Chronodisruption that dampens output of the central clock abolishes rhythms in metabolome profiles and elevates acylcarnitine levels in the liver of female rats.

AIM: Exposure to light at night and meal time misaligned with the light/dark (LD) cycle-typical features of daily life in modern 24/7 society-are associated with negative effects on health. To understand the mechanism, we developed a novel protocol of complex chronodisruption (CD) in which we exposed female rats to four weekly cycles consisting of 5-day intervals of constant light and 2-day intervals of food access restricted to the light phase of the 12:12 LD cycle. METHODS: We examined the effects of CD on behavior, estrous cycle, sleep patterns, glucose homeostasis and profiles of clock- a

Exercise as a Synchronizer: Effects on Circadian Re-Entrainment of Core Body Temperature and Metabolism Following Light-Dark Cycle Inversion in Mice.

Core body temperature (CBT) is a crucial marker of circadian synchrony, reflecting behavioral, metabolic, and environmental adaptations. Disruptions to CBT rhythms, as seen in shift workers or jetlag, indicate desynchronization and can lead to significant health consequences. Exercise is a potent non-photic zeitgeber that may help align circadian rhythms with external cues, but its role in re-entrainment following abrupt phase shifts remains unclear. This study investigated whether voluntary exercise accelerates the re-entrainment of CBT and metabolic rhythms in mice subjected to a 12-h light-

Artificial light at night disrupts fertility in Drosophila melanogaster.

Artificial light at night (ALAN) can disrupt numerous biological processes, and is increasingly studied in animal models. Here, we evaluated the impact of red and blue ALAN on Drosophila melanogaster, focusing on fertility, development, circadian rhythms, and gene expression. All results were compared to those of a control group maintained under a 12 h white light/12 h dark cycle. Red ALAN exposure increased the number of eggs laid but reduced the hatching rate and shortened the larval period. Conversely, blue ALAN led to fewer eggs laid, fewer emerging adults, and lower hatching success. Sign

Monochromatic Photophase Light Alters Diurnal Profiles of Melatonin Pathway Indoles in the Rat Pineal Gland.

Light is a major environmental factor that regulates circadian rhythms and pineal melatonin synthesis. While the influence of nighttime light exposure on melatonin suppression has been extensively investigated, much less is known about the impact of photophase light wavelength on pineal function. The aim of the study was to determine the influence of monochromatic light during the photophase on diurnal changes in melatonin-related indoles in the rat pineal gland. Wistar rats were exposed for 7 days to 150 lx of monochromatic blue (463 ± 10 nm), green (523 ± 10 nm), or red (623 ± 10 nm) LED lig

Altered circadian rhythm, sleep, and rhodopsin 7-dependent shade preference during diapause in Drosophila melanogaster.

To survive adverse environments, many animals enter a dormant state such as hibernation, dauer, or diapause. Various Drosophila species unde … peak at a time similar to when nondiapausing flies take a siesta. Temperatures ≤15 °C, rather than photoperiod, primarily drive this behavior. At cool temperatures, flies rapidly enter a deep-sleep state that lacks the sleep cycles of flies at higher temperatures and require high levels of stimulation for arousal. Furthermore, we show that at 25 °C, flies pr …

The male effect associated with prostaglandins and reproductive outcomes in photo-stimulated Saanen goats during the non-breeding season.

The efficiency of combining oestrous induction via a light program (16 h of light and 8 h of darkness for 60 days, ending on Day 0 - D0) with cloprostenol administration, followed by the male effect or not, was tested in acyclic Saanen goats during the non-breeding season (June/2019 to January/2020). Initially, all animals (males and females) were submitted to the described light program; 60 days after its ending (D60), the females were divided into two groups, with (G1; n = 67) or without (G2; n = 61) a male effect from D60 to D75 after the light program. At D75, both groups received two clop

Evidence for Internal Desynchrony Caused by Circadian Clock Resetting.

Circadian disruption has been linked to markers for poor health outcomes in humans and animal models. What is it about circadian disruption that is problematic? One hypothesis is that phase resetting of the circadian system, which occurs in response to changes in environmental timing cues, leads to internal desynchrony within the organism. Internal desynchrony is understood as acute changes in phase relationships between biological rhythms from different cell groups, tissues, or organs within the body. Do we have strong evidence for internal desynchrony associated with or caused by circadian c

Seasonal variation of BMI at admission in German adolescents with anorexia nervosa.

OBJECTIVE: Recent preliminary studies indicated a seasonal association of BMI at admission to inpatient treatment for anorexia nervosa (AN), indicating lower BMI in the cold season for restrictive AN. An impaired thermoregulation was proposed as the causal factor, based on findings in animal models of AN. However, findings regarding seasonality of BMI and physical activity levels in the general population indicate lower BMI and higher physical activity in summer than in winter. Therefore, we aimed to thoroughly replicate the findings regarding seasonality of BMI at admission in patients with A

Time-of-day influences on respiratory sequelae following maximal electroshock-induced seizures in mice.

