Crossover fungal pathogens: the biology and pathogenesis of fungi capable of crossing kingdoms to infect plants and humans.

The outbreak of fungal meningitis associated with contaminated methylprednisolone acetate has thrust the importance of fungal infections into the public consciousness. The predominant pathogen isolated from clinical specimens, Exserohilum rostratum (teleomorph: Setosphaeria rostrata), is a dematiaceous fungus that infects grasses and rarely humans. This outbreak highlights the potential for fungal pathogens to infect both plants and humans. Most crossover or trans-kingdom pathogens are soil saprophytes and include fungi in Ascomycota and Mucormycotina phyla. To establish infection, crossover f

Two decades of plant-based candidate vaccines: a review of the chimeric protein approaches.

Genetic engineering revolutionized the concept of traditional vaccines since subunit vaccines became reality. Additionally, over the past two decades plant-derived antigens have been studied as potential vaccines with several advantages, including low cost and convenient administration. More specifically, genetic fusions allowed the expression of fusion proteins carrying two or more components with the aim to elicit immune responses against different targets, including antigens from distinct pathogens or strains. This review aims to provide an update in the field of the production of plant-bas

EDS1 contributes to nonhost resistance of Arabidopsis thaliana against Erwinia amylovora.

Erwinia amylovora causes fire blight in rosaceous plants. In nonhost Arabidopsis thaliana, E. amylovora triggers necrotic symptoms associated with transient bacterial multiplication, suggesting either that A. thaliana lacks a susceptibility factor or that it actively restricts E. amylovora growth. Inhibiting plant protein synthesis at the time of infection led to an increase in necrosis and bacterial multiplication and reduced callose deposition, indicating that A. thaliana requires active protein synthesis to restrict E. amylovora growth. Analysis of the callose synthase-deficient pmr4-1 muta

Cyanide, a coproduct of plant hormone ethylene biosynthesis, contributes to the resistance of rice to blast fungus.

Rice (Oryza sativa) plants carrying the Pi-i resistance gene to blast fungus Magnaporthe oryzae restrict invaded fungus in infected tissue via hypersensitive reaction or response (HR), which is accompanied by rapid ethylene production and formation of small HR lesions. Ethylene biosynthesis has been implicated to be important for blast resistance; however, the individual roles of ethylene and cyanide, which are produced from the precursor 1-aminocyclopropane-1-carboxylic acid, remain unevaluated. In this study, we found that Pi-i-mediated resistance was compromised in transgenic rice lines, in

SARD1 and CBP60g redundantly regulate N-hydroxypipecolic acid- and salicylic acid-dependent systemic immunity but are dispensable for the activation of indolic defense metabolism.

We investigated the roles of the cognate, redundantly acting transcription factors SARD1 and CPB60g in the metabolic reprogramming of leaves of Arabidopsis thaliana suffering from bacterial infection or UV light exposure. Comprehensive metabolite analyses reveal that SARD1/CBP60g exert distinct influences on major stress-inducible metabolic pathways. The SARD1/CBP60g transcriptional node decisively boosts the biosynthesis of the systemic acquired resistance (SAR)-inducing metabolites N-hydroxypipecolic acid (NHP) and salicylic acid (SA), as well as their metabolism to major glucose conjugates.

Phosphoproteomic analysis uncovers phosphorylated proteins in response to salicylic acid and N-hydroxypipecolic acid in Arabidopsis.

BACKGROUND: The phytohormone salicylic acid (SA) serves as a crucial signaling molecule within the realm of plant immunity, playing an indispensable role in both local and systemic acquired resistance (SAR). N-hydroxypipecolic acid (NHP), a derivative of L-lysine, is integral to the induction of SAR. Recent investigations have illuminated the intricate manner in which NHP orchestrates the establishment of SAR in conjunction with the immune signal SA. METHODS AND RESULTS: To further explore the mechanisms governing the synergistic regulation of SAR by SA and NHP, we conducted an extensive phos…

Peach extract induces systemic and local immune responses in an experimental food allergy model.

Peach allergy is among the most frequent food allergies in the Mediterranean area, often eliciting severe anaphylactic reactions in patients. Due to the risk of severe symptoms, studies in humans are limited, leading to a lack of therapeutic options. This study aimed to develop a peach allergy mouse model as a tool to better understand the pathomechanism and to allow preclinical investigations on the development of optimized strategies for immunotherapy. CBA/J mice were sensitized intraperitoneally with peach extract or PBS, using alum as adjuvant. Afterwards, extract was administered intragas

Novel transgenic rice-based vaccines.

