In a multi-institutional collaborative project, 1473 patients with myeloproliferative neoplasms (MPN) were screened for isocitrate dehydrogenase 1 (IDH1)/IDH2 mutations: 594 essential thrombocythemia (ET), 421 polycythemia vera (PV), 312 primary myelofibrosis (PMF), 95 post-PV/ET MF and 51 blast-phase MPN. A total of 38 IDH mutations (18 IDH1-R132, 19 IDH2-R140 and 1 IDH2-R172) were detected: 5 (0.8%) ET, 8 (1.9%) PV, 13 (4.2%) PMF, 1 (1%) post-PV/ET MF and 11 (21.6%) blast-phase MPN (P<0.01). Mutant IDH was documented in the presence or absence of JAK2, MPL and TET2 mutations, with similar mu

Human conducting airways contain two anatomically distinct epithelial cell compartments: surface epithelium and submucosal glands (SMG). Surface epithelial cells interface directly with the environment and function in pathogen detection, fluid and electrolyte transport, and mucus elevation. SMG secrete antimicrobial molecules and most of the airway surface fluid. Despite the unique functional roles of surface epithelia and SMG, little is known about the differences in gene expression and cellular metabolism that orchestrate the specialized functions of these epithelial compartments. Reactive oxygen species (ROS)-mediated cell injury contributes to the pathophysiology of cardiovascular disease and myocardial dysfunction. Protection against ROS requires maintenance of endogenous thiol pools, most importantly, reduced glutathione (GSH), by NADPH. In cardiomyocytes, GSH resides in two separate cellular compartments: the mitochondria and cytosol. Although mitochondrial GSH is maintained largely by transhydrogenase and isocitrate dehydrogenase, the mechanisms responsible for sustaining cytosolic GSH remain unclear. Glucose-6-phosphate dehydrogenase (G6PD) functions as the first

The effect of the three-monthly injectable contraceptive depot medroxyprogesterone acetate (DMPA) on liver and lipid function was assessed in Thai women with liver fluke (Opisthorchis viverrini) infestation, DMPA administration being started in the immediate postpartum period. Immediate postpartum IUD and sterilization acceptors with fluke infestation were recruited as a comparison (control) group for the fluke-positiv DMPA acceptors. Comparable groups of fluke-negative acceptors were recruited in an area of Thailand free of liver fluke transmission. Results are presented for the first 6 follo

White spot syndrome virus (WSSV) drives extensive metabolic reprogramming to support viral replication, including activation of reductive carboxylation, a pathway in which host isocitrate dehydrogenase 1 (IDH1) has a central role. However, the mechanisms by which viral factors regulate IDH1 and associated metabolic networks remain unclear. Here, we identified WSSV355 as a key viral regulator that contributed to IDH1 binding and activation during infection. Using yeast two-hybrid screening, WSSV355 was identified as an LvIDH1-interacting protein and further validated by co-immunoprecipitation a

BACKGROUND: Brain reserve refers to the structural capacity of the brain and correlates with age and Karnofsky Performance Status (KPS). While age and KPS are known independent predictors of survival in glioblastoma patients, the role of brain reserve in influencing glioblastoma outcomes remains unexplored. This study investigates the relationship between baseline brain-reserve metrics and survival outcomes in isocitrate dehydrogenase 1-wildtype (IDH1-WT) glioblastoma.
METHODS: We analyzed 841 patients with IDH1-WT glioblastoma using clinical data and presurgical MR images sourced from 4 inst

Adult-type diffuse gliomas are defined by the isocitrate-dehydrogenase (IDH) axis, yet how this biology constrains malignant cell-state repertoires and their immune-modulatory potential remains unclear. We profiled 69 tumors using single-cell RNA sequencing (scRNA-seq), single-nucleus RNA sequencing (snRNA-seq), and spatial transcriptomics, and resolved seven recurrent malignant cell states: astrocyte-like progenitor cells (ALPC), hypoxia, cycling G1/S, cycling G2/M, oligo-lineage, neuro-oligo-lineage, and neuro-lineage. The IDH axis strongly biased state occupancy and malignant-immune composi

This article of discusses blast crisis chronic myeloid leukemia (CML), which is the most aggressive CML phase marked by rapid progression, substantial mutational complexity, and resistance to standard tyrosine kinase therapies. The methodology combines whole exome sequencing and machine learning to identify the molecular subtypes of blast crisis-CML and repurpose existing Food and Drug Administration-approved drugs on the basis of the Catalogue of Somatic Mutations in Cancer mutational patterns. In a pilot cohort (n = 7), three exploratory genomic clusters were identified: Breast cancer gene 2

