Molecular Mechanisms Underlying Flax (Linum usitatissimum L.) Tolerance to Cadmium: A Case Study of Proteome and Metabolome of Four Different Flax Genotypes.

Cadmium is one of the most toxic heavy metal pollutants, and its accumulation in the soil is harmful to agriculture. Plants have a higher cadmium tolerance than animals, and some species can be used for phytoremediation. Flax (Linum usitatissimum L.) can accumulate high amounts of cadmium, but the molecular mechanism behind its tolerance is unknown. Here, we employed four genotypes representing two fiber cultivars, an oilseed breeding line, and a transgenic line overexpressing the metallothionein domain for improved cadmium tolerance. We analyzed the proteome of suspensions and the proteome an

Genetic and agronomic approaches to decreasing acrylamide precursors in crop plants.

Progress in developing genetic and agronomic approaches for reducing the levels of the principal precursors of acrylamide, asparagine and sugars in crop plants is reviewed. The factors that affect asparagine and sugar accumulation, particularly in cereal seeds and potato tubers, are described. Asparagine levels appear to be the key parameter in determining acrylamide formation in processed wheat flour and agronomic strategies for reducing asparagine accumulation in wheat grain are reviewed. Sulphur availability has been shown to be particularly important, with sulphur deprivation causing a dra

To buzz or to sip: Asymmetric pollination effectiveness hints at the evolutionary transition of floral rewards.

BACKGROUND AND AIMS: Evolutionary transitions between floral reward types, such as the repeated loss of nectar, are common across angiosperms, yet the ecological mechanisms driving these shifts remain poorly understood. In Pedicularis, the evolutionary loss of nectar is hypothesized to be associated with shifts to buzz pollination. However, empirical comparisons of pollination effectiveness between nectar- and pollen-foraging behaviours within a single transitional system are largely lacking. METHODS: We investigated Pedicularis alaschanica and its principal pollinator, Bombus friseanus, in t

Multiple environmental test and genetic mapping jointly reveal genomic loci and associated genes underlying cotton Verticillium wilt resistance.

Based on GWAS, QTL mapping, BSA and RNA‒seq jointly reveal the genetic mechanisms underlying Verticillium wilt resistance in cotton and identify three candidate genes associated with this trait. Verticillium wilt, caused by the soil-borne vascular pathogen Verticillium dahliae, is a devastating disease that severely threatens global cotton production. However, the genetic basis of resistance to this disease remains poorly uncovered, hindering the advancement of genomics-assisted breeding programs for Verticillium wilt (VW) resistance. Here, we systematically evaluated VW resistance in a recomb

Characterization of the sexual reproductive process in the perennial tree Robinia pseudoacacia.

Sexual reproduction in perennial plants, especially woody species, remains poorly understood, and most studies have focused on annuals such as Arabidopsis thaliana. This gap hinders understanding of perennial reproductive processes and limits tree breeding. Here we characterized the sexual reproductive process of Robinia pseudoacacia, a perennial leguminous tree, and established standardized methods for studying its reproduction. We optimized cytological assays for pollen viability, in vitro germination, and pollen tube dynamics, achieving over 90% pollen viability and germination efficiency w

Redox-active di-O-methylated coumarins exudation contributes to genotype-dependent iron deficiency tolerance in soybean.

Iron (Fe) deficiency is a widespread disorder limiting global soybean (Glycine max (L.) Merr.) production. Although root exudation is a key adaptive mechanism for Fe scarcity in species like Arabidopsis, a detailed chemical characterization of soybean exudates is lacking. Here, we examined the accumulation and secretion of phenolic compounds in soybean roots and their correlation with intraspecific tolerance to Fe-deficiency chlorosis. Seven soybean genotypes with contrasting tolerance, derived from U.S. breeding programs, were analyzed. Root exudates from Fe-deficient soybean plants solubiliz

Analysis of the Nutritional Composition and Drought Tolerance Traits of Sweet Potato: Selection Criteria for Breeding Lines.

Sweet potato is an important world staple with the potential to address hunger and malnutrition. The Agricultural Research Council of South Africa has led sweet potato breeding efforts in the country since 1952 and released several important cultivars. More detailed quality assessments are necessary in addition to general breeding criteria. The present study investigated sweet potato cultivars and elite clones for (1) their nutritional composition; (2) biochemical reaction to drought stress; (3) correlate biochemical reaction to root yield for possible identification of screening methods for d

Examination of Genomic and Transcriptomic Alterations in a Morphologically Stable Line, MU1, Generated by Intergeneric Pollination.

Interspecific hybridization creates genetic variation useful for crop improvement. However, whether pollen from a different genus affects the genomic stability and/or transcriptome of the recipient species during intergeneric pollination has not been investigated. Here, we crossed japonica rice cv. Z12 with the maize accession B73 (pollen donor) and obtained a morphologically stable line, MU1, exhibiting moderate dwarfism, higher tiller number, and increased grain weight compared with Z12. To reveal the genetic basis of these morphological changes in MU1, we performed whole-genome resequencing

Reducing recalcitrance of black pepper to Agrobacterium-mediated transformation: an efficient way through nucellar apomixis to establish transgenic and genome-edited plants at high frequency and scale-up through bioreactor.

