Primate Swallowing Is Powered by Both Rotation and Contraction of Suprahyoid Muscles.

OBJECTIVES: Swallowing biomechanics in primates and other mammals is poorly understood, and the effect of hyoid descent on swallowing biomechanics lacks experimental interrogation. In macaques, which share similar swallowing kinematics with humans, the base of the tongue and the food bolus are hypothesized to be driven into the oropharynx by a hydraulic mechanism, at the core of which is elevation and protraction of the hyoid. Here, the musculoskeletal mechanisms driving these hyoid movements are experimentally evaluated in macaque primates. MATERIALS AND METHODS: We integrate XROMM-based mea…

Evolutionary echoes of emotion: Humans mimic other primate expressions.

Humans readily mimic the emotional behavior of conspecifics -- a behavior linked to empathy. Yet, whether humans unconsciously mimic the emotional expressions of non-human primates remains an open question. Human observers watched short videos with positive (play-face), negative (open-mouth threat display) or neutral expressions by monkeys and apes (while their own facial expressions were filmed and automatically coded), rated the expressions for emotional content and indicated their degree of liking of and closeness to the primates. Participants mimicked both positive and negative expressions…

Coordinated neural progenitor adaptations contribute to primate neocortical downscaling.

Most primates have a large, highly folded (gyrencephalic) neocortex, a feature present in the primate common ancestor. In contrast, several New World monkey species display a comparatively small and largely smooth, unfolded (lissencephalic) neocortex. One prominent example is the common marmoset, an increasingly popular model in neuroscience. This phenotype likely reflects an evolutionary reduction from the ancestral primate condition, implying modifications in neurodevelopmental programs. One essential basis for neocortical development is the activity and behavior of neural progenitor cells (…

Working Memory of Multi-Object Scenes in Primate Frontal Cortex.

Working memory allows primates to reason about complex scenes, yet how the brain maintains multiple objects in memory simultaneously remains unclear. Competing theories propose that objects are stored in discrete slots1,2, represented dynamically through switching3-6, or encoded by weighted combinations of single-object representations7-11. We formalized these hypotheses in terms of their quantitative predictions at the level of single neurons and tested them against densely recorded neural data from the dorsomedial frontal cortex and frontal eye field of monkeys trained to perform a novel mul…

De novo fast motion computation in the primate visual cortex.

Objects move through space and time, generating sequential visuotopic activations in all sighted animals, leading to velocity perception defined by direction and speed. The primary visual cortex (V1) only encodes directionality at low speed, yet we effortlessly perceive velocities ranging from 0.25°/s to 500°/s. To resolve this paradox, we recorded neuronal responses to moving dots, gratings, and movies across the lateral geniculate nucleus (LGN), V1, middle temporal (MT) area, and medial superior temporal (MST) area of the macaque motion pathway. Regardless of cell type and motion stimuli, V1

The cellular and molecular basis of gyrus-sulcus specialization in the primate brain.

Gyri and sulci are key anatomical features of the primate cerebral cortex, distinguished by their structural and functional specialization, yet their cellular and molecular foundations remain poorly understood. Here we utilized single-cell spatial transcriptomics data from adult male macaque monkey brains and developed gsModel, a customized deep learning framework that distinguishes sulci from gyri. Relying solely on local cell-type composition, gsModel achieved over 90% accuracy in the classification task, outperforming alternative machine learning models and predictors based on anatomical fe

Visual activity in primate superior colliculus requires geniculostriate input.

The superior colliculus (SC) is an ancient visual structure whose principle source of visual drive comes directly from the retina. In primates, however, the SC also receives geniculostriate input from primary visual cortex (V1) via the lateral geniculate nucleus (LGN), making it unclear which pathway normally drives visually evoked spiking. Here we tested whether visually evoked spiking in the primate SC depends on retinal signals routed through LGN and V1, rather than on direct retinal input alone. We recorded from macaque SC neurons before and during reversible inactivation of the ipsilatera…

Evolutionary Pathways in Brain Morphology, Development, and Behavior in the Primate Lineage.

The article collection 'How Primates Became the Brainiest of All Mammals: A Closer Look at the Evolution of the Primate and Human Cortex' presents a wide-ranging analysis of encephalization across primates and humans. The papers in the collection tackle multiple classic themes in primatology, from brain ontogeny and relative brain size to cortical reorganization and metabolism. Taken together, the contributions reveal how the primate brain is more than a scaled-up version of the basic mammalian design. The primate brain uniquely evolved towards the cognitive scaffolding that ultimately brought…

This review summarizes our current knowledge of the anatomy of the primate (including human) retina. Understanding the structure and functio … types of ganglion cells. In the mouse retina, the number of bipolar types is comparable to that in primates, but the number of amacrine (over 60 types) and ganglion (over 30 types) cell types is significantly higher, indicating a reduction in the number of retinal pathways in primates. Other important features found in primate retinas (but absent from mou …

Gut bacterial regulation of primate adaptive thermogenesis at high altitude.

