic approach to diagnosing hair shaft disorders.

The hair shaft is a unique structure composed of an inner cortex and a protective outer cuticle. Any defects in this normal structure due to genetics or the environment can lead to variations in physical properties. Thus one should suspect a hair shaft disorder if a patient presents with an abnormality or change in hair texture, appearance, manageability or ability to grow hair long. A key feature of the clinical evaluation is to determine whether there is hair breakage (increased fragility) by looking for broken hairs and performing a tug test. Once this determination is made, an algorithmic

Charting the computational landscape of single-cell genetic perturbation.

BACKGROUND: Single-cell genetic perturbation screens are increasingly used to study gene function at single-cell resolution. However, their analysis remains difficult because available computational methods differ in assumptions, input requirements, and output definitions, which can complicate cross-study comparison and consistent tool selection. AIM OF REVIEW: We summarize current methods by organizing single-cell genetic perturbation analysis into a five-step computational workflow: basic data preprocessing, data refinement and correction, perturbation-specific quality control and filtering

Eyebrow Madarosis: An Updated Review of the Etiology and Management, Part II- Scarring Disorders.

BACKGROUND: Eyebrows play an essential role in facial symmetry, communication, and aesthetic identity. Scarring eyebrow alopecia results from inflammatory, autoimmune, infectious, genetic, traumatic, and iatrogenic processes that irreversibly damage follicular stem cells and limit regrowth potential. Despite its clinical and psychosocial impact, evidence-based guidance for diagnosis and management remains limited. METHODS: A comprehensive PubMed search (1960–2025) was performed using terms related to "eyebrow alopecia," "eyebrow madarosis," and etiology-specific conditions. English-lang

Interactive networks of donors and recipients of the TetM gene and its evolutionary dynamics across the bacterial domain.

Identifying primary donors and recipients of the tetM gene is crucial for gaining a deeper understanding of the dynamics underlying the dissemination of resistance to tetracyclines in natural bacterial populations, including those of human and animal pathogens. This study modeled the major donor-recipient network of tetM, also providing important insights into the primary evolutionary mechanisms of this gene. The RDP4- and SplitsTree-embedded algorithms were used to detect genetic recombination events of tetM loci from different bacterial species and genera. FUBAR, MEME, and MEGA11 were employ

The interplay of age, gender and amyloid on brain and cognition in mid-life and older adults.

Deficits in memory are seen as a canonical sign of aging and a prodrome to dementia in older adults. However, our understanding of age-related cognition and brain morphology occurring throughout a broader spectrum of adulthood remains limited. We quantified the relationship between cognitive function and brain morphology (sulcal width, SW) using three cross-sectional observational datasets (PISA, AIBL, ADNI) from mid-life to older adulthood, assessing the influence of age, sex, amyloid (Aβ) and genetic risk for dementia. The data comprised cognitive, genetic and neuroimaging measures of a tota

Genotypic and phenotypic features of 39 Chinese patients with glycogen storage diseases type I, VI, and IX.

Glycogen storage diseases (GSDs) are abnormally inherited glycogen metabolism mainly affecting the liver, muscles, and heart. Deficiency of proteins involved in glycogen metabolism caused by genetic mutations are responsible for different subtype of GSDs. However, there are still some challenges in diagnosing GSD. This study includes 39 suspected GSDs patients from unrelated families in China. Next-generation sequencing (NGS) was used to investigate the reason for their diseases at the genetic level. Finally, all 39 patients were diagnosed with GSDs, including 20 GSD-Ia, 4 GSD-VI, and 15 GSD I

What's new in pediatric genetic cholestatic liver disease: advances in etiology, diagnostics and therapeutic approaches.

