Shape, form, function and Leishmania pathogenicity: from textbook descriptions to biological understanding.

The shape and form of protozoan parasites are inextricably linked to their pathogenicity. The evolutionary pressure associated with establishing and maintaining an infection and transmission to vector or host has shaped parasite morphology. However, there is not a 'one size fits all' morphological solution to these different pressures, and parasites exhibit a range of different morphologies, reflecting the diversity of their complex life cycles. In this review, we will focus on the shape and form of Leishmania spp., a group of very successful protozoan parasites that cause a range of diseases

Coarse-resolution Ecology of Etiological Agent, Vector, and Reservoirs of Zoonotic Cutaneous Leishmaniasis in Libya.

Cutaneous leishmaniasis ranks among the tropical diseases least known and most neglected in Libya. World Health Organization reports recognized associations of Phlebotomus papatasi, Psammomys obesus, and Meriones spp., with transmission of zoonotic cutaneous leishmaniasis (ZCL; caused by Leishmania major) across Libya. Here, we map risk of ZCL infection based on occurrence records of L. major, P. papatasi, and four potential animal reservoirs (Meriones libycus, Meriones shawi, Psammomys obesus, and Gerbillus gerbillus). Ecological niche models identified limited risk areas for ZCL across the n…

Structure of a phleboviral envelope glycoprotein reveals a consolidated model of membrane fusion.

An emergent viral pathogen termed severe fever with thrombocytopenia syndrome virus (SFTSV) is responsible for thousands of clinical cases and associated fatalities in China, Japan, and South Korea. Akin to other phleboviruses, SFTSV relies on a viral glycoprotein, Gc, to catalyze the merger of endosomal host and viral membranes during cell entry. Here, we describe the postfusion structure of SFTSV Gc, revealing that the molecular transformations the phleboviral Gc undergoes upon host cell entry are conserved with otherwise unrelated alpha- and flaviviruses. By comparison of SFTSV Gc with that

Spatial distribution of P. argentipes in association with agricultural surrounding environment in North Bihar, India.

INTRODUCTION: Phlebotomus argentipes is considered to be one of the major control measure targets in eradicating visceral leishmaniasis (VL). This study demonstrates the spatial association of P. argentipes abundance in relation to agricultural environment in the endemic area of north Bihar. METHODOLOGY: Vector data were collected from the 95 villages of the study area using a handheld aspiration technique. Space technology and ground observationwere made to estimate the environmental characteristics. RESULTS: A total of 1,663 P. argentipes were collected between April and November 2011. Res…

Zoonotic disease in a peripheral population: persistence and transmission of Leishmania major in a putative sink-source system in the Negev Highlands, Israel.

Populations at the edge of their geographic distributions are referred to as peripheral populations. Very little attention has been given to this topic in the context of persistence of infectious disease in natural populations. In this study, we examined this question using zoonotic cutaneous leishmaniasis (ZCL) caused by Leishmania major in the Negev Desert of Israel as a model system. Here, we suggest that the regional persistence of Phlebotomus papatasi populations and L. major transmission in the Sede Boqer region could be explained through processes akin to sink-source and/or mainland-isl…

Quantifying the contribution of hosts with different parasite concentrations to the transmission of visceral leishmaniasis in Ethiopia.

BACKGROUND: An important factor influencing the transmission dynamics of vector-borne diseases is the contribution of hosts with different p … ntified the host parasitemias that are mostly responsible for the infection of vector, the sand fly Phlebotomus orientalis. Our findings indicate that a 3.2% of the most infected people were responsible for the infection of between 53% and 79% (mean - 62%) of the infected sand fly vector population. SIGNIFICANCE: Our modeling framework can easily be extended …

Undifferentiated febrile illnesses amongst British troops in Helmand, Afghanistan.

OBJECTIVES: Undifferentiated febrile illnesses have been a threat to British expeditionary forces ever since the Crusades. The infections responsible were identified during the Colonial Era, both World Wars and smaller conflicts since, but nearly all remain a significant threat today. Undiagnosed febrile illnesses have occurred amongst British troops in Helmand, Afghanistan since 2006 and so a fever study was performed to identify them. METHODS: From May to October 2008, all undifferentiated fever cases seen at the British field hospital in Helmand, Afghanistan were assessed using a standard

Unusual presentation of life-threatening Toscana virus meningoencephalitis.

This case report describes a brother and a sister with severe meningoencephalitis caused by Toscana virus (TOSv). The clinical presentation was characterized by stiff neck, deep coma, maculopapular rash, diffuse lymphadenopathy, hepatosplenomegaly, renal involvement, tendency to bleeding, and diffuse intravascular coagulation. The boy had epididymo-orchitis. Recovery with neurologic sequelae as hydrocephalus was observed. Microbiological diagnosis was obtained by serological tests and reverse transcriptase-polymerase chain reaction. Sequencing of polymerase chain reaction products from the S a

[Geographical variability of the causative agent of zoonotic cutaneous leishmaniasis].

A study of the phenetics of the zoonotic cutaneous leishmaniasis agent in Uzbekistan and subzone of southern and northern deserts has made it possible to separate a northern population (or a group of populations) of the agent. It differs from the southern population (or southern ones) in having smaller sizes of the amastigote stage, greater abundance of parasites in cutaneous affections of gerbils, milder progress of leishmaniasis in white mice and great gerbils. This population can be called priaral, i.e. belonging to the subzone of northern deserts of the Turan lowland.

Exploring Emerging Challenges: Survey on Phlebotomine Sand Flies and Leishmania infantum at the Northern Endemic Border in Europe.

Although Switzerland is currently not endemic for canine leishmaniosis (CanL), imported cases of this emerging zoonosis are regularly detect … ught there. In Ticino, 15 out of 20 sites surveyed (75%) were positive for phlebotomine sand flies. Phlebotomus perniciosus, a potential vector of L. infantum, was the most abundant species, with site densities ranging from 0.1 to 5.3. The parasite was not detected in females. Leishmania infantum seroprevalences of 3.0% and 1.6% were observed in 101 and 126 d …

Development of a mouse model for the study of Toscana virus pathogenesis.

Toscana virus (TOSV) has recently been recognized as an emerging virus transmitted by phlebotomus vectors, responsible for acute neurological diseases in Mediterranean countries. In our study, we demonstrated that adult Balb/c mice were susceptible to TOSV when infected intracerebrally (i.c.) or subcutaneously (s.c.) with a neuroadapted strain of the virus. We have shown that by performing serial passages of a wild type human isolate of TOSV in mouse brains, selection occurs for a highly virulent variant which replicates efficiently in the central nervous system (CNS) of i.c.-injected mice, ca…
