ABC transporters in the CNS - an inventory.

In the present review we provide a summary of ATP-binding cassette (ABC) transporters in the central nervous system (CNS). Our review is focused on transporters of the ABC A, B, C, D, and G families that have been detected in the cells of the neurovascular unit/blood-brain barrier including brain capillary endothelial cells, pericytes, astrocytes, and neurons, as well as in other brain cells, such as microglia, oligodendrocytes, and choroid plexus epithelial cells. In this review, we provide an overview, organized by ABC family, of transporter expression, localization, and function. We summari…

Primary pulmonary hemangiopericytoma in a 3-year-old child.

Hemangiopericytoma is an uncommon and potentially malignant mesenchymal tumor that apparently arises from spindle-shaped pericytes. The tumor usually grows insidiously until considerable size has been reached, at which point symptoms may indicate invasion of the chest wall, mediastinum, or pleura. Because of its malignant potential, hemangiopericytoma must be distinguished from other vascular tumors. While this tumor has been reported in adolescents and adults, it has never before, to our knowledge, been reported in a small child. We present, within the context of a brief review, a case of app…

Localization of smooth muscle myosin-containing cells in the aqueous outflow pathway.

Cells containing smooth muscle myosin were localized in the human aqueous outflow pathway by immunohistochemical techniques. In the majority of eyes, immunoreactive cells were observed adjacent to the collector channels and slightly distal to the outer wall of Schlemm's canal. In a few eyes, smooth muscle myosin was localized to cells in the juxtacanalicular tissue and the trabecular meshwork. The immunoreactive cells from these regions may be true smooth muscle cells or pericytes, which can contain smooth muscle myosin. No obvious differences were observed in the pattern of distribution of sm…

Microprobe analysis of chlorpromazine pigmentation.

We describe the histochemical, ultrastructural, and microanalytical features of a skin biopsy specimen obtained from a patient with chlorpromazine pigmentation. Golden-brown pigment granules were present in the dermis, predominantly in a perivascular arrangement. The granules stained positively with the Fontana-Masson stain for silver-reducing substances and negatively with Perl's stain for iron. Electron microscopy revealed dense inclusion bodies in dermal histiocytes, pericytes, endothelial cells, and Schwann cells, as well as lying free in the extracellular matrix. These "chlorpromazine bod…

Subretinal neovascularization after experimental ocular histoplasmosis in a subhuman primate.

Subretinal neovascularization was demonstrated in a subhuman primate eye (Macaca speciosa) one year after the internal carotid injection of … to be continuous, had tight junctional complexes, and a well-developed basal lamina with occasional pericytes. Failure of fluorescein angiography to demonstrate the presence of this neovascular net may be explained by the presence of tight junctions within the subretinal capillaries.

EXPRESS: Microvascular no-reflow after endovascular therapy: The molecular ceiling on hemodynamic optimization and the next therapeutic frontier.

Endovascular thrombectomy restores macrovascular patency but fails to achieve tissue-level perfusion in approximately 30% of cases, likely because of the no-reflow phenomenon. Even after optimized hemodynamic management, 40% of patients with angiographically successful recanalization fail to achieve functional independence, revealing a biological ceiling beyond blood pressure optimization. I propose that pericyte contraction via endothelin-1/ET-A receptor signaling, neutrophil CD11b/CD18-dependent capillary plugging, and platelet microthrombus, mechanisms operating orthogonally to systemic hem

A placental chorionic villous mesenchymal core cellular origin for infantile haemangioma.

AIMS: To investigate the expression of the placental cell-specific associated proteins in infantile haemangioma (IH). METHODS: Immunohistochemical staining was used to investigate the expression of human chorionic gonadotrophin (hCG), human placental lactogen (hPL), human leucocyte antigen-G (HLA-G), cytokeratin 7 (CK7) and smooth muscle actin in paraffin-embedded sections of proliferating and involuted IHs. RESULTS: The proteins hCG and hPL were expressed by the endothelium but not the pericyte layer of proliferating IH, but these proteins were not detected in involuted lesions. There was n

Sinonasal haemangiopericytoma: a case report.