BACKGROUND: Adaptive thermogenesis is a fundamental physiological process by which mammals maintain their body temperature through shivering and non-shivering thermogenesis to adapt to environmental changes. The activation of brown adipose tissue (BAT) is the core of non-shivering thermogenesis. However, the specific mechanisms by which gut-derived bacteria trigger BAT to activate adaptive thermogenesis remain poorly understood. Environmental variations across different altitudes create unique temperature gradients, providing an ideal condition for identifying bacteria associated with cold ada

Neurotensin Regulates Primate Ovulation Via Multiple Neurotensin Receptors.

Neurotensin (NTS), a small neuropeptide, was recently established as a key paracrine mediator of ovulation. NTS mRNA is highly expressed by granulosa cells in response to the luteinizing hormone surge, and multiple NTS receptors are expressed by cells of the ovulatory follicle. To identify the role of NTS receptors NTSR1 and SORT1 in ovulation in vivo, the dominant follicle of cynomolgus macaques (Macaca fascicularis) was injected with either vehicle control, the general NTS receptor antagonist SR142948, the NTSR1-selective antagonist SR48692, or the SORT1-selective antagonist AF38469. hCG was

Imaging of Microtubules in a Nonhuman Primate Model of Alcohol Use Disorder.

Microtubules (MTs), as structural components of the cytoskeleton, are vital for axonal transport, information processing, and signaling within the brain. Disruption of MT integrity has been associated with neurological disorders, including alcohol use disorder (AUD). This study used positron emission tomography (PET) imaging with the radiotracer [11C]MPC-6827 to investigate alcohol-induced changes in MT dynamics in a nonhuman primate model of AUD. Dynamic PET scans (0-120 min) were conducted in male cynomolgus monkeys (n = 4) before and after ∼7 months of ethanol self-administration (EtOH SA).

Convergent evolution through independent rearrangements in the primate amylase locus.

Structurally complex regions of the genome are increasingly recognized as engines of evolutionary convergence due to their propensity to generate recurrent gene duplications that give rise to similar gene expression patterns and traits across lineages. However the mutational mechanisms driving these duplications and the regulatory changes enabling novel expression patterns remain poorly understood. The primate amylase locus, marked by independent gene duplications, provides an ideal model to investigate these dynamics. Leveraging high-quality genome assemblies from 53 primates and multi-tissue…

The Evolution of Immunosuppressive Therapy in Pig-to-Nonhuman Primate Organ Transplantation.

An overview is provided of the evolution of strategies towards xenotransplantation during the past almost 40 years, focusing on advances in gene-editing of the organ-source pigs, pre-transplant treatment of the recipient, immunosuppressive protocols, and adjunctive therapy. Despite initial challenges, including hyperacute rejection resulting from natural (preformed) antibody binding and complement activation, significant progress has been made through gene editing of the organ-source pigs and refinement of immunosuppressive regimens. Major steps were the identification and deletion of expressi

ASIC currents in cultured primate retinal amacrine/ganglion cells.

Acid-sensing ion channels (ASICs) are proton-gated cation channels belonging to the epithelial Na + channel/degenerin superfamily. In the CNS, ASICs are involved in synaptic plasticity, learning/memory, and acidosis-mediated injury. Previous studies showed that ASICs are expressed in rodent retina where activation likely participates in the phototransduction process and retinal integrity. However, there have been no studies examining the expression of ASICs in primate retina. Using molecular biology and patch-clamp techniques, we explored the expression of ASICs in monkey retina and cultured m

Optogenetic stimulation of single ganglion cells in the living primate fovea.

Though the responses of the rich variety of retinal ganglion cells (RGCs) reflect the totality of visual processing in the retina and provide the sole conduit for those processed responses to the brain, we have much to learn about how the brain uses these signals to guide behavior. An impediment to developing a comprehensive understanding of the role of retinal circuits in behavior is the paucity of causal studies in the intact primate visual system. Here, we demonstrate the ability to optogenetically activate individual RGCs with flashes of light focused on single RGC somas in vivo, with no e

Applied anatomy and morphology of Meibomian glands in the non-human primate.

Dry eye disease (DED) is a common ocular condition that has been estimated to affect ~ 10 to 55.4% of the global population. Symptoms of DED include eye irritation, ocular pain and discomfort, inflammation, and photophobia, and, if left untreated, can lead to infection, corneal neuropathy, corneal scarring and impaired vision. Studies have shown that over 70% of all DED cases are caused by some form of Meibomian gland dysfunction (MGD). To date the etiology of MGD remains unknown, therefore, there is a need for further research into understanding the development, homeostasis and pathology of t

Molecular characterization of Bertiella studeri infecting a primate in South India.

Bertiella spp. is a mite-borne cestode parasite that inhabits the small intestine of wide range of mammals, including non-human primates. In the present study, the morphological and molecular analysis of Bertiella studeri recovered from the small intestine of a bonnet macaque (Macaca radiata) from Wayanad, Kerala (South India) was performed. Acetic alum carmine staining identified the cestode morphologically based on the characters like broader proglottids, which contain irregularly alternating genital pores, single set of reproductive organs, 280 testes and a tubular transverse uterus. Molecu…
