The Drosophila homolog of APP promotes Dscam expression to drive axon terminal growth, revealing interaction between Down syndrome genes.

Down syndrome (DS) is caused by triplication of human chromosome 21 (HSA21). Although several HSA21 genes have been found to be responsible for aspects of DS, whether and how HSA21 genes interact with each other is poorly understood. DS patients and animal models present with a number of neurological changes, including aberrant connectivity and neuronal morphology. Previous studies have indicated that amyloid precursor protein (APP) and Down syndrome cell adhesion molecule (DSCAM) regulate neuronal morphology and contribute to neuronal aberrations in DS. Here, we report the functional interact

Nicotine Addiction and Psychiatric Disorders.

Even though smoking rates have long been on the decline, nicotine addiction still affects 20% of the US population today. Moreover, nicotine dependence shows high comorbidity with many mental illnesses including, but are not limited to, attention deficit hyperactivity disorder, anxiety disorders, and depression. The reason for the high rates of smoking in patients with mental illnesses may relate to attempts to self-medicate with nicotine. While nicotine may alleviate the symptoms of mental disorders, nicotine abstinence has been shown to worsen the symptoms of these disorders. In this chapter

Nutrition, insulin-like growth factor-1 and retinopathy of prematurity.

Retinopathy of prematurity is a potentially blinding disease starting with impaired retinal vessel growth in the neonatal period. Weeks to months later, peripheral retinal hypoxia induces pathologic neovascularization that may lead to retinal detachment and blindness. Current treatment strategies target late stage disease and it would be advantageous if retinopathy of prematurity could be prevented. Poor general growth after very preterm birth is a universal problem associated with increased risk of retinopathy. Loss of the maternal-fetal interaction results not only in loss of nutrients but a

Neisseria meningitidis endogenous endophthalmitis with meningitis in an immunocompetent child.

Neisseria meningitidis is a major cause of childhood morbidity and mortality worldwide. We describe an exceptional case of an immunocompetent 15-month-old child presenting with a unilateral anterior uveitis, hypopyon, and sepsis. Anterior chamber aspirate demonstrated gram-negative cocci before Neisseria meningitidis was identified in blood and cerebrospinal fluid. Meningococcal endophthalmitis presents variably with sepsis, meningitis, or isolated ocular symptoms. Diagnosis is a clinical challenge, requiring diagnostic sampling and treatment from both pediatricians and ophthalmologists. Delay

Upstream effect of esophageal distention: effect on airway.

The pharyngoesophageal segment of the foregut has an important function in steering clear of luminal contents from the airway, across the age spectrum from a premature neonate to an aging adult. This complex neuromuscular interaction between the esophagus and the airway is maintained by a variety of mechanisms mediated by the parasympathetic and sympathetic afferent and efferent outflows involving the myenteric plexus, glossopharyngeal and vagus cranial nerves, phrenic nerve, and brainstem nuclei. The esophageal provocation during gastroesophageal reflux events results in esophageal distention

The testosterone hypothesis: Assessment since Geschwind and Behan, 1982.

The Geschwind hypothesis proposes a causal interaction among non-right-handedness, immune disorders, and learning disabilities, including dyslexia, via the intrauterine action of the male hormone testosterone. Some epidemiologic studies have supported at least a statistical association among the three traits; others have not. The associations between learning disorders and immune disease and between learning disorders and non-righthandedness appear to be better supported than that between immune disorders and non-right-handedness. However, none of data thus far accumulated are conclusive becau

Brain tissue transplantation in neonatal rats prevents a lesion-induced syndrome of adipsia, aphagia and akinesia.

Previous experiments have proven brain tissue transplantation effective in reversing lesion-induced behavioral deficits in mature rats. This study reversed the usual experimental paradigm, so that fetal substantia nigra was transplanted into intact neonatal rats and allowed to mature in the host brain. Upon maturation substantia nigra lesions were made bilaterally to reveal the functional contribution of the transplanted tissue. In control animals these lesions depleted striatal dopamine, producing rigidity, poverty of movement and abnormal posture comparable to Parkinson's disease in the huma

Repeated Tick Infestations Impair Borrelia burgdorferi Transmission in a Non-Human Primate Model of Tick Feeding.

The blacklegged tick, Ixodes scapularis, is the predominant vector of Borrelia burgdorferi, the agent of Lyme disease in the USA. Natural hosts of I. scapularis such as Peromyscus leucopus are repeatedly infested by these ticks without acquiring tick resistance. However, upon repeated tick infestations, non-natural hosts such as guinea pigs, mount a robust immune response against critical tick salivary antigens and acquire tick resistance able to thwart tick feeding and Borrelia burgdorferi transmission. The salivary targets of acquired tick resistance could serve as vaccine targets to prevent

Modulation of innate immune responses by virus-host interactions in Orthoflavivirus.

