Insecticide resistance reduces the effectiveness of current malaria-vector-control interventions. To understand the mechanisms underlying permethrin resistance in field-derived Anopheles gambiae vectors collected from Cameroon, we used ultra-high-performance liquid chromatography coupled with high-resolution tandem mass spectrometry (UHPLC-HRMS/MS) to comprehensively analyze metabolic changes in resistant and susceptible samples to gain insights into mechanisms driving permethrin resistance. Resistant mosquitoes exhibited upregulated levels of inosine, nicotinic acid, dipeptides, amino acids, …

Detoxification (detox) plays a major role in pesticide action and resistance. The mechanisms involved are sometimes part of the discovery and development process in seeking new biochemical targets and metabolic pathways. Genetically modified and chemical-safener-modified crops are a marked exception and often involve herbicide detox by design to achieve the required crop tolerance. This perspective evaluates the role of detox by design or chance and target-site-based selectivity in insecticide, herbicide, and fungicide action and human health and environmental effects.

A wind-sensitive vane apparatus was designed and implemented specifically to accommodate the attachment of otherwise unidirectional insecticide treatment cages used in ground ultra-low volume mosquito adulticide field tests. This cage support system is useful in keeping the potential West Nile virus and St. Louis encephalitis vector Culex quinquefasciatus caged mosquitoes oriented into the wind during field efficacy tests. Testing capacity for resistance surveillance was tripled during the 2005 season, and more reliable results were achieved as a consequence.

The common body infestations, their incidence, diagnosis and treatment are discussed. Pediculosis capitis still has a high incidence in Britain. The louse could probably be effectively eradicated provided a national campaign of treatment and education were undertaken. Intensive therapy with a malathion containing preparation should be used before the louse develops resistance to this new organophosphorus insecticide. Similarly, scabies can be effectively treated. All doctors should be aware of the signs and symptoms of this common dermatosis and its treatment.

India's National Malaria Elimination Program, which achieved notable success in reducing cases, is now facing challenges because of the rise in malaria cases in 2023 and 2024. This increase is linked to multiple factors, including disruptions in healthcare priorities during the coronavirus disease 2019 pandemic, drug and insecticide resistance, and the under-detection of asymptomatic carriers and pfhrp2-deleted parasites. The ultimate goal is to achieve sustainable reductions in malaria incidence and transmission, paving the way for a malaria-free India by 2030.

Following unsuccessful eradication attempts there was a resurgence of malaria towards the end of the 20th century. Renewed control efforts using a range of improved tools, such as long-lasting insecticide-treated bednets and artemisinin-based combination therapies, have more than halved the global burden of disease, but it remains high with 445 000 deaths and more than 200 million cases in 2016. Pitfalls in individual patient management are delayed diagnosis and overzealous fluid resuscitation in severe malaria. Even in the absence of drug resistance, parasite recurrence can occur, owing to hi

BACKGROUND: Chagas disease control relies primarily on vector control using pyrethroid insecticide sprays with residual action in domestic and peridomestic environments. However, the necessity for repeated applications and the development of pyrethroid resistance in some countries have undermined this strategy. Alternative control tools are needed, and host-targeted systemic insecticides offer an alternative approach by creating toxic blood meals for hematophagous vectors. In peridomestic settings, chickens are major blood meal sources for triatomines. This study evaluated the insecticidal act

BACKGROUND: Owing to pyrethroid resistance in An. gambiae, the carbamate and organophosphate insecticides are currently regarded as alternat … rb resistance in An. gambiae populations is a clear indication that calls for the implementation of insecticide resistance management strategies. The ace-1(R) mutation could not entirely explain the resistance to bendiocarb observed and is highly suggestive of involvement of other resistance mechanisms such as metabolic detoxification.

Susceptibility tests were conducted on the adults of two species of Japanese encephalitis vectors viz., Culex pseudovishnui and C. tritaeniorhynchus against diagnostic doses of DDT, dieldrin, malathion, fenitrothion, propoxur and permethrin at different exposure duration. C. pseudovishnui was found susceptible to permethrin and resistant to dieldrin and propoxur while C. tritaeniorhynchus was found susceptible to permethrin and resistant to DDT, dieldrin, fenitrothion and propoxur. A verification, however, was required with other insecticides for both the species.

