BACKGROUND: Lymphoedema is a chronic and progressive disease characterised by excessive accumulation of lymph in the interstitial compartment, leading to tissue swelling and fibroadipose deposition. Lymphangiogenesis is partly regulated by ketone body oxidation, and a ketogenic diet (KD) has shown therapeutic efficacy in a preclinical mouse tail lymphoedema model. BACKGROUND: Lymphedema (LE) is characterized by inadequate interstitial fluid drainage because of disrupted lymphatic vasculature. Despite the pivotal role of angiopoietin (Ang)-Tie2 signaling in lymphatics, the therapeutic potential of Tie2 modulation in LE remains unknown.
METHODS: This study investigates the molecular and pathologic changes associated with LE and transcriptomic analyses. In addition, a mouse hindlimb LE model was established by obstructing superficial and deep lymphatic drainage, allowing the evaluation of structural, functional, and molecular alterations accompanying LE.

Indocyanine green lymphography has recently been used to assess lymphatic vessel function in lymphedema patients. Postoperative collateral lymphatic vessels toward ipsilateral axillary lymph nodes are rarely seen above the umbilical level in lower lymphedema patients.