Sudden unexpected death in epilepsy (SUDEP) is the leading cause of death in refractory epilepsy patients. Although specific mechanisms underlying SUDEP are not well understood, evidence suggests most SUDEP occurs due to seizure-induced respiratory arrest. SUDEP also tends to happen at night. Although this may be due to circumstances in which humans find themselves at night, such as being alone without supervision or sleeping prone, or to independent influences of sleep state, there are a number of reasons why the night (i.e., circadian influences) could be an independent risk factor for SUDEP

Diurnal physiological and immunological responses to a 10-km run in highly trained athletes in an environmentally controlled condition of 6 °C.

PURPOSE: The Clara cell protein CC16, secreted from Clara cells in the lung, is discussed as a potential biomarker for toxic effects on the airways. An increased concentration of CC16 in serum may be caused by increased permeability of the lungs. To investigate the changes in P-CC16 in response to an intense exercise bout performed at different times of day (9 am and 4 pm) of highly trained individuals. METHOD: Using a crossover randomized design, 8 runners (mean VO2max 71 ml kg-1 min-1, SD 6) performed a 10-km time trial run, at 9 am and 4 pm, in an environmental chamber set at 6 °C. Lung fu

Photic and circadian regulation of melatonin production in the Mozambique tilapia Oreochromis mossambicus.

Diverse circadian systems related to phylogeny and ecological adaptive strategies are proposed in teleosts. Recently, retinal photoreception was reported to be important for the circadian pacemaking activities of the Nile tilapia Oreochromis niloticus. We aimed to confirm the photic and circadian responsiveness of its close relative-the Mozambique tilapia O. mossambicus. Melatonin production in cannulated or ophthalmectomized fish and its secretion from cultured pineal glands were examined under several light regimes. Melatonin production in the cannulated tilapias was measured at 3-h interval

Endogenous alpha-ketol linolenic acid levels in short day-induced cotyledons are closely related to flower induction in Pharbitis nil.

Alpha-ketol linolenic acid [KODA, 9,10-ketol-octadecadienoic acid, that is 9-hydroxy-10-oxo-12(Z),15(Z)-octadecadienoic acid] is a signal compound found in Lemna paucicostata after exposure to stress, such as drought, heat or osmotic stress. KODA reacts with catecholamines to generate products that strongly induce flowering, although KODA itself is inactive [Yokoyama et al. (2000) Plant Cell Physiol. 41: 110; Yamaguchi et al. (2001) Plant Cell Physiol. 42: 1201]. We examined the role of KODA in the flower-induction process of Pharbitis nil (violet). KODA was identified for the first time in se

Phase advance after one or three simulated dawns in humans.

A specially designed apparatus that can simulate the waveform of the dawn or dusk signal at any latitude and any day of the year has been shown to phase shift the circadian pacemaker in rodents and primates at a fraction of the illuminance previously used. Until recently, it was considered that rather high illuminances or rather long exposure episodes to room light were necessary to phase shift human circadian rhythms. This experiment shows that, under controlled conditions of a modified constant routine protocol, a single dawn signal is sufficient to phase advance the timing of the onset of s

Individual pineal cells in chick possess photoreceptive, circadian clock and melatonin-synthesizing capacities in vitro.

Chick pineal cells express a circadian rhythm of melatonin release under light-dark (LD) cycles, with an increase during the dark period and a decrease during the light period, and this rhythm persists under constant darkness (DD). We cultured individual single pineal cells with 15 microl of medium per well in a Terasaki plate and measured melatonin secretion every 12 h under LD, DL and DD. Individual cells secreted more melatonin during the dark period than during the light period under both LD and DL conditions, and those rhythmic secretions persisted under DD. These results suggest that ind

Rhythm in the vertical distribution of Danio rerio under photic and magnetic zeitgebers.

Circadian rhythms in the vertical distribution of adult zebrafish and their potential sensitivity to geomagnetic fluctuations remain poorly … fluctuations may function as an extremely weak zeitgeber compared to the dominant influence of the photoperiod. Prior exposure to an atypical light-dark regime, which may have reset the biochemical processes sustaining the endogenous oscillators, appears to be required for the manifestation of magnetically driven rhythm entrainment.

Daytime aversive stimuli do not phase shift behavioral rhythms under light-dark cycles in a strictly diurnal rodent.

Recent studies have shown that cyclic aversive stimuli (time-specific footshocks) act as a nonphotic zeitgeber, shifting circadian behaviors to the daytime in nocturnal rodents through entrainment. It has remained untested whether diurnal species exhibit similar plasticity in behavioral timing. This study investigated whether antelope ground squirrels (Ammospermophilus leucurus, AGS), naturally diurnal rodents, shift activity timing in response to cyclic aversive stimuli delivered at specific phases of the light-dark (LD) cycle. We conducted two experiments with 20 AGS housed in custom cages f

Circadian clock period length is not consistently linked to chronotype in a wild songbird.

Circadian clock properties vary between individuals and relate to variation in entrained timing in captivity. How this variation translates into behavioural differences in natural settings, however, is poorly understood. Here, we tested in great tits whether variation in the free-running period length (tau) under constant dim light (LL) was linked to the phase angle of the entrained rhythm ("chronotype") in captivity and in the wild, as recently indicated in our study species. We also assessed links between tau and the timing of first activity onset and offset under LL relative to the last exp
