Intact pituitary function is decisive for the catabolic response to TNF-α: studies of protein, glucose and fatty acid metabolism in hypopituitary and healthy subjects.

CONTEXT: TNF-α generates inflammatory responses and insulin resistance, lipolysis, and protein breakdown. It is unclear whether these changes depend on intact hypothalamo-pituitary stress hormone responses to trigger the release of cortisol and growth hormone. OBJECTIVE: To define differential effects of TNF-α on glucose, protein, and lipid metabolism in hypopituitary patients (without intact hypothalamo-pituitary axis) and healthy controls. DESIGN: Randomized, placebo controlled, single-blinded. Setting, Participants, and Intervention: We studied eight hypopituitary (HP) patients and eight

Effectiveness of grease interceptors in food service establishments for controlling fat, oil and grease deposition in the sewer system.

Источник пометил публикацию как отозванную

The water industry worldwide experiences numerous sewer blockages each year, partially attributed to the accumulation of fat, oil and grease (FOG). Managing this issue involves various strategies, including the requirement for installation of grease interceptors (GIs) installation. However, the claimed efficacy of commercial GIs of eliminating 99 % of FOG has been questioned for many years because FOG deposit formation occurs despite food service establishments (FSEs) using GIs, therefore detailed understanding of FOG wastewater compositions and its removal by GIs is required. This study provi

Inhibition of Hepatic AMPK Pathway Contributes to Free Fatty Acids-Induced Fatty Liver Disease in Laying Hen.

Metabolism-associated fatty liver disease (MAFLD) is one of the most common causes of liver disease; however, the underlying processes remain unknown. This study aimed to investigate the changes of free fatty acids (FFA) on the expression of genes related to the AMP-activated protein kinase (AMPK) signaling pathway in the primary hepatocytes of laying hens. The primary hepatocytes of laying hens were treated with FFA (containing a 2:1 ratio of oleic and palmitic acids) for 24 h. FFA significantly increased lipid droplet accumulation, decreased glycogen synthesis, increased the levels of trigly…

Fatty acids secreted from head and neck cancer induce M2-like Macrophages.

Tumor-infiltrating monocytes can mature into Macrophages that support tumor survival or that display antitumor properties. To explore mechanisms steering Macrophage maturation, we assessed the effects of supernatants from squamous cell carcinoma cell lines (FaDu and SCC) on monocyte-derived Macrophage maturation. Purified monocytes were incubated in medium or medium supplemented with supernatants from FaDu and SCC9 or the leukemia monocytic cell line, THP-1. Macrophages were examined for markers of maturation (CD14, CD68), activation (HLA-DR, CD86, IL15R), scavenger receptor (CD36), toll-like

Potential of Whole Pequi (Caryocar spp.) Fruit-Pulp, Almond, Oil, and Shell-as a Medicinal Food.

Pequi (Caryocar) pulp, the most consumed component of pequi fruit, is one of the richest Brazilian carotenoid sources, and the most important carotenoid food source native to the Cerrado. However, there are considerable differences among pequi species regarding total carotenoids content and carotenoids profile. Caryocar brasiliense Camb. pulp presents higher content of total carotenoids than Caryocar villosum (Aubl.) Pers. Regarding the carotenoids profile, few studies are available in the literature, mainly with C. brasiliense. Pequi pulp also has high contents of lipids, dietary fiber, zinc,

The Association of Prenatal Exposure to Perfluorinated Chemicals with Maternal Essential and Long-Chain Polyunsaturated Fatty Acids during Pregnancy and the Birth Weight of Their Offspring: The Hokkaido Study.

BACKGROUND: Fatty acids (FAs) are essential for fetal growth. Exposure to perfluorinated chemicals (PFCs) may disrupt FA homeostasis, but there are no epidemiological data regarding associations of PFCs and FA concentrations. OBJECTIVES: We estimated associations between perfluorooctane sulfonate (PFOS)/perfluorooctanoate (PFOA) concentrations and maternal levels of FAs and triglyceride (TG) and birth size of the offspring. METHODS: We analyzed 306 mother-child pairs in this birth cohort between 2002 and 2005 in Japan. The prenatal PFOS and PFOA levels were measured in maternal serum samples

Photoactivated azido fatty acid irreversibly inhibits anion and proton transport through the mitochondrial uncoupling protein.

The protonophoretic function of uncoupling protein (UCP) is activated by fatty acids. According to the "docking site" hypothesis (Jezek, P., and Garlid, K. D., J. Biol. Chem. 265, 19303-19311, 1990), the fatty acid binding site is identical with the anion channel of UCP. Skulachev (Skulachev, V. P. (1991) FEBS Lett. 294, 158-162) extended this hypothesis by suggesting that fatty acid anions are transported by UCP and that H+ are delivered by back-diffusion of the protonated fatty acid through the lipid bilayer. In this model, UCP does not transport H+ at all but rather enables fatty acids to a

The effects of different plasma insulin concentrations on lipolytic and ketogenic responses to epinephrine in normal and type 1 (insulin-dependent) diabetic humans.