Oral vaccination can induce both systemic and mucosal antigen-specific immune responses. To control rampant mucosal infectious diseases, the development of new effective oral vaccines is needed. Plant-based vaccines are new candidates for oral vaccines, and have some advantages over the traditional vaccines in cost, safety, and scalability. Rice seeds are attractive for vaccine production because of their stability and resistance to digestion in the stomach. The efficacy of some rice-based vaccines for infectious, autoimmune, and other diseases has been already demonstrated in animal models. W

Endogenous peptide defense signals in Arabidopsis differentially amplify signaling for the innate immune response.

AtPep1, a 23-aa peptide encoded by Arabidopsis PROPEP1, a member of a small, six-member gene family, activates expression of the defense gene PDF1.2 (encoding defensin) and its own precursor gene, PROPEP1, through the jasmonate/ethylene signaling pathway, mediated by a cell-surface receptor, PEPR1. Overexpression of two family members, PROPEP1 and PROPEP2, enhances resistance of Arabidopsis plants against the pathogen Pythium irregulare, and PROPEP2 and PROPEP3 are expressed at highly elevated levels in Arabidopsis in response to pathogen infections and to several pathogen-associated molecules

Ecological risks of microplastics in soils of major Angelica sinensis production areas and biological responses of roots to polyethylene microplastics.

Farmland microplastic pollution has become an emerging environmental issue in agricultural ecosystems; however, systematic studies on soil microplastic contamination in the main production areas of Angelica sinensis in China and its potential effects on medicinal roots remain limited. This study analyzed the composition, ecological risks, and accumulation and biological effects of microplastics in A. sinensis roots through field investigation combined with controlled experiments. The results indicate that farmland soils in the main production areas of Gansu Province contain relatively high lev

Soil microbial legacy mediated by buckwheat flavonoids enhances cabbage resistance to clubroot disease.

BACKGROUND: The legacy of plant growth significantly impacts the health of subsequent plants, yet the mechanisms by which soil legacies in crop rotation systems influence disease resistance through rhizosphere plant-microbiome interactions remain unclear. Using a buckwheat-cabbage rotation model, we investigated how microbial soil legacies shape cabbage growth and clubroot disease resistance. RESULTS: Three-year field trials revealed that buckwheat rotation sustainably reduced clubroot severity by 67%-97%, regardless of pathogen load. Soil sterilization eliminated this suppression, implicatin

Multi-omics analyses reveal the biocontrol potential of endophytic Paenibacillus peoriae 3-B4 against maize seedling blight.

INTRODUCTION: Maize seedling blight, caused by the phytopathogenic fungus Fusarium verticillioides, is a common and rapidly spreading disease that reduces grain quality and yield. Control of this pathogen is challenging due to its complex infection process and the development of resistance to chemical pesticides. Biological control agents are therefore recognized as a sustainable and environmentally friendly strategy for the maize seedling blight. RESULTS: In this study, we isolated and identified Paenibacillus peoriae 3-B4 from maize leaves, which exhibited a 59.92% inhibitory rate against F

Ectopic expression of BOTRYTIS SUSCEPTIBLE1 reveals its function as a positive regulator of wound-induced cell death and plant susceptibility to Botrytis.

Programmed cell death (PCD) is integral to plant life and required for stress responses, immunity, and development. Our understanding of the regulation of PCD is incomplete, especially concerning regulators involved in multiple divergent processes. The botrytis-susceptible (bos1) mutant of Arabidopsis is highly susceptible to fungal infection by Botrytis cinerea (Botrytis). BOS1 (also known as MYB108) regulates cell death propagation during plant responses to wounding. The bos1-1 allele contains a T-DNA insertion in the 5'-untranslated region upstream of the start codon. This insertion results

Characterization of the interaction between Oidium heveae and Arabidopsis thaliana.

Oidium heveae, an obligate biotrophic pathogen of rubber trees (Hevea brasiliensis), causes significant yield losses of rubber worldwide. However, the molecular mechanisms underlying the interplay between O. heveae and rubber trees remain largely unknown. In this study, we isolated an O. heveae strain, named HN1106, from cultivated H. brasiliensis in Hainan, China. We found that O. heveae HN1106 triggers the hypersensitive response in a manner that depends on the effector-triggered immunity proteins EDS1 (Enhanced Disease Susceptibility 1) and PAD4 (Phytoalexin Deficient 4) and on salicylic ac