Orthoflavivirus are single-stranded positive-sense RNA viruses that pose a significant threat to human and animal health globally, accounting for more than 400 million infections annually. They are predominantly transmitted by arthropods and cause a plethora of clinical manifestations, ranging from mild self-limiting infections to lethal encephalitis, with the potential to cause epidemics and pandemics. The complication in effectively treating Orthoflavivirus infections stems from the intricate interactions between virus and host proteins. A prominent hallmark of Orthoflavivirus infection is t

Drug interactions with paracetamol.

Paracetamol (acetaminophen) is one of the most commonly used analgesic antipyretic drugs worldwide, and it is widely available by prescription and over the counter (OTC). Fortunately, few clinically significant drug interactions have been documented. There is probable potentiation of hepatotoxicity following an overdose from the paracetamol metabolite NAPQI by enzyme-inducing drugs. There is considerable controversy regarding the possible interaction with warfarin in its potential to increase its anticoagulant effects because of discrepancies between observational studies and those in healthy

The influence of ovarian activity and menopause on mental health: Evidence from animal models and women.

Hormonal variations occurring throughout the female reproductive cycle have a significant impact on physical and mental health, particularly due to the influence of estradiol (E2) and progesterone (P4). These changes are directly related to alterations in neurological systems, being associated with conditions such as premenstrual syndrome (PMS), premenstrual dysphoric disorder (PMDD), and mood disorders during hormonal transition phases, such as perimenopause and menopause. Studies conducted in humans and animal models indicate that these fluctuations affect neurotransmitters, neural plasticit

Long-Tailed Macaques (Macaca fascicularis) in Urban Landscapes: Gastrointestinal Parasitism and Barriers for Healthy Coexistence in Northeast Thailand.

Gastrointestinal parasites have diverse life cycles that can involve people, animals, and the environment (e.g., water and soil), demonstrating the utility of One Health frameworks in characterizing infection risk. Kosumpee Forest Park (Thailand) is home to a dense population of long-tailed macaques (Macaca fascicularis) that frequently interact with tourists and local residents. Our study investigated the presence of zoonotic parasites, and barriers to healthy coexistence by conducting stool analysis on macaques (N = 102) and people (N = 115), and by examining risk factors for infection with

Molecular Mechanisms of Bartonella and Mammalian Erythrocyte Interactions: A Review.

Bartonellosis is an infectious disease caused by Bartonella species that are distributed worldwide with animal and public health impact varying according to Bartonella species, infection phase, immunological characteristics, and geographical region. Bartonella is widely present in various mammals including cats, rodents, ruminants, and humans. At least 13 Bartonella species or subspecies are zoonotic. Each species has few reservoir animals in which it is often asymptomatic. Bartonella infection may lead to various clinical symptoms in humans. As described in the B.tribocorum-rat model, when Ba

Comparison of Q fever serology methods in cattle, goats, and sheep.

Coxiella burnetii is an obligate intracellular bacterium that is responsible for the zoonotic disease Q fever. The distribution of this agent is worldwide except for New Zealand, and infection can be asymptomatic in both human beings and animals. Chronic exposures can produce abortions, stillbirths, and infertility issues in animals and endocarditis in human beings. A commercial enzyme-linked immunosorbent assay (ELISA) kit marketed in the European Union was purchased to compare C. burnetii antibody detection methods. The current study examined the agreement of ELISA and complement fixation re

Prospects of vaccination against schistosomiasis.

The development of a vaccine against schistosomiasis is necessary in order to reduce the risk of reinfection after drug treatment. Recent reports converge on a message strongly supporting the existence of naturally acquired human immunity to this infection and reinforcing the hypothesis that at least partial protection can be achieved by artificial means. Advances in molecular biology have led to the identification and characterization of an array of protective schistosome antigens, and the introduction of new sophisticated methods for their production enables a bypass of previous low-yielding

Evaluation of the Interactions between Mumps Virus and Guinea Pig.

Mumps is a highly contagious viral disease that can be prevented by vaccination. In the last decade, we have encountered repeated outbreaks of mumps in highly vaccinated populations, which call into question the effectiveness of available vaccines. Animal models are crucial for understanding virus-host interactions, and viruses such as mumps virus (MuV), whose only natural host is the human, pose a particular challenge. In our study, we examined the interaction between MuV and the guinea pig. Our results present the first evidence that guinea pigs of the Hartley strain can be infected in vivo

Divergence between hepatic insulin-like growth factor (IGF)-I mRNA expression and IGF-I serum levels in Leishmania (Leishmania) infantum chagasi-infected dogs.

Visceral leishmaniasis (VL) caused in the New World by Leishmania (Leishmania) infantum chagasi has dog as important peridomestic reservoir in its transmission cycle. Since VL in infected animals is similar to human VL, its study is interesting for human pathology. During Leishmania infection, insulin-like growth factor-I (IGF-I) plays a role in the host-parasite interaction, favoring parasite growth, particularly acting directly on Leishmania. We evaluated IGF-I mRNA expression in different organs/tissues, which was differently modulated in dogs naturally infected by L. (L.) infantum chagasi.