PURPOSE OF REVIEW: To highlight recent advances in pediatric cholestatic liver disease, including promising novel prognostic markers and new therapies. FINDINGS: Additional genetic variants associated with the progressive familial intrahepatic cholestasis (PFIC) phenotype and new genetic cholangiopathies, with an emerging role of ciliopathy genes, are increasingly being identified. Genotype severity predicts outcomes in bile salt export pump (BSEP) deficiency, and post-biliary diversion serum bile acid levels significantly affect native liver survival in BSEP and progressive familial intrahep

Clinically accessible amplitude-based multiplex ddPCR assay for tryptase genotyping.

Hereditary α tryptasemia (HαT) is an autosomal dominant trait characterized by increased TPSAB1 copy number (CN) encoding α-tryptase. The determination of HαT is being discussed as an important biomarker to be included in risk assessment models and future diagnostic algorithms for patients with mastocytosis and anaphylaxis. Due to the complex genetic structure at the human tryptase locus, genetic testing for tryptase gene composition is presently notably limited and infrequently pursued. This study aimed to develop, optimise and validate a multiplex droplet digital PCR (ddPCR) assay that can r

Evaluating the performance of a clinical genome sequencing program for diagnosis of rare genetic disease, seen through the lens of craniosynostosis.

PURPOSE: Genome sequencing (GS) for diagnosis of rare genetic disease is being introduced into the clinic, but the complexity of the data poses challenges for developing pipelines with high diagnostic sensitivity. We evaluated the performance of the Genomics England 100,000 Genomes Project (100kGP) panel-based pipelines, using craniosynostosis as a test disease. METHODS: GS data from 114 probands with craniosynostosis and their relatives (314 samples), negative on routine genetic testing, were scrutinized by a specialized research team, and diagnoses compared with those made by 100kGP. The impact of post-alignment processing procedures on whole-exome sequencing data.

The use of post-alignment procedures has been suggested to prevent the identification of false-positives in massive DNA sequencing data. Insertions and deletions are most likely to be misinterpreted by variant calling algorithms. Using known genetic variants as references for post-processing pipelines can minimize mismatches. They allow reads to be correctly realigned and recalibrated, resulting in more parsimonious variant calling. In this work, we aim to investigate the impact of using different sets of common variants as references to facilitate variant calling from whole-exome sequencing d

Primary immunodeficiencies and invasive fungal infection: when to suspect and how to diagnose and manage.

PURPOSE OF REVIEW: Invasive fungal infections (IFIs) most often occur secondary to acquired immunodeficiency states such as transplantation, AIDS or immune-modulatory treatment for neoplastic and autoimmune disorders. Apart from these acquired conditions, several primary immunodeficiency disorders (PIDs) can present with IFIs in the absence of iatrogenic immunosuppression. This review highlights recent advances in our understanding of PIDs that cause IFIs, which may help clinicians in the diagnosis and management of such infections. RECENT FINDINGS: A growing number of PIDs that cause varying

Blood-based biomarkers predicting response to antidepressants.

Major depressive disorder is a common, serious and in some cases, life-threatening condition and affects approximately 350 million people globally. Although there is effective treatment available for it, more than 50% of the patients fail to respond to the first antidepressant they receive. The selection of a distinct treatment is still exclusively based on clinical judgment without incorporating lab-derived objective measures. However, there is growing evidence of biomarkers that it helps to improve diagnostic processes and treatment algorithms. Here genetic markers and blood-based biomarkers

High-resolution HLA haplotype frequencies of stem cell donors in Germany with foreign parentage: how can they be used to improve unrelated donor searches?

In hematopoietic stem cell transplantation, human leukocyte antigens (HLA), usually HLA loci A, B, C, DRB1 and DQB1, are required to check histocompatibility between a potential donor and the recipient suffering from a malignant or non-malignant blood disease. As databases of potential unrelated donors are very heterogeneous with respect to typing resolution and number of typed loci, donor registries make use of haplotype frequency-based algorithms to provide matching probabilities for each potentially matching recipient/donor pair. However, it is well known that HLA allele and haplotype frequ

Prediction of conversion to psychosis: review and future directions.