Haemangiopericytoma (HPC) is a rare vascular tumour that is thought to originate from the vascular pericytes of Zimmerman. HPC may arise in any part of the body, and from 15 to 30% of these tumours are found in the head and neck, with a rare involvement of the sinonasal region The main symptoms of nasal HPC, epistaxis and nasal obstruction, are not typical. The final diagnosis is based on the histopathology and immunochemistry, and whether the tumour is benign or malignant is defined on the basis of the clinical history. HPC located in the sinonasal area is generally benign. We report the case…

In Vivo Profiling of the Vascular Cell Surface Proteome in Murine Models of Bacteremia.

Vascular dysfunction is a hallmark of systemic inflammatory responses such as bacterial sepsis. The luminal surface of the blood vessels is coated with a dense layer of glycans and proteoglycans, collectively known as the glycocalyx. Surface associated glycoproteins of endothelial origin, or derived from pericytes, intravascular leukocytes, and plasma, are other important components of the glycocalyx, constituting a vascular cell surface proteome that is dynamic, tissue-specific, and sensitive to changes in vascular homeostasis, blood infection, and inflammation. Here, we describe an experimen…

Molecular parallels between neural and vascular development.

The human central nervous system (CNS) features a network of ~400 miles of blood vessels that receives >20% of the body's cardiac output and uses most of its blood glucose. Many human diseases, including stroke, retinopathy, and cancer, are associated with the biology of CNS blood vessels. These vessels originate from extrinsic cell populations, including endothelial cells and pericytes that colonize the CNS and interact with glia and neurons to establish the blood-brain barrier and control cerebrovascular exchanges. Neurovascular interactions also play important roles in adult neurogenic nich…

Intranasal myopericytoma. A tumour with perivascular myoid differentiation: the changing nomenclature for haemangiopericytoma.

We present a case report of a patient who developed a sinonasal myopericytoma treated by surgical excision through a lateral rhinotomy. Some … s constitute a single morphological spectrum with differentiation towards perivascular myoid cells (pericytes). Currently myopericytoma is the most appropriate and accepted term embracing all these entities. A review of the literature has been reassuring in identifying these tumours as benign but with a reasonably high rate of local recurrence (17 per cent) …

Giant hemangiopericytoma of mandible: a propos of a case: a variant of the surgical technique for protection of the articular fosse.

The hemangiopericytoma is a tumor of the pericytes. It represents 1% of vascular tumors. It is frequently located in inferior limbs, the pelvis, and the retroperitoneum. A painless mass of slow growth is main presentation. Imaging is not specific. Only microscopic and immunohistochemical findings can verify the lesion. The surgical treatment is elective. The search of the world medical literature revealed the publication of only four cases of hemangiopericytoma of the jaw. We present the case of a patient with a giant hemangiopericytoma that originated in the jaw. Studies showed an enormous ma…

Bacillary angiomatosis of the conjunctiva.

A 70-year-old man had unilateral congestion of the right upper eyelid, which contained a nodular mass. A biopsy was performed, and histologic, immunocytochemical, and ultrastructural studies disclosed a pseudoneoplastic proliferation of endothelial cells and pericytes in a region containing clumps of bacteria. This combination of histologic features is characteristic of bacillary angiomatosis, which has been described in the skin, particularly in association with immunodeficient states, especially acquired immunodeficiency syndrome, but not in the conjunctiva. A second biopsy contained a diffu…

An investigation into the role of inflammatory cells in Dupuytren's disease.

An immunohistochemical study was performed on nodules excised from the palmar fascia of patients with Dupuytren's contracture. In cellular nodules, antibodies to actin (used as a marker for myofibroblasts), desmin, vimentin, Mac 387 (a macrophage marker) and leucocyte common antigen were used. A correlation was demonstrated between the numbers of macrophages and the presence of myofibroblasts. The presence of myofibroblasts is generally considered to indicate the active stage of the disease. Inflammatory cells other than macrophages were largely absent from the nodules, although lymphocytes we