This study was performed to verify: (1) the ability of different insulin concentrations to restrict the lipolytic and ketogenic responses to exogenous epinephrine administration; (2) whether the ability of insulin to suppress the lipolytic and ketogenic responses during epinephrine administration is impaired in Type 1 (insulin-dependent) diabetic patients. Each subject was infused on separate occasions with insulin at rates of 0.2, 0.4, and 0.8 mU.kg-1.min-1 while normoglycaemic. To avoid indirect adrenergic effects on endocrine pancreas secretions, the so-called "islet clamp" technique was us

Fatty acid synthase in chemoresistance: mechanisms and therapeutic opportunities.

Chemoresistance has been a major obstacle to the efficient treatment of cancer. Recently, targeting lipid metabolism has gained significant attention because of its roles not only in promoting cancer progression but also in inducing chemotherapy resistance. Fatty acid synthase (FAS) is the sole enzyme that is in charge of catalyzing the synthesis of palmitate, a long-chain lipid that is essential for membrane construction and post-translational modification in cell biology. Both FAS and its product, palmitate, have been validated as critical players in mediating or causing chemoresistance in c

Protectin DX Ameliorates Hepatic Steatosis by Suppression of Endoplasmic Reticulum Stress via AMPK-Induced ORP150 Expression.

Docosahexaenoic acid (DHA) and its bioactive compounds may have suppressive effects on inflammation, endoplasmic reticulum (ER) stress, and insulin resistance. Protectin DX (PDX), a double lipoxygenase product from DHA has shown a suppressive effect on inflammation and insulin resistance. However, the effects of PDX on ER stress and hepatic steatosis have not been elucidated yet. Herein we report that PDX could stimulate the AMP-activated protein kinase (AMPK) phosphorylation, thereby upregulating oxygen-regulated protein 150 (ORP150) expression in a dose-dependent manner. Treatment of HepG2 c

Transport of plasma FFA in cats; effect of hypothalamic stimulation.

Two tracer methods, repetitive single injection of [3-H]palmitate and continuous infusion of [1 minus 14-C]palmitate, were applied simultaneously to study tracer kinetics of the free fatty acid (FFA) miscible pool in anesthetized cats before, during, and after electrical stimulation of the hypothalamus. The results indicated that the FFA pool of the cat behaved as though it consisted of two compartments which exchange FFA with each other. The data were analyzed according to a two-compartmental mammillary system model. The space of the central compartment which represents the plasma was found t

Sustainable and affordable pest control: In silico study and toxicological effects of black soldier fly, Hermetia illucens (Diptera: Stratiomyidae), oil, and its nanoemulsion against Agrotis ipsilon (Lepidoptera: Noctuidae) larvae.

Black soldier fly, Hermetia illucens (Linnaeus, 1758) (Diptera: Stratiomyidae) larvae decompose several organic wastes into valuable biomass (oil, protein, chitin, and frass) for promising applications. Black Soldier Fly Oil (BSFO) is extracted using a mechanical pressing method, an affordable technique that yields about 24.56% oil. Gas chromatography-mass spectrometry analysis of oil showed a high content of fatty acids, with 3 predominant fatty acids, oleic acid (40.14%), palmitic acid (24.53%), and lauric acid (10.00%). Black soldier fly oil nanoemulsion (BSFO-NE) exhibited a droplet size o

Preconditioning and supplementation with glutathione ameliorates oxidative stress, mitochondrial dysfunction, and restores metabolic imbalance in insulin-resistant myotubes.

Insulin resistance, defined as the inability of the insulin-target tissues including skeletal muscles to insulin action, has been identified as a major pathophysiology associated with the development of metabolic disorders including type 2 diabetes mellitus. Palmitic acid (PA) and related saturated free fatty acids contribute significantly to the development of insulin resistance by perturbing the redox homeostasis in the cells. The deficiency of the intracellular antioxidant - glutathione (GSH) has been documented in insulin-resistant states, and dietary supplementation of GSH and its precurs

Improvement in the Extraction of Antioxidant-Related Compounds from Parastrephia quadrangularis ("tola") Using Ethanol-Modified Supercritical Carbon Dioxide.

Parastrephia quadrangularis (tola) is a native plant of the Chilean Andean Altiplano that is traditionally used for its anti-inflammatory properties. In this study, the aerial parts of the plant were analysed to determine their fatty acid (FA) profile and to identify bioactive compounds using gas chromatography-mass spectrometry (GC-MS). Both conventional extraction methods and Supercritical Fluid Extraction (SFE) were employed, using a 23 factorial design with centre-point replicates. The variables included temperature (30-60 °C), pressure (15-45 MPa), and ethanol as a cosolvent (0-30% v/v).