This article reviews recent findings on predictors of conversion to psychosis among youth deemed at ultra high risk (UHR) based on the presence of subpsychotic-intensity symptoms or genetic risk for psychosis and a recent decline in functioning. Although transition rates differ between studies, the most well powered studies have observed rates of conversion to full psychosis in the 30-40% range over 2-3 years of follow-up. Across studies, severity of subthreshold positive symptoms, poorer social functioning, and genetic risk for schizophrenia appear to be consistent predictors of conversion to

Demand side management with electric vehicles and optimal renewable resources integration under system uncertainties.

The rapid growth of integrating electrical vehicles (EVs) into the distribution network has introduced complexities and power flow inefficiencies. To address these challenges, optimal renewable energy resources (RERs) integration along with applied demand-side management (DSM) contribute to managing load profiles and generation thus reducing costs. This should be smartly attained through selecting efficient optimization techniques to improve power quality, voltage profile, and reliability. This paper aims to investigate the effect of integrating EVs and applying peak load shifting (PLS) as a D

Combining RGB imaging with a two-stage deep learning method to reveal genetic variation of wheat sprouting traits.

Wheat emergence rate and emergence uniformity are key indicators for evaluating seed vigor and sowing quality, and they play an important role in wheat growth and yield formation. Traditional methods for measuring emergence rate and evaluating emergence uniformity rely on manual assessment, which is inefficient, highly subjective, and unable to meet the demand for large scale, high efficiency, and precise acquisition of wheat emergence data. In this study, RGB images and a two-stage deep learning algorithm were used to extract and analyze seedling traits of 420 wheat varieties under two nitrog

A multi-objective optimization method for seed distribution in prostate cancer low-dose-rate brachytherapy.

BACKGROUND: Brachytherapy planning places radioactive sources around the tumor target to ensure that the tumor is covered by the prescribed dose while maximizing dose sparing for normal organs. In brachytherapy, the mainstream inverse planning methods are Hybrid Inverse Planning Optimization and Inverse Planning Simulated Annealing. Most existing algorithms use a single objective function with weighting parameters to balance tumor and normal tissue importance, but choosing these weights is often subjective and difficult, which can result in biased or suboptimal plans. PURPOSE: To propose a se

Leveraging Natural Language Processing to Accurately Identify Histologically Confirmed Esophageal and Gastric Premalignant and Malignant Conditions Using a Large-Scale Health-Record Linked Genomic Biobank.

BACKGROUND AND AIMS: Our understanding of the epidemiology and natural history of gastroesophageal precancerous conditions is limited by a lack of robust analyses using large-scale individual-level data. We aimed to develop and validate a natural language processing (NLP) algorithm to identify and phenotype esophageal and gastric precancerous conditions and cancer and apply this to the Million Veteran Program (MVP), a uniquely powerful nationwide genomic biobank linked to individual-level electronic health record data. METHODS: We identified 121,808 individuals in MVP who underwent upper endo

Heart rate monitoring and therapeutic devices: A wavelet transform based approach for the modeling and classification of congestive heart failure.

Heart rate monitoring and therapeutic devices include real-time sensing capabilities reflecting the state of the heart. Current circuitry can be interpreted as a cardiac electrical signal compression algorithm representing the time signal information into a single event description of the cardiac activity. It is observed that some detection techniques developed for ECG signal detection like artificial neural network, genetic algorithm, Hilbert transform, hidden Markov model are some sophisticated algorithms which provide suitable results but their implementation on a silicon chip is very compl

Analysis and multiclass classification of pathological knee joints using vibroarthrographic signals.

BACKGROUND AND OBJECTIVE: Vibroarthrography (VAG) is a method developed for sensitive and objective assessment of articular function. Although the VAG method is still in development, it shows high accuracy, sensitivity and specificity when comparing results obtained from controls and the non-specific, knee-related disorder group. However, the multiclass classification remains practically unknown. Therefore the aim of this study was to extend the VAG method classification to 5 classes, according to different disorders of the patellofemoral joint. METHODS: We assessed 121 knees of patients (95
