An Operationalized Multisymptomatic Model of Neuroses (OMMON): toward a reintegration of diagnosis and treatment in behaviour therapy.

Experimental results from a sample of 216 patients with four different "symptom-neuroses" show that 65% to 90% of these patients have different combinations of multiple symptomatology. With a background of these data, we present an Operationalized Multisymptomatic Model of Neuroses (OMMON), based on self-rating assessment of these patients on 4 symptom scales. Individual ratings on each scale are dichotomized into (+) or (-) results with regard to defined cut-off points and the model is derived from their 16 mathematically possible combinations. Subsequent analysis of these data (from a single

Temporal dynamics of students' physiological arousal in personalized learning contexts: a quasi-experimental study based on electrodermal activity.

Physiological arousal varies throughout classroom learning, yet its temporal dynamics during personalized learning in authentic primary-school classrooms remain unclear. This quasi-experimental study used electrodermal activity (EDA) to examine the lesson-wide temporal trend, dynamic trajectory, and phase-specific arousal level and activation frequency in an "Introduction to Circles" lesson. Two intact fourth-grade classes in Shanghai participated (analytic sample: experimental n = 23; control n = 27). The experimental class received personalized learning based on cognitive diagnosis, targetin

Logical analysis of diffuse large B-cell lymphomas.

OBJECTIVE: The goal of this study is to re-examine the oligonucleotide microarray dataset of Shipp et al., which contains the intensity levels of 6817 genes of 58 patients with diffuse large B-cell lymphoma (DLBCL) and 19 with follicular lymphoma (FL), by means of the combinatorics, optimisation, and logic-based methodology of logical analysis of data (LAD). The motivations for this new analysis included the previously demonstrated capabilities of LAD and its expected potential (1) to identify different informative genes than those discovered by conventional statistical methods, (2) to identif

Reference equation for the 2-minute walk test in adults and the elderly.

BACKGROUND: The 2-min walk test (2MWT) has been used in several health conditions, but the interpretation of its results is limited due to a lack of reference values. The aim of this study was to establish a reference equation to predict the distance walked (DW) in the 2MWT for healthy adults and the elderly and to test its reproducibility. METHODS: We evaluated 390 healthy subjects (195 male), 18-89 y old, with normal spirometry and no history of previous chronic diseases. Two 2MWTs were performed on the same day, 30 min apart. To test the reliability of the prediction equation, 70 subjects

Prioritising Data Quality Governance for AI in Prostate Cancer: A Methodological Proof-of-Concept Study Using Neural Networks for Risk Stratification.

Background: An accurate D'Amico risk stratification is mandatory for prostate cancer (PCa) management. The purpose of this proof-of-concept study was to establish a methodological framework for integrating validated clinical nomograms with strict data-quality governance in order to generate reliable artificial neural networks (ANNs), even when the sample is small. Methods: We performed a retrospective analysis of a curated cohort of 49 patients from one centre. A multilayer perceptron (MLP) was trained using 11 variables, including the ISUP biopsy grade and Briganti nomogram. Model development

Mathematical Modeling of HDV RNA, HBV DNA, and HBsAg Dynamics During Lonafarnib-Based Therapy: Insights from the LOWR HDV-1 Study.

Lonafarnib (LNF) is an investigational drug targeting hepatitis delta virus (HDV) but not hepatitis B virus (HBV), providing a unique opportunity to model HDV kinetics and how changes in HDV affect HBV. We performed a detailed kinetic analysis and developed a mathematical model to explain serum HBV DNA, HDV RNA and hepatitis B surface antigen (HBsAg) kinetics in 15 HBV/HDV coinfected patients receiving LNF-based treatment. After a delay of 0-2 days, patients experienced a rapid 1st-phase HDV-decline followed by either a viral plateau, 2nd slower-decline phase, or viral breakthrough (VB). LNF m

Modeling Combination Therapies and T Cell Exhaustion Dynamics in the Tumor Under Immune Checkpoint Blockade.

