Alpha-2,3 sialylation differentiate the limbal and corneal epithelial cell phenotypes.

PURPOSE: The initial differentiation event for the corneal epithelial cell lineage occurs as the limbally localized stem cells yield, through mitosis, the highly proliferative, transiently amplifying corneal peripheral cells. This differentiation is characterized by the expression of tissue-specific cytokeratins, as well as the loss of alpha-enolase and pigmentation. All these are intracellular events. The aim of this study was to identify and characterize, through lectin analysis, changes in cell surface properties associated with differentiation. METHODS: Cryostat sections of the limbo-corn

Rho kinase inhibitor Y-27632 and dual media culture approach promote the construction and transplantation of rabbit limbal epithelial cell sheets via cell spheroid culture and auto-bioprinting.

The shortage of corneal donors and the limitations in tissue engineering grafts, such as biocompatibility and mechanical properties, pose significant challenges in corneal transplantation. Here, for the first time, we investigate the effect of Rho kinase inhibitor Y-27632 and a dual media culture approach, including proliferative media (M1) and stabilizing media (M2), on rabbit limbal epithelial stem cells (LESCs), aiming to explore the feasibility of constructing corneal cell sheets in vitro through auto-bioprinting and assessing their corneal wound healing capacity in vivo. Y-27632 has prima

Functional study of Krt17 in corneal epithelial regeneration and stemness regulation.

Corneal limbal epithelial stem cells (LSCs) are essential for maintaining corneal epithelial homeostasis and mediating injury repair. Although various LSC-associated markers have been reported, the dynamic expression characteristics of the stress-inducible keratin 17 (Krt17) during corneal development and injury, as well as its potential association with LSC function, remain insufficiently explored. In this study, we systematically evaluated the temporal expression pattern of Krt17 in mouse corneal development and wound healing, and further validated its functional relevance to stemness, proli

Development of a microscopy-based diagnostic test for alkali injury-induced limbal stem cell deficiency through Autofluorescence Multispectral Imaging.

Diagnosing limbal stem cell deficiency (LSCD) remains challenging, and this study evaluates a newly developed Autofluorescence Multispectral Imaging (AFMI) technology for its diagnostic potential. Unstained fixed corneal cross-sections from 36 mice were used in this study. The right eyes of these mice had alkali-induced LSCD, while the left eyes served as healthy controls. Autofluorescence multispectral images of the corneal epithelium were acquired across multiple spectral channels to track changes in endogenous fluorophore spectra. Big data analytics and machine learning classifiers were app

Limbal and Conjunctival Epithelial Cell Cultivation on Contact Lenses-Different Affixing Techniques and the Effect of Feeder Cells.

OBJECTIVES: Corneal blindness due to limbal stem-cell deficiency can be treated by transplantation of cultivated limbal epithelial stem cells (LESCs). We examined LESC cultivation on a contact lens (CL) carrier. Our goal was to optimize explant affixation and assess the possible benefit of 3T3 feeder cells. METHODS: Human cadaver limbal and conjunctival explants were allowed to attach to CLs under the airflow of the laminar box (dried group) or affixed on CLs using suturing (sutured group) or tissue adhesives (glued group), then cultivated with or without 3T3 feeder cells. Outgrowth efficienc

Amniotic membrane modulates innate immune response inhibiting PRRs expression and NF-κB nuclear translocation on limbal myofibroblasts.

Corneal damage observed in a viral infection such as herpetic stromal keratitis is mainly caused by proinflammatory molecules released by resident cells in the response to viral antigens. There are pattern recognition receptors like MDA5, RIG-1, and TLR3, that recognize viral dsRNA and after activation, the innate immune response is exacerbated inducing the synthesis and secretion of inflammatory cytokines through NF-κB activation. Amniotic membrane (AM) has demonstrated to reduce inflammation by several mechanisms, however the effect of AM on innate immune receptors such as MDA5, RIG-1, and T

Fresh-tissue corneolimbal covering graft for large corneal perforation following childhood trachoma.

BACKGROUND AND OBJECTIVE: When corneal perforation develops in patients with severe ocular surface disease and limbal stem cell deficiency following childhood trachoma infection, penetrating keratoplasty is doomed to fail. To overcome this problem, the use of a fresh-tissue corneolimbal covering graft is described. PATIENTS AND METHODS: Retrospective interventional case series of four patients who presented to a tertiary center from 2003 to 2005 with a large corneal perforation due to ocular surface disease. Emergency surgery consisted of suturing a fresh-tissue corneolimbal covering graft to

The effect of in vivo grown corneal epithelium transplantation on persistent epithelial defects with limbal stem cell deficiency.

We report our experience with corneal epithelium, grown in vivo, transplantation in three patients with persistent epithelial defect (PED). The three patients had ocular surface disease unresponsive to standard treatments and were therefore chosen for transplantation. They underwent transplantation of epithelial sheets, grown in vivo, to the most affected eye. In vivo cultivation was carried out in the cornea of a living related donor. After epithelialization was completed, the epithelium grown on an amniotic membrane was harvested gently; it was then transplanted into the patient's eye after

[A new operation in the treatment of refractory glaucoma: limbo-sclerectomy with valvular drainage of supraciliary space].

A new antiglaucoma operation has been developed: limbosclerectomy with valvular drainage of the supraciliary space. The operation was performed on 82 eyes with artiphakia, pseudophakia, and previously operated primary open-angle glaucoma. The patients were observed for 1.3 +/- 0.6 years after the intervention. Compensation of intraocular pressure and a significant increase in the coefficient of liquid discharge facility were observed in remote periods in all patients: in 95.1% patients without hypotensive therapy and in 4.9% cases instillations of 0.5% thimolol maleate twice a day were needed.

Steroid inhibition of limbal blood and lymphatic vascular cell growth.

Steroids are widely used in the prevention of corneal neovascularization in a wide range of natural and experimental situations. However, no information is available on their effect on the growth of the individual limbal blood vascular cells or of lymphatic cells involved in corneal neovascularization. In addition, tritiated thymidine labelling index is commonly used as an indicator of cell population but doubt exists as to whether it truly represents cell growth. Remote thermal cautery of the rat cornea was used to elicit corneal neovascularization. New cell growth was measured by tritiated t
