Antibiotic susceptibility of bacteria infecting total joint arthroplasty sites.

BACKGROUND: Currently, there is no consensus regarding the principles of empiric antibiotic treatment of suspected periprosthetic infection … ombination of cefazolin and gentamicin therapy. All chronic and acute postoperative infections with Gram-positive bacteria and all cases in which a Gram stain fails to identify bacteria should be managed with vancomycin. Infections with Gram-negative bacteria should be managed with a third or fourth-generation cephalosporin. Infections with mixed Gram-positive and Gram- …

Preparation of Membrane Models of Gram-Negative Bacteria and Their Interaction with Antimicrobial Peptides Studied by CD and NMR.

The antibiotic activity of antimicrobial peptides is generally derived via some type of disruption of the cell membrane(s). The most common models used to mimic the properties of bacterial membranes consist of mixtures of various zwitterionic and anionic phospholipids. This approach works reasonably well for Gram-positive bacteria. However, since the membranes of Gram-negative bacteria contain lipopolysaccharides, as well as zwitterionic and anionic phospholipids, a more complex model is required to simulate the outer membrane of Gram-negative bacteria. Herein we present a protocol for the pre…

Identification of bacteria in blood culture broths using matrix-assisted laser desorption-ionization Sepsityper™ and time of flight mass spectrometry.

Matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS) is a novel method for the direct identification of bacteria from blood culture broths. We evaluate for the first time, the performance of the MALDI Sepsityper™ Kit and MS for the identification of bacteria compared to standard phenotypic methods using the manufacturer's specified bacterial identification criteria (spectral scores ≥1.700-1.999 and ≥2.000 indicated identification to genus and species level, respectively). Five hundred and seven positive blood culture broths were prospectively examined, of

Multidrug-resistant organisms in lung transplant: a narrative review.

PURPOSE OF REVIEW: The purpose of this narrative review is presenting the current knowledge of multidrug-resistant (MDR) pathogens in lung transplant recipients, considering both Gram-positive and Gram-negative bacteria. RECENT FINDINGS: Overall prevalence of Gram-negative pathogens has increased remarkably (4.33/1000 recipient-days) in solid organ transplant recipients, while the prevalence of Gram-positive bacteria seems to be decreasing (0.20 cases/100 transplant-years). In lung transplant, the prevalence of postoperative infections due to MDR-GN bacteria has been assessed between 31 and 5…

A New Simplified Coralmycin Derivative with Potent Anti-Gram-Negative Activity and Its Congeners Produced by the Myxobacterium Corallococcus coralloides M23.

Multidrug-resistant (MDR) Gram-negative bacterial infections urgently require new antibacterial agents with novel chemical scaffolds. In our continued investigation of minor metabolites produced by the myxobacterium Corallococcus coralloides M23, we isolated three new coralmycin derivatives, coralmycins J (1), K (2), and L (3), from large-scale fermentation cultures. Spectroscopic analyses revealed that these compounds represent structurally simplified derivatives of coralmycin with varying degrees of truncation. Compounds 1-4 (coralmycins J, K, L, and A) are new natural products isolated in o

Bactericidal Metal-Organic Gallium Frameworks - Synthesis to Application.

Gallium, a trace metal not found in its elemental form in nature, has garnered significant interest as a biocide, given its ability to interfere with iron metabolism in bacteria. Consequently, several gallium compounds have been developed and studied for their antimicrobial properties but face challenges of poor solubility and formulation for delivery. Organizing the metal into three-dimensional, hybrid scaffolds, termed metal-organic frameworks (MOFs), is an emerging platform with potential to address many of these limitations. Gallium MOFs show improved solubility and antibacterial potency r
