Increased risk for congenital heart defects in children carrying the ABCB1 Gene C3435T polymorphism and maternal periconceptional toxicants exposure.

BACKGROUNDS: The etiology of congenital heart defect (CHD) is commonly believed to involve the interaction of multiple environmental and genetic factors. This study aimed to explore the joint effects of the ABCB1 gene C3435T polymorphism and maternal periconceptional toxicants exposure on the CHD risk in a Han Chinese population. METHODS: An age and gender matched case-control study with standardized data collection involving 201 pairs was conducted. Periconceptional toxicants exposure was obtained through a structured questionnaire. A job exposure matrix (JEM) was used for toxicants exposure

Maternal occupational exposure to polycyclic aromatic hydrocarbons: effects on gastroschisis among offspring in the National Birth Defects Prevention Study.

BACKGROUND: Exposure to polycyclic aromatic hydrocarbons (PAHs) occurs in many occupational settings. There is evidence in animal models that maternal exposure to PAHs during pregnancy is associated with gastroschisis in offspring; however, to our knowledge, no human studies examining this association have been conducted. OBJECTIVE: Our goal was to conduct a case-control study assessing the association between estimated maternal occupational exposure to PAHs and gastroschisis in offspring. METHODS: Data from gastroschisis cases and control infants were obtained from the population-based Nati…

Fetal plasma insulin and thyroid hormone levels during acute in utero ethanol exposure in a maternal-fetal sheep model.

The effects of acute in utero ethanol (ETOH) treatment on basal and stimulated thyroid and insulin levels in fetal plasma were studied in chronically cannulated fetal sheep. In test situations, pregnant ewes (0.78-0.88 gestation) which were chronically cannulated received 2 g/kg ETOH [25% (vol/vol) in isotonic saline] for 2 h; this was followed by a maintenance iv infusion of 0.13 g/kg ETOH. Control animals received isovolemic infusions of isotonic saline. Fetal arterial plasma samples were obtained after the 2-h infusion, and basal levels of T3, T4, glucose, and insulin were measured. The 2-h

Maternal Diet and Offspring ASD and ADHD: A Critical Narrative Review of Dietary Patterns, Choline, and Polyphenol-Related Exposures.

Maternal diet may influence fetal neurodevelopment through nutrient availability, placental function, and maternal-fetal metabolite signaling. This critical narrative review used a structured, purposive search to organize human evidence on prenatal dietary patterns, choline, and polyphenol-related exposures according to outcome directness: diagnosed autism spectrum disorder (ASD) or attention-deficit/hyperactivity disorder (ADHD), related dimensional traits, and general neurodevelopment. Whole-diet studies provide the most direct human evidence, but diagnostic findings are cohort-dependent and

Intrauterine exposure to maternal diabetes and the risk of developing epilepsy in children: a national cohort study of 2.3 million children.

BACKGROUND: Approximately half of the epilepsy cases lack a documented cause. Despite growing evidence linking maternal diabetes to pregnancy complications and impaired child neurodevelopment, little is known about its potential association with child epilepsy. This study aims to determine whether intrauterine exposure to maternal pre-existing type 1 diabetes mellitus (T1DM), type 2 diabetes mellitus (T2DM), or gestational diabetes mellitus (GDM) increases the risk of epilepsy in children. METHODS: This study included live-born children without major malformations in Sweden from 1998 to 2021.

Gene expressions in newborns and the association with prenatal exposure to phthalate plasticizers - The Taiwan Maternal and Infant Cohort Study (TMICS).

Phthalates, notably di-(2-ethylhexyl) phthalate (DEHP), are pervasive endocrine-disrupting chemicals used in consumer products. Prenatal DEHP exposure has been linked to adverse developmental and immune outcomes, yet the molecular mechanisms remain unclear. We conducted a prospective study using cord blood transcriptomics to examine associations between maternal DEHP exposure levels and neonatal gene expression. Maternal urinary DEHP metabolites were measured in the Taiwan Maternal and Infant Cohort Study, and RNA sequencing was performed on cord blood from 125 newborns. After adjustment for m

Progressive motor dysfunction and loss of cerebellar Purkinje and granule cells in rat offspring after maternal exposure to imidacloprid.

Imidacloprid (IMI), a major neonicotinoid insecticide, raises concerns about neurodevelopmental abnormalities, particularly attention deficit hyperactivity disorder. However, the involvement of cerebellar development in IMI-induced developmental neurotoxicity has not been studied. Here, this study investigated the maternal exposure effects of IMI on the developing cerebellum in rats. Pregnant Sprague-Dawley rats were fed diet containing IMI at 0 (control), 83, 250 or 750 ppm from gestational day 6 through gestation, and dams treated with the diet during lactation until day 21 postpartum. Male …

Maternal exposure to fine particulate matter and pregnancy outcomes in women undergoing in vitro fertilization: A multicenter retrospective study.

Few large-scale studies have systematically examined the effects of maternal exposure to fine particulate matter (PM2.5) on live birth in women undergoing in vitro fertilization (IVF). This study aimed to investigate the associations between ambient PM2.5 exposure and live birth in women treated with IVF, and determine critical periods, key failure events, and vulnerable populations affected by such exposure. We included 58,637 patients from six reproductive centers in China between 2016 and 2021. We defined six exposure windows and adopted logistic regression with random-effect models to inve…

Maternal exposure to Waste-to-Energy plants and risks of adverse birth outcomes and hypertensive pregnancy disorders in Lazio, Italy.

