Expression of immunoreactive activin A protein in remodeling lesions associated with interstitial pulmonary fibrosis.

The expression of activin A, one of the transforming growth factor-beta supergene family, was studied in various pulmonary conditions associated with interstitial pulmonary fibrosis (3 cases with diffuse alveolar damage, 6 cases with idiopathic pulmonary fibrosis, and 1 case with pulmonary fibrosis associated with rheumatoid arthritis) using immunohistochemical techniques on paraffin-embedded sections. Controls consisted of 10 cases with normal pulmonary parenchyma, and 2 cases with primary pulmonary hypertension and 1 case with secondary pulmonary hypertension were also studied. The lung spec

Regulation of organogenesis. Common molecular mechanisms regulating the development of teeth and other organs.

Vertebrate organs develop from epithelial and mesenchymal tissues, and during their early development they share common morphological features. These include condensation of the mesenchymal cells and thickening, folding or branching of epithelial sheets. Sequential and reciprocal interactions between the epithelial and mesenchymal tissues play central roles in regulation of the morphogenesis of all organs. During recent years increasing amounts of molecular data have accumulated from studies describing developmental changes in expression patterns of molecules, as well as from functional in vit

Cellular signals in atherosclerosis.

Atherosclerosis is a complex, incompletely understood process involving several cell types, including smooth muscle cells, endothelial cells, and platelets, and the signals between them. Atherosclerotic lesions result from an excessive inflammatory fibroproliferative response to various forms of insult to the endothelium and smooth muscle of the artery wall. Numerous growth regulatory molecules/factors participate in this process. This article reviews the atherosclerotic process and provides new scientific insights into the prevention and treatment of this disease. The implications for nursing

The hamster cheek pouch carcinogenesis model.

The Syrian golden hamster cheek pouch carcinogenesis model is probably the best-known animal system that closely compares to events involved in the development of premalignant and malignant human oral cancers. Furthermore, it is one of the most well-characterized models for squamous cell carcinomas (SCCs). However, stages of carcinogenesis (initiation, promotion, and progression) have not been well-defined in this system. Basic understanding of the mechanism(s) of carcinogenesis in this organ is instrumental for the development of new strategies for chemoprevention and early chemointervention.

Estrogen use and cancer risk: a review.

The role of estrogens as carcinogens, cocarcinogens or tumor promoters, as well as their mechanism(s) of action on cancer cells, are thoroughly reviewed. Although there is ample evidence that estrogens (natural and synthetic) can induce multiple benign and malignant tumors in animals, and most of these tumors are histologically similar to that in humans, there is no direct evidence that natural estrogens (estradiol-17 beta, estrone) are carcinogenic in humans. Recent evidence in cellular and molecular oncology revealed that estrogens act by genetic and epigenetic mechanisms on cancer cells, an

[Effects of growth factors on proliferation of cultured human peritoneal mesothelial cells].

Loss of ultrafiltration during continuous ambulatory peritoneal dialysis (CAPD) is often caused by the structural peritoneal membrane alteration, namely the disappearance of mesothelial cells and the proliferation of peritoneal collagen fibers. The interleukin hypothesis has been proposed to explain the etiology of peritoneal fibrosis. The CAPD procedure has been shown to induce macrophages and lymphocytes in the peritoneum, resulting in the production of interleukin-1 (IL-1) and interferon-gamma (IFN-gamma), which may be promote to the development of peritoneal fibrosis. On the other hand, th

Modulation of peritoneal re-epithelialization by postsurgical macrophages.

The studies reviewed here show that postsurgical macrophages are capable of modulating the proliferation of TRC. That is, macrophages either suppress or enhance the proliferation of TRC depending on the culture time and the medium used as a comparison, i.e., culture medium with only serum or spent medium from cultures of resident peritoneal macrophages. Postsurgical macrophages also modulate the morphology of (spindly or rounded appearance) and the secretion of extracellular matrices by TRC. The responsivity of TRC to control by postsurgical macrophage-spent media or growth factors changes as

[Determination of renotropic activity in the sera of 35 uninephrectomy patients].

The renotropic growth factor (RGF) does increase in postuninephrectomy serum although the majority of studies were conducted in experiment. As regard to whether there is postnephrectomy RGF in human being, we have not hitherto reached a conclusion. By incorporating tritiated thymidine (3H-TdR) into primary human kidney cells in culture. We have determined renotropic activity in the sera of 35 uninephrectomy patients. The RGF was found increased in 68% and 76% patients at postnephrectomy days 2 and 5 respectively, showing an average increase of 14% and 33% as compared with that of preoperation

Sex steroids and growth factors in mammary cancer.

We have studied the effect of mitogens on the proliferation of the human breast cancer cell line MCF7 under growth factor-defined culture conditions. Under these conditions MCF7 cells showed the strict hormone dependence that has been established in vivo (in nude mice). E2, insulin or insulin-like growth factors were found to be essential mitogens for these cells. E2 synergistically stimulated proliferation in combination with suboptimal doses of these growth factors. This latter combination of mitogens could be important for in vivo growth regulation of hormone-dependent breast tumours. Synth

Co-cultures: their relevance to assisted reproduction.