Chronic antigen exposure in the tumor microenvironment drives CD 8 + T cell exhaustion, marked by increased inhibitory receptors and diminished effector functions. Immune checkpoint blockade seeks to prevent or reverse exhaustion, but its success relies on the pre-existing state of tumor-infiltrating T cells. To investigate this, we developed a mathematical model examining: (1) how T cell exhaustion disrupts tumor-immune equilibrium, (2) anti-PD-L1 efficacy across exhaustion states, and (3) efficacy of next-generation therapies (e.g., IFN α -anti-PD-L1, PD1-IL2v). Stability analysis and simul

In vivo Quantification of Neural Criticality and Complexity in Mouse Cortex and Striatum in a Model of Cocaine Abstinence.

UNLABELLED: Self-organized criticality is a hallmark of complex dynamic systems at phase transitions. Systems that operate at or near criticality have large-scale fluctuations or "avalanches", the frequency and duration power of which are best fit with a power law revealing them to be scale-free and fractal, and such power laws are ubiquitous. It is an attractive concept in neuroscience since spiking avalanches are exhibited by neural tissue, and may underpin how minuscule events could scale up to circuits and provide adaptive psychobiological function. Much is yet to be understood about criti

Synergetic Control Framework for Autonomous UAV Swarm Coordination.

This study presents a synergetic control approach for coordinating the motion of a UAV swarm based on self-organization principles and energy potential interactions. The proposed method effectively addresses the known limitations of traditional potential field techniques by introducing a procedure for synthesizing the synergetic control of swarm motion using a virtual force field with symmetric attraction and repulsion functions. A Lyapunov/LaSalle stability analysis and a geometric safety radius argument show that this keeps the swarm's motion conflict-free, subject to the platform's kinemati

Variational free energy as a conceptual framework for understanding habituation in spinal cord stimulation.

INTRODUCTION: Habituation, or loss of clinical benefit over time, is a frequent problem in spinal cord stimulation (SCS). Existing programming strategies such as waveform cycling or closed-loop stimulation try to address it, but there is no common conceptual framework. This paper introduces the variational free energy (VFE) principle as a way to think about habituation in terms that connect clinical observation with computational models of the brain. METHODS: We reviewed the core ideas of VFE, as formulated by Friston and colleagues-recognition density, prior, posterior, accuracy, complexity,

Membrane-decisional architecture: a framework for multigenomic signal prioritization in the plasma membrane nanobiome.

BACKGROUND: Classical models of cellular signaling assume that ligand recognition at the plasma membrane is sufficient to initiate intracellular cascades. However, experimental evidence consistently shows that extracellular signals can engage receptors without producing functional cellular responses and that identical ligand-receptor interactions may generate divergent outcomes depending on membrane organization, receptor clustering, and intracellular routing dynamics. CONCEPTUAL FRAMEWORK: To address this discrepancy, the concept of membrane-decisional architecture is introduced, in which pl

Fluctuating arch symmetry: a comparison of two methods of assessment - applicability and efficiency.

INTRODUCTION: Symmetry is balance, some correspondence in the size, form, and arrangements of parts on opposite sides of a plane, line, or point. The opposite of this concept is asymmetry, or imbalance. OBJECTIVE: This retrospective study compared two methods for assessing arch symmetry with linear measurements based on triangles, to determine their applicability and efficiency. METHODS: Two groups were enrolled: children (n=20) and adults (n=20), and the arch symmetry was assessed from linear measurements. Method 1: the incisor-canine (INC), canine-molar (CM), and incisor-molar (INM) distan

Measurement Uncertainty and Risk of False Compliance Assessment Applied to Carbon Isotopic Analyses in Natural Gas Exploratory Evaluation.