Evidence on the effects of waste incineration on adverse birth outcomes (ABO) and hypertensive disorders of pregnancy (HDP) is limited. We investigated the association between maternal exposure to a waste-to-energy (WTE) plant in Lazio Region and ABO/HDP. Employing a difference-in-differences (DiD) design, we assessed changes related to the plant's deactivation (2017), comparing outcomes to a control WTE plant that remained operational. Mothers with geocoded residential address within 7 km from Colleferro (exposed area) and San Vittore del Lazio (control area) plants who delivered between 2007…

Charting the Impact of Maternal Antibodies and Repeat Exposures on Sapovirus Immunity in Early Childhood From a Nicaraguan Birth Cohort.

BACKGROUND: Sapovirus is an important cause of acute gastroenteritis in childhood. While vaccines against sapovirus may reduce gastroenteritis burden, a major challenge to their development is a lack of information about natural immunity. METHODS: We measured sapovirus-specific IgG in serum collected between 2017 and 2020 of mothers soon after delivery and at 6 time points in Nicaraguan children until 3 years of age (n = 112 dyads), using virus-like particles representing 3 sapovirus genotypes (GI.1, GI.2, GV.1). RESULTS: Of the 112 children, 16 (14.3%) experienced at least 1 sapovirus gastr

The relationship between maternal exposure to ambient air pollutants and premature rupture of membranes: A systematic review and meta-analysis.

Air pollution is an environmental stimulus that may predispose pregnant women to preterm rapture of membrane (PROM). However, the relationship of maternal exposure to air pollutants and PROM is still unclear. To investigate the relationship between the long-term and short-term maternal exposure to air pollution and PROM. We searched all studies published in PubMed, Embase and Web of Science up to February 2024. The studies provided quantitative effect estimates with 95% confidence intervals, for the impact of short-term (<30 days) or long-term (≥30 days) maternal exposure to air pollutants on …

Maternal exposure to fine particulate matter before and during pregnancy, and the risk of birth defects: A population-based study.

Although epidemiological evidence has linked maternal exposure to air pollution with adverse birth outcomes, the association between exposure to fine particulate matter (PM2.5) and the risk of total birth defects in highly polluted developing regions remains limited. The objective of this study was to assess the relationship between maternal exposure to PM2.5 before and during pregnancy and the occurrence of birth defects. This study included 16,080 births with birth defects and 567,483 controls born between 2014 and 2019 in 14 cities in Liaoning Province, China. The assessment of exposure was…

Maternal exposure to fine particulate matter in the air and risk for fetal congenital heart defects: A case-control study.

Prior research into the association between fine particulate matter (PM2.5) exposure and the risk for fetal congenital heart defect (CHD) has yielded inconclusive and conflicting results. More epidemiologic evidence from different regions is necessary. A case-control study was conducted with 360 CHD cases and 3600 healthy newborns. Both the cases and the controls were registered by the mothers in the Prenatal Health Care System during the first trimester and gave birth at hospitals in the Tongzhou District of Beijing between 2013 and 2018. Information on PM2.5 was obtained from satellite remot

Prenatal exposure to maternal hypertension and higher body mass index and risks of neurodevelopmental and psychiatric disorders during childhood.

INTRODUCTION: Hypertensive disorders of pregnancy (HDP) or prepregnancy overweight/obesity are independently associated with the risk for certain neurodevelopmental and psychiatric disorders in offspring. These two conditions often co-exist but the risk from combined exposure is unknown. We investigated whether specific subtypes of maternal HDP, along with prepregnancy overweight/obesity, were associated with the distinct risk of neurodevelopmental and psychiatric disorders in offspring during childhood. MATERIAL AND METHODS: This prospective, population-based cohort study used data from 652

Maternal exposure to bisphenols, phthalates, perfluoroalkyl acids, and trace elements and their associations with gestational diabetes mellitus in the APrON cohort.

The increasing global prevalence of gestational diabetes mellitus (GDM) has been hypothesized to be associated with maternal exposure to environmental chemicals. Here, among 420 women participating in the Alberta Pregnancy Outcomes and Nutrition (APrON) cohort study, we examined associations between GDM and second trimester blood or urine concentrations of endocrine disrupting chemicals (EDCs): bisphenol-A (BPA), bisphenol-S (BPS), twelve phthalate metabolites, eight perfluoroalkyl acids (PFAAs), and eleven trace elements. Fifteen (3.57%) of the women were diagnosed with GDM, and associations …

Impacts of prenatal environmental exposures on fetal-placental-maternal bile acid homeostasis and long-term health in offspring.

During pregnancy, the maternal body undergoes a series of adaptative physiological changes, leading to a slight increase in serum bile acid (BA) levels. Although the fetal liver can synthesize BAs since the first trimester through the alternative pathway, the BA metabolic system is immature in the fetus. Compared to adults, the fetus has a distinct composition of BA pool and limited expression of BA synthesis enzymes and transporters. Besides, the "enterohepatic circulation" of BAs is absent in fetus. Thus, fetal BAs need to be transported to the mother through the placenta for further metabol