Assisted reproductive techniques have contributed significantly to alleviating subfertility in the childless couple. However, the take-home baby rates have been very low throughout the world. One of the contributory causes has been the reduced viability of replaced embryos brought about by suboptimal in vitro conditions. The culture of human embryos in the presence of passaged human tubal ampullary monolayers (co-cultures) is an attractive approach to improving the viability of embryos for assisted reproduction. Seventy per cent of blastocysts can be produced in human ampullary co-culture as c

The effects of interleukin-1 on articular cartilage destruction as observed in arthritic diseases, and its therapeutic control.

At present there is substantial evidence to suggest that interleukin 1 (IL-1) may act as a key mediator in the normal physiologic regulation of cartilage as well as in the pathogenesis of cartilage destruction in arthritic disorders. IL-1 induces stimulation of chondrocyte catabolism and alters chondrocyte biosynthesis in articular cartilage. These actions of IL-1 may lead to destruction and inappropriate repair following degradation of the cartilage matrix. Moreover, IL-1 induced biological activities in chondrocytes may be influenced by growth factors (e.g. fibroblast growth factor, insulin-

Effect of different oestrogen doses on final height reduction in girls with constitutional tall stature.

The effects of different doses of oestrogens in constitutionally tall girls were evaluated in two centres for paediatric endocrinology. In one centre, 38 girls were treated with a high oestrogen dose of 0.3 to 0.5 mg ethinyloestradiol (EE) daily. In the other, 44 girls received a comparably low dose of 0.1 mg EE per day. Height prediction (HP), chronological age (CA), and height at the onset of treatment were comparable in both groups. Although the duration of treatment was significantly longer in those receiving the low dose, the cumulative oestrogen dose was still significantly lower. The do

The Northern Ireland experience of growth hormone therapy for short stature.

In 1967 the first patient in Northern Ireland commenced growth hormone treatment for short stature. By the end of December 1988 a total of 89 patients had been treated. Thirty-two had idiopathic isolated growth hormone deficiency, an incidence of 1.5 new cases per year (in a population of 1.5 million with approximately 30,000 births per year). Since 1967 the mean age at starting treatment has fallen from 18 years to 10 years and the height standard deviation score has fallen from -4.7 +/- 0.6 to -3.4 +/- 0.3. The group with classical growth hormone deficiency (maximum GH less than 7 mU/l durin

Bovine and human pineal glands contain substantial quantities of endothelial cell stimulating angiogenic factor.

Using a quantitative assay the amount of a low Mr endothelial cell stimulating angiogenesis factor (ESAF) has been determined in human pineal glands and bovine brain, retina, pineal gland, liver and kidney. Pineal glands contain approximately ten times as much ESAF as the retina or grey and white matter from the cerebral cortex and a hundred times as much as highly vascular tissues such as liver and kidney. The relevance of these findings to the highly vascular nature of the pineal gland is discussed.

Periarticular fibrosis associated with idiopathic retroperitoneal fibrosis.

We describe a previously unreported association of retroperitoneal fibrosis with biopsy proven periarticular inflammatory fibrosis. Histologic features were strikingly similar at the 2 sites. Plasma levels of a platelet derived growth factor, connective tissue activating peptide III, were found to be elevated and may play a role in pathogenesis. A dramatic response of the periarticular process to corticosteroids correlated with improvement of other variables of disease activity.

Variant sublines of the human B-lymphoma cells Namalwa are at different stages of differentiation.

Variant sublines of the Burkitt's lymphoma cell line Namalwa, previously shown to differ in MHC class II antigen and immunoglobulin expression, have been tested with monoclonal antibodies defining B-cell specific clusters of differentiation (CD). The Namalwa sublines express antigens recognized by the CD antibodies in patterns that indicate that the cells are arrested at different stages of differentiation.

Anti-proliferative effects of interferons on Daudi Burkitt lymphoma cells: induction of cell differentiation and loss of response to autocrine growth factors.

Treatment of Daudi B-lymphoblastoid cells with low concentrations of either natural or recombinant human alpha-interferons inhibits cell proliferation and modulates the expression of a number of cell-surface antigens. Using a panel of monoclonal antibodies (MAbs) identifying determinants expressed at the surface of normal plasma cells, and polyclonal antibodies against surface and cytoplasmic immunoglobulin, we have found that growth inhibition is accompanied by plasmacytoid differentiation. Assays of growth stimulation of heterologous cells indicate that the culture medium from interferon-tre

Vasoactive peptides in the pathogenesis of psoriasis.

Psoriasis, a chronic inflammatory skin disease, is believed to be exacerbated by stress. The exact mechanism of this phenomenon is not fully understood, however, it has been postulated that different substances released from dermal nerve endings during stress may take part in initiation or modulation of psoriasis. One of the most interesting group of mediators are polypeptides, also named as neuropeptides, that possess vasoactive properties. It was documented that these polypeptides could not only be released from nerve endings, but may also be directly synthesised in the skin and liberated fr