The concept of uncertainty in an isotopic analysis is not uniform in the scientific community worldwide and can compromise the risk of false compliance assessment applied to carbon isotopic analyses in natural gas exploratory evaluation. In this work, we demonstrated a way to calculate one of the main sources of this uncertainty, which is underestimated in most studies focusing on gas analysis: the δ13C calculation itself is primarily based on the raw analytical data. The carbon isotopic composition of methane, ethane, propane, and CO2 was measured. After a detailed mathematical treatment, the

Transport-based morphometry of nuclear structures of digital pathology images in cancers.

Alterations in nuclear morphology are useful adjuncts and even diagnostic tools used by pathologists in the diagnosis and grading of many tumors, particularly malignant tumors. Large datasets such as TCGA and the Human Protein Atlas, in combination with emerging machine learning and statistical modeling methods, such as feature extraction and deep learning techniques, can be used to extract meaningful knowledge from images of nuclei, particularly from cancerous tumors. Here we describe a new technique based on the mathematics of optimal transport for modeling the information content related to

Calculating stochastic inactivation of individual cells in a bacterial population using variability in individual cell inactivation time and initial cell number.

The traditional log-linear inactivation kinetics model considers microbial inactivation as a process that follows first-order kinetics. A basic concept of log reduction is decimal reduction time (D-value), which means time/dose required to kill 90% of the relevant microorganisms. D-value based on the first-order survival kinetics model is insufficient for reliable estimations of bacterial survivors following inactivation treatment. This is because the model does not consider the inactivation curvature and variability in bacterial inactivation. However, although the D-value has some limitations

Testing of a novel percutaneous temporary aortic valve in a rabbit model of acute severe aortic regurgitation: an experimental study.

PURPOSE: Device management of hemodynamic instability due to acute aortic regurgitation is not available. A novel, catheter-based, temporary aortic valve (TAV) has been in development. Early prototypes (balloon-based TAV) have undergone proof-of-concept studies in mathematical, bench and animal models. The redesigned membrane-based TAV prototype is evaluated in a rabbit model of acute severe aortic regurgitation. METHODS: Acute aortic regurgitation was simulated by deploying a self-expanding endovascular stent across the aortic annulus. Eight rabbits of body weights ranging 4.9-5.4 kg were ra

Mathematical models for predicting human mobility in the context of infectious disease spread: introducing the impedance model.

BACKGROUND: Mathematical models of human mobility have demonstrated a great potential for infectious disease epidemiology in contexts of data scarcity. While the commonly used gravity model involves parameter tuning and is thus difficult to implement without reference data, the more recent radiation model based on population densities is parameter-free, but biased. In this study we introduce the new impedance model, by analogy with electricity. Previous research has compared models on the basis of a few specific available spatial patterns. In this study, we use a systematic simulation-based ap

Contribution of intestine, bone, kidney, and dialysis to extracellular fluid calcium content.

Calcium (Ca) balance is the net of Ca intake and output from the body over a period of time. The concept of Ca balance does not consider the redistribution of Ca that often occurs in patients with chronic kidney disease (CKD), especially those who are on dialysis, which is often in the form of soft tissue and/or vascular calcification. In this article, we consider movement of Ca with respect to the extracellular fluid (ECF) and develop a mathematical formulation for Ca homeostasis with respect to the ECF that includes input and output from the diet, the bone, the kidney, and dialysis. We consi

The development of an age-structured model for trachoma transmission dynamics, pathogenesis and control.

BACKGROUND: Trachoma, the worldwide leading infectious cause of blindness, is due to repeated conjunctival infection with Chlamydia trachomatis. The effects of control interventions on population levels of infection and active disease can be promptly measured, but the effects on severe ocular sequelae require long-term monitoring. We present an age-structured mathematical model of trachoma transmission and disease to predict the impact of interventions on the prevalence of blinding trachoma. METHODOLOGY/PRINCIPAL FINDINGS: The model is based on the concept of multiple reinfections leading to