Cholangiocarcinomas (CCAs) are cancers that arise in the peripheral or central bile ducts. They have substantial heterogeneity in their etiologies and histopathologic phenotypes, ranging from cholangiolar carcinomas arising from the smallest bile ducts or from dedifferentiating hepatocytes through cellular plasticity, to ductular cancers in medium and large-sized bile ducts. The surgical delineation of intrahepatic, perihilar, and distal CCAs does not match the biological classification, as intrahepatic CCAs include both small duct cholangiolar carcinoma and intermediate duct ductular or mucin

Nanomaterials are increasingly recognized as stressors in biological wastewater treatment systems, yet the effects on biomass yield remain poorly understood. This study systematically evaluated the effects of low-dose graphene oxide (GO; 0.1 and 1 mg/L) on pollutant removal and sludge yield in activated sludge systems. At day 40, sludge yields were 0.356, 0.298, and 0.237 g VSS/g COD in the Control, 0.1 and 1 mg/L GO systems, respectively, corresponding to reductions of 16.29% (p = 0.055) and 33.43% (p < 0.05) without compromising nitrogen removal. GO exposure also loosened floc structure and

Glioblastoma multiforme (GBM), isocitrate dehydrogenase (IDH)-wildtype, is the most aggressive primary malignant tumor of the central nervous system, characterized by poor prognosis and high recurrence rates despite standard multimodal treatment. This study investigates the molecular response of glioblastoma cells to 5-aminolevulinic acid (5-ALA)-based photodynamic therapy (PDT), focusing on gene expression changes associated with apoptosis, ferroptosis, and oxidative stress. Human glioblastoma T98G cells were treated with 5-ALA followed by light irradiation, and gene expression was analyzed u

Targeting metabolic dependencies of leukemic stem cells (LSC) may open avenues to improve outcomes of patients suffering from acute myeloid leukemia (AML). LSCs rely heavily on an active tricarboxylic acid (TCA) cycle and mitochondrial oxidative phosphorylation whereas healthy hematopoietic stem and progenitor cells (HSPCs) possess more metabolic flexibility. Here, we identify the TCA cycle enzyme isocitrate dehydrogenase 3 (IDH3) as a critical and selective regulator of LSC maintenance. IDH3 is more abundant in LSCs compared to healthy HSPCs, and TCA cycle activity correlates with inferior cl

BACKGROUND: Malignant pleural effusion (MPE) is a common manifestation in advanced malignancies. However, its differentiation from benign pleural effusions, such as parapneumonic effusion (PPE) and tuberculous pleural effusion (TPE), remains challenging. This study aimed to profile differentially expressed proteins and metabolites between MPE, PPE, and TPE.
METHODS: Pleural effusion samples from patients with MPE, PPE, and TPE were collected and subjected to proteomic and metabolomic profiling. Differentially expressed proteins (DEPs) and differentially abundant metabolites (DAMs) were identi

BACKGROUND AND OBJECTIVES: Resection of insular gliomas remains challenging because of their deep-seated location and proximity to eloquent neurovascular structures. Although the transsylvian approach through a standard pterional craniotomy is widely used, we present a minimally invasive alternative using a single burr hole technique.
METHODS: A prospective analysis was performed on 6 patients who underwent burr hole surgery for insular gliomas using a technique termed burr hole microsurgery.
RESULTS: In 6 patients, resection of insular tumors was performed using a 14-mm burr hole approach.

Most human salmonellosis cases are linked to the consumption of poultry products contaminated with Salmonella enterica serovar Typhimurium. In poultry, infections cause intestinal inflammation, decreased weight gain, lower egg production, and potential economic losses for the poultry sector. This study examined the effects of a natural antimicrobial mixture (AuraShield, As) on Salmonella enterica serovar Typhimurium SE10/72 infection of chicken macrophage-like cells (HD11), focusing on infection inhibition, inflammatory response, cell viability, metabolic changes, and oxidative stress. The HD1

80% of patients with low-grade gliomas containing mutations of the metabolic enzyme isocitrate dehydrogenase (IDH) experience seizures, of which nearly half are drug-refractory. Studies in isolated neuronal cultures have led to the hypothesis that the metabolite of the mutant IDH, D-2-hydroxyglutarate (D-2-HG), is epileptogenic by mimicking glutamate and activating NMDA receptors. To test this hypothesis, we used whole-cell patch-clamp techniques to determine the direct effects of D-2-HG on neuronal and synaptic functions and epileptiform activity in neocortical slices that preserve natural an
