Association between phospholipid metabolism in plasma and spontaneous preterm birth: a discovery lipidomic analysis in the cork pregnancy cohort.

INTRODUCTION: Preterm birth (PTB) is defined as birth occurring before 37 weeks' gestation, affects 5-9% of all pregnancies in developed countries, and is the leading cause of perinatal mortality. Spontaneous preterm birth (sPTB) accounts for 31-50% of all PTB, but the underlying pathophysiology is poorly understood. OBJECTIVE: This study aimed to decipher the lipidomics pathways involved in pathophysiology of sPTB. METHODS: Blood samples were taken from SCreening fOr Pregnancy Endpoints (SCOPE), an international study that recruited 5628 nulliparous women, with a singleton low-risk pregnanc…

Fatty acid profiles as a potential lipidomic biomarker of exposure to brevetoxin for endangered Florida manatees (Trichechus manatus latirostris).

Fatty acid signature analysis (FASA) is an important tool by which marine mammal scientists gain insight into foraging ecology. Fatty acid profiles (resulting from FASA) represent a potential biomarker to assess exposure to natural and anthropogenic stressors. Florida manatees are well studied, and an excellent necropsy program provides a basis against which to assess this budding tool. Results using samples from 54 manatees assigned to four cause-of-death categories indicated that those animals exposed to or that died due to brevetoxin exposure (red tide, or RT samples) demonstrate a distinct

Deciphering the effect of sugar-to-water mass ratios on key aroma formation in roasted ducks: Insights from volatilomics, lipidomics, and Maillard reaction.

Sugar-coating is a pivotal step in roasted duck processing, yet its optimization has relied on artisanal experience rather than standardized protocols. This study explored how varying sugar-to-water mass ratios affect the aroma formation. The 1:6 sugar-to-water mass ratio produced the richest aroma compounds in roasted ducks compared with the ratios of 1:5, 1:7 and 1:8. This optimal condition likely facilitated the glucose penetration from the skin into the meat, enhancing Maillard reactions with arginine and alanine for pyrazine formation. It also promoted the contribution of triglycerides, c

A lipidomics approach identifies the lipid classes modulated in response to hepatic steatosis in obese fa/fa rats relative to lean Zucker rats.

Mass spectrometry-based lipidomics analysis of liver tissues from obese fa/fa and lean Zucker rats (n = 8/group) was carried out. Hepatic steatosis, the accumulation of >5% lipid (w/w) in liver due to obesity forms the physiological context of this study, and differences in the lipid profile of steatotic and healthy livers are expected to provide insight into how lipid metabolic processes respond to obesity. A total of 1000 lipids within 21 different classes of lipids were identified using an iterative auto-MSMS analyses of all liver tissues combined. Among these, 175 lipids were significantly…

Research on the mechanism of the anti-myocardial infarction effect of the Qiliqiangxin capsule on heart failure rats via nontargeted metabolomics and lipidomics.

BACKGROUND: Qi Li Qiang Xin (QLQX) capsule has a solid theoretical basis and clinical efficacy in the treatment of chronic heart failure; however, the underlying mechanisms remain obscure. This study was designed to determine the effect of the QLQX on the treatment of heart failure and delineate the underlying mechanisms via a nontargeted metabolomics and lipidomics approach. METHODS: A rat model of heart failure after myocardial infarction (MI) was established via permanent ligation of the anterior descending branch of the left coronary artery. The rats were then randomly divided into the SH…

Advanced unified monophasic lipid extraction protocol with wide coverage on the polarity scale optimized for large-scale untargeted clinical lipidomics analysis of platelets.

Lipid extraction is a critical step in sample preparation of lipidomics studies. Biphasic liquid-liquid extraction protocol with methyl tert-butyl ether (MTBE)/methanol (MeOH) as organic solvents are widely adopted by researchers nowadays as an eco-friendly replacement of classic Folch, and Bligh&Dyer protocols. Yet, it has some limitations such as suboptimal performance for the most polar lipids (e.g. acylcarnitines), complicated handling as it requires phase separation, and is therefore non-ideal for large-scale clinical studies. To advance the extraction protocol for large-scale clinical li…

Effect of micro-pressure stewing technology on the presence of key aroma compounds in chickens: New insight from lipidomics and heat transfer analysis.

Micro-pressure (104 kPa) stewing technology has become a popular cooking method in Chinese households, but the mechanisms underlying the aroma presence in meat products remain unclear. The aroma formation in chicken during micro-pressure stewing was characterized using lipidomics, volatilomic, and heat conduction approach. Twenty aroma compounds were identified as key odorants, particularly aldehydes and alcohols. Most aroma compounds displayed a remarkable increase in concentration during micro-pressure stewing, peaking at 24-30 min, with the highest cumulative contribution rate of OAVs obser…

Lipidomics Analysis of Free Fatty Acids in Human Plasma of Healthy and Diabetic Subjects by Liquid Chromatography-High Resolution Mass Spectrometry (LC-HRMS).

Targeted analytical methods for the determination of free fatty acids (FFAs) in human plasma are of high interest because they may help in identifying biomarkers for diseases and in monitoring the progress of a disease. The determination of FFAs is of particular importance in the case of metabolic disorders because FFAs have been associated with diabetes. We present a liquid chromatography-high resolution mass spectrometry (LC-HRMS) method, which allows the simultaneous determination of 74 FFAs in human plasma. The method is fast (10-min run) and straightforward, avoiding any derivatization st

Metabolomics and Lipidomics Study of Mouse Models of Type 1 Diabetes Highlights Divergent Metabolism in Purine and Tryptophan Metabolism Prior to Disease Onset.

With the increase in incidence of type 1 diabetes (T1DM), there is an urgent need to understand the early molecular and metabolic alterations that accompany the autoimmune disease. This is not least because in murine models early intervention can prevent the development of disease. We have applied a liquid chromatography (LC-) and gas chromatography (GC-) mass spectrometry (MS) metabolomics and lipidomics analysis of blood plasma and pancreas tissue to follow the progression of disease in three models related to autoimmune diabetes: the nonobese diabetic (NOD) mouse, susceptible to the develop…

Modeling the organizational heterogeneity of cholesterol-enriched microdomains in the neuronal membranes of gray and white matter of Alzheimer's brain: a computational lipidomics study.

Alzheimer's disease (AD) is a leading cause of death among the elderly, with no existing treatment. The development of therapies is further hindered by a limited understanding of the molecular pathogenesis and the absence of reliable early-detection biomarkers. Neuroimaging and lipidomic studies reveal structural and biochemical alterations in both gray and white matter in AD patients, including disruptions in membrane organization and neuronal signaling pathways. In the present work, we employed lipidomics-guided modeling of membranes in gray and white matter regions under healthy and disease…