BACKGROUND: Bemisia tabaci is a globally invasive agricultural pest. The B. tabaci Q/MED has rapidly replaced B/MEAM1, largely due to Q/MED's high resistance to neonicotinoid insecticides. Although cytochrome P450s (P450s) are major detoxification enzymes involved in insecticide resistance, the genome-wide identification and functional contributions to differential clothianidin resistance between B/MEAM1 and Q/MED remain poorly understood.
RESULTS: A total of 129 P450 genes were identified in both B/MEAM1 and Q/MED genomes, with marked expansions observed in clan 3 and clan 4. Comparative chr…

The extensive use of Bacillus thuringiensis (Bt) insecticidal proteins in pest control has led to the evolution of resistance in target insects. Reduced toxin binding to the midgut epithelial membrane is considered a major resistance mechanism. Current methods for characterizing toxin binding suffer from limitations in physiological relevance or practical applicability. Here, we developed an in situ binding (ISB) method to visualize and quantify the binding of Cry1Ac toxin to the midgut epithelium of Helicoverpa armigera larvae. By culturing dissected midguts in a cell culture medium containin

BACKGROUND: Helicoverpa zea and Spodoptera frugiperda are destructive insect pests of cotton and corn. Both have evolved practical resistance to transgenic crops producing Bacillus thuringiensis (Bt) toxins in the United States. GS-omega/kappa-Hxtx-Hv1a (ω/κ-Hv1a) is a novel biopesticide derived from spider venom with activity against lepidopteran pests. The efficacy of ω/κ-Hv1a is dependent on combining it with a facilitator product such as a Bt-based insecticide. However, it is unknown if ω/κ-Hv1a can be an alternative for the management of Bt resistance. In this study, we tested the effect

Tuta absoluta is a major invasive pest of Solanaceous crops, largely managed through chemical interventions. Despite its economic importance, the genomic landscape of its detoxification machinery remains poorly understood. We performed a genome-wide identification of 23 GST genes (TabsGSTs) in T. absoluta, characterized by amino acid lengths of 147-289 aa and a predominantly 1- or 5-exon genomic architecture (∼65%). The identification of four tandemly arranged gene clusters highlights potential evolutionary hotspots for insecticide resistance. Phylogenetic analysis categorized these genes into…

Cochliomyia hominivorax (Calliphoridae) is one of the most important myiasis-causing flies and is responsible for severe economic losses to the livestock industry throughout the Neotropical region. In Brazil, C. hominivorax has been controlled mainly with organophosphate (OP) insecticides, although the inappropriate use of these chemicals can result in the selection of resistant flies. Changes in carboxylesterase activity have been associated with OP insecticides in some arthopodan species. In this work, we isolated and characterized part of the E3 gene in C. hominivorax (ChalphaE7), which con

Tigolaner is a recently developed long-acting ectoparasiticide with potent activity against fleas and ticks, but its molecular mode of action remains poorly defined. In this study, we investigated how tigolaner interacts with insect γ-aminobutyric acid (GABA)-gated chloride channels by combining receptor pharmacology, insecticide bioassays using Drosophila point mutants, and molecular docking. The pore-associated A301S substitution markedly reduced the inhibitory activity of the classical non-competitive antagonist α-endosulfan but caused only an approximately two-fold decrease in tigolaner po

Continuous use of organophosphate for dengue vector management has increased the risk of resistance development in Aedes aegypti, threatening the effectiveness of current control programmes. This study evaluated susceptibility and mechanisms associated with organophosphate resistance in Ae. aegypti populations from five dengue-endemic districts of sub-Himalayan West Bengal, India. Larvae and adults were subjected to insecticide bioassays, synergist assays, biochemical assays, gene expression analysis, partial sequencing of the acetylcholinesterase gene (ace-1), and molecular docking. Spodoptera exigua is a significant invasive pest that has developed resistance to various commonly applied insecticides. The detoxification of insecticides in insects may partially depend on ATP-binding cassette (ABC) transporters. Although the functions of ABCB subfamily genes are diverse in insects, the specific roles of ABCBs in S. exigua remain largely unexplored. This research focused on identifying ten SeABCB genes and characterizing their expression profiles across various developmental stages and tissues, as well as in response to different insecticide treatments. Our findings indicate

Mosquitoes are important vectors of human diseases, with Anopheles sinensis posing substantial health risks across Asia. Although pyrethroids remain widely deployed, their effectiveness is compromised by resistance associated with sodium channel mutations. Spider peptide neurotoxins are potential biopesticides. Here, we evaluated the insecticidal efficacy of the Pardosa pseudoannulata neurotoxin PPTX-04, both alone and in combination with pyrethroids, against A. sinensis. Bioassays were conducted on laboratory and field populations, and functional analyses were performed on wild-type and mutan