Nucellar apomixis is truly clonal and is a powerful tool for broadening the genetic base of crops. Black pepper (Piper nigrum L.), the 'King of Spices' is difficult to improve through conventional breeding. Although transgenesis and genome editing are prime strategies for rapid crop improvement, recalcitrance hinders genetic modifications. Here, we report a highly efficient Agrobacterium-mediated procedure for generating genetically modified black pepper plants using nucellar apomixis-derived embryos of the varieties Sreekara and Karimunda, with a 99% survival rate. Both Agrobacterium tumefaci

Integration of targeted metabolome and transcript profiling of Pseudomonas syringae-triggered changes in defence-related phytochemicals in oat plants.

A gene-to-metabolite approach afforded new insights regarding defence mechanisms in oat plants that can be incorporated into plant breeding programmes for the selection of markers and genes related to disease resistance. Monitoring metabolite levels and changes therein can complement and corroborate transcriptome (mRNA) data on plant-pathogen interactions, thus revealing mechanisms involved in pathogen attack and host defence. A multi-omics approach thus adds new layers of information such as identifying metabolites with antimicrobial properties, elucidating metabolomic profiles of infected an…

Elicitation of jasmonate-mediated host defense in Brassica juncea (L.) attenuates population growth of mustard aphid Lipaphis erysimi (Kalt.).

The productivity of Brassica oilseeds is severely affected by its major pest: aphids. Unavailability of resistance source within the crossable germplasms has stalled the breeding efforts to derive aphid resistant cultivars. In this study, jasmonate-mediated host defense in Indian mustard Brassica juncea (L.) Czern. was evaluated and compared with regard to its elicitation in response to mustard aphid Lipaphis erysimi (Kalt.) and the defense elicitor methyl jasmonate (MeJ). Identification of jasmonate-induced unigenes in B. juncea revealed that most are orthologous to aphid-responsive genes, id

Relationship between cyanogenic compounds in kernels, leaves, and roots of sweet and bitter kernelled almonds.

The relationship between the levels of cyanogenic compounds (amygdalin and prunasin) in kernels, leaves, and roots of 5 sweet-, 5 slightly bitter-, and 5 bitter-kernelled almond trees was determined. Variability was observed among the genotypes for these compounds. Prunasin was found only in the vegetative part (roots and leaves) for all genotypes tested. Amygdalin was detected only in the kernels, mainly in bitter genotypes. In general, bitter-kernelled genotypes had higher levels of prunasin in their roots than nonbitter ones, but the correlation between cyanogenic compounds in the different

Twenty-one years of three-way hybrid breeding shaped diversity and complementarity of male and female gene pools in sugar beet.

Three-way hybrid breeding in sugar beet is associated with strong genomic differentiation between male and female parental pools and persistent haplotypic differentiation at agronomically relevant loci over the last two decades. Three-way hybrid breeding has been used in several major crops to increase seed yield and manage heterosis. Sugar beet (Beta vulgaris ssp. vulgaris) breeding relies exclusively on it and combines cytoplasmic male sterility (CMS) and monogermity to exploit heterosis by combining male and female parental pools. The genomic consequences of this breeding scheme remain poor

Genome-wide identification and functional analysis of the BES1-like (VfBES1) gene family in Vernicia fordii reveals its role in floral development.

BACKGROUND: Vernicia fordii Hemsl (also known as Tung tree), an significant commercial oil-producing tree species, is a monoecious and diclinous species with male and female flowers on the same inflorescence; however, the molecular mechanisms governing its floral sex determination remain elusive, particularly the genetic basis underlying the skewed female-to-male flower ratio and the evolutionary dynamics of sex-related gene families, which severely restrict targeted breeding for yield enhancement. In the model plant Arabidopsis, the BRI1 EMS SUPPRESSOR 1 (BES1) transcription factor family pla

Elevated serum CA72-4 levels predict poor prognosis in pancreatic adenocarcinoma after intensity-modulated radiation therapy.

UNLABELLED: Purple blotch (PB), caused by Alternaria porri (Ellis) Cifferi, is one of the most destructive diseases of onion worldwide. Rapid development and deployment of resistant onion varieties is the most effective approach to control this disease. A single dominant gene, ApR1 was previously linked to PB resistance in onion cultivar 'Arka Kalyan'. In this study, an advanced RIL population derived from a cross between the resistant (Arka Kalyan) and susceptible (Agrifound Rose) cultivar of onion was used to fine map the resistant locus with SNP markers. Twenty plants from the RIL populatio

Nucleotide diversity, natural variation, and evolution of Flexible culm-1 and Strong culm-2 lodging resistance genes in rice.

Lodging resistance is one of the vital traits in yield improvement and sustainability. Culm wall thickness, diameter, and strength are different traits that can govern the lodging resistance in rice. The genes SCM2 and FC1 have been isolated for culm thickness, strength, and flexibility, but their functional nucleotide variations were still unknown. We used a 13× deep sequence of 795 diverse genotypes to present the functional variation and SNP diversity in SCM2 and FC1. The major functional variant for the SCM2 gene was at position 27480181 and for the FC1 gene at position 31072992. Haplotype
