Population Genetics of Phlebotomus papatasi from Endemic and Nonendemic Areas for Zoonotic Cutaneous Leishmaniasis in Morocco, as Revealed by Cytochrome Oxidase Gene Subunit I Sequencing.

Zoonotic cutaneous leishmaniasis (ZCL) caused by Leishmania major Yakimoff & Shokhor and transmitted by Phlebotomus papatasi (Scopoli) is a public health concern in Morocco. The disease is endemic mainly in pre-Saharan regions on the southern slope of the High Atlas Mountains. The northern slope of the High Atlas Mountains and the arid plains of central Morocco remain non-endemic and are currently considered high risk for ZCL. Here we investigate and compare the population genetic structure of P. papatasi populations sampled in various habitats in historical foci and non-endemic ZCL areas. A f

Combining spatial analysis and host population genetics to gain insights into the mode of transmission of a pathogen: The example of Toxoplasma gondii in mice.

Toxoplasma gondii is a ubiquitous highly prevalent zoonotic protozoan. Cats are the definitive hosts, while all other warm-blooded animals are intermediate hosts for this parasite. Commensal rodents, being the main prey of cats, are probably the major reservoir for T. gondii in the domestic environment. Rodents can acquire infection after ingestion of oocysts that have sporulated in the environment. However, experimental evidence shows that vertical transmission can be sufficient for the perpetuation of transmission between generations of mice. In natural settings, the relative epidemiological

Genetics and meta-analysis of recessive non-syndromic hearing impairment and Usher syndrome in Maghreb population: lessons from the past, contemporary actualities and future challenges.

Hereditary hearing impairment (HI) is a heterogeneous condition with over 130 genes associated with genetic non-syndromic HI (NSHI) and Usher syndrome (USH). Approximately 80% of hereditary NSHI cases have autosomal recessive (AR) mode of inheritance. The high rate of consanguinity and endogamy in the Maghreb countries, including Tunisia, Algeria and Morocco, represents a major contributing factor to the development of ARHI. Since the 90s, those populations, with their particular large familiar structure, represented an effective key towards the discovery of the first HI loci and genes. In thi

Plague circulation and population genetics of the reservoir Rattus rattus: the influence of topographic relief on the distribution of the disease within the Madagascan focus.

BACKGROUND: Landscape may affect the distribution of infectious diseases by influencing the population density and dispersal of hosts and vectors. Plague (Yersinia pestis infection) is a highly virulent, re-emerging disease, the ecology of which has been scarcely studied in Africa. Human seroprevalence data for the major plague focus of Madagascar suggest that plague spreads heterogeneously across the landscape as a function of the relief. Plague is primarily a disease of rodents. We therefore investigated the relationship between disease distribution and the population genetic structure of th

Pleiotropy and eQTL analysis identify shared genetic architecture of lipid traits in African-ancestry populations.

Dyslipidemia is a leading modifiable risk factor for cardiovascular disease (CVD), yet the shared genetic architecture of lipid traits remains poorly characterized, particularly in African-ancestry populations who bear a disproportionate CVD burden but are underrepresented in genomic research. We conducted multi-trait meta-analyses of genome-wide association summary statistics for HDL cholesterol, LDL cholesterol, triglycerides, and total cholesterol in 34 039 individuals of African ancestry, comprising 27 289 discovery participants from three continental African cohorts (ACCME, AWI-Gen, and U

Genetic Diversity and Haplotype Architecture of Non-Classical HLA Class I Genes in the Japanese Population.

The non-classical Class I genes (HLA-E, HLA-F and HLA-G) show limited protein-level diversity; however, full-length analyses that include regulatory and noncoding regions have not been sufficiently performed in East Asian populations. We designed new primers for long-range PCR and conducted PacBio SMRT sequencing to generate full-length gene sequences of HLA-E, HLA-F and HLA-G, including 5' and 3' untranslated regions (UTRs) as well as coding and intronic regions in 531 healthy Japanese individuals. These data enabled the full phasing of HLA-E, HLA-F and HLA-G, including upstream and downstrea

Pan-ethnic preconception screening: evidence from a large-scale programme in a genetically diverse population.

BACKGROUND: Preconception genetic screening is a key preventive strategy for identifying at-risk couples (ARCs) of inherited disorders and supporting informed reproductive decision-making. However, increasing population admixture and the widespread adoption of next-generation sequencing challenge the validity of ancestry-driven risk stratification. METHODS: A total of 107 739 individuals participated in the national screening programme, including 51 722 couples and 4295 individuals tested independently, representing 33 ethnic groups. Under the sequential testing strategy, all women underwent

The MexMAGIC population reveals the genetic architecture of traits exhibiting clinal variation in Mexican native maize.

Defining the genetic basis of local adaptation is a key goal of evolutionary biology and crop improvement. Theory predicts that when selective pressures follow differences in the environment, a cline will be established. Clines can be exploited to uncover adaptive variation by association of alleles with the environment. However, monotonic phenotypic change over a cline is not necessarily mirrored in the behavior of genetic variants and population structure can further complicate analysis. To study genetic and phenotypic variation across the environment, we developed a multi-parent advanced ge

Targeted population genomics uncovers demographic history and genetic divergence in north American wild cranberry.

Wild populations of North American cranberry (Vaccinium macrocarpon Aiton) are reservoirs of genetic variation that may contribute to the improvement of breeding-relevant traits. However, the extent to which wild genetic variation is geographically structured and represented in elite germplasm remains unclear. We analysed 179 wild cranberry accessions from the upper Midwest and Eastern North America to estimate nucleotide diversity (π), population structure, and loci associated with genetic differentiation and environmental variables using a genome-informed targeted genotyping panel. Additiona

Contrasting Patterns of Genetic Isolation Among Diadromous, Landlocked, and Non-Migratory Gobiid Fish Populations.

Dams can landlock populations of amphidromous fishes that formerly migrated between rivers and seas. However, the genetic consequences of these life-cycle changes remain unclear. Rhinogobius is a speciose genus of freshwater gobies widely distributed in Asia, including non-migratory R. duospilus and facultatively amphidromous R. similis. Using genome-wide SNPs from Hong Kong, southern China, we compared the genetic diversity and structure of R. duospilus (six populations), non-landlocked R. similis (three populations), and landlocked R. similis (four populations) that had been isolated for 58-
