Lysine Malonylation and Its Links to Metabolism and Diseases.

Malonylation is a recently identified post-translational modification with malonyl-coenzyme A as the donor. It conserved both in prokaryotes and eukaryotes. Recent advances in the identification and quantification of lysine malonylation by bioinformatic analysis have improved our understanding of its role in the regulation of protein activity, interaction, and localization and have elucidated its involvement in many biological processes. Malonylation has been linked to diverse physiological processes, including metabolic disorders, inflammation, and immune regulation. This review discusses mal

Anti-epileptic drugs and bone loss: Phenytoin reduces pro-collagen I and alters the electrophoretic mobility of osteonectin in cultured bone cells.

Phenytoin is an antiepileptic drug used in the management of partial and tonic-clonic seizures. In previous studies we have shown that valproate, another antiepileptic drug, reduced the amount of two key bone proteins, pro-collagen I and osteonectin (SPARC, BM-40), in both skin fibroblasts and cultured osteoblast-like cells. Here we show that phenytoin also reduces pro-collagen I production in osteoblast-like cells, but does not appear to cause a decrease in osteonectin message or protein production. Instead, a 24h exposure to a clinically relevant concentration of phenytoin resulted in a dose

Ubiquitin signaling in tumor-associated macrophage reprogramming: mechanisms and therapeutic opportunities in cancers.

Tumor-associated macrophages (TAMs) are key regulators of tumor progression and immune suppression within the tumor microenvironment (TME). Emerging evidence identifies ubiquitination, a dynamic post-translational modification that regulates protein stability, localization, and signaling activity, as a critical determinant of TAM function. Ubiquitin signaling orchestrates macrophage inflammatory responses, metabolic reprogramming, and tumor cell-immune cell crosstalk through coordinated actions of ubiquitin-activating enzymes, ubiquitin-conjugating enzymes, ubiquitin-protein ligases (E3s), and

Cardiac Protein Palmitoylation: Mechanisms, Functions, and Therapeutic Potential.

Protein S-palmitoylation is a reversible lipid modification which regulates the localisation, stability, and activity of a substantial fraction of the proteome. In the heart S-palmitoylation controls the function of membrane proteins which regulate cardiac excitability and contractility. Recent work has moved the S-palmitoylation field beyond descriptive cataloguing towards mechanistic understanding, revealing how site-specific S-palmitoylation can modulate protein structure and function in a precise and quantifiable manner. These advances position S-palmitoylation as a tuneable regulator of c

Polystyrene nanoplastics drive neuronal senescence via PP2A-B56γ-targeted p-Ebp1Ser335 dephosphorylation-mediated ribosome biogenesis dysfunction.

Nanoplastics (NPs) exhibit neurotoxicity, yet the precise molecular mechanisms remain elusive. In this study, we established a human-relevant polystyrene nanoplastics (PS-NPs, 50 mg kg-1) oral exposure model in C57BL/6 mice in vivo and a neuro-immune microglial-neuron co-culture system (HMC-3/SH-SY5Y cells) in vitro to dissect these mechanisms. We demonstrate that PS-NPs exposure triggers microglial M1 activation and drives neuronal senescence. Mechanistically, PS-NPs activate the protein phosphatase 2A (PP2A)-B56γ subunit, which selectively dephosphorylates the ribosome biogenesis regulator E

When metabolic enzymes meet lactylation: a bidirectional dialogue in health and disease.

Lactate accumulation occurs in diverse physiological and pathological states, including hypoxia, ischemia, development, wound healing, immune activation, and cancer, where it functions as a key regulator of cellular energy metabolism, signal transduction, microenvironmental homeostasis, and immune responses. Protein lactylation was first described in 2019, revealing that lactate can serve as a substrate for post-translational protein modification. This review focuses on the bidirectional interplay between metabolic enzymes and lactylation. On one hand, metabolic enzymes control lactate product

Protein arginine methyltransferases in protozoan parasites: a new path for antiparasitic chemotherapy?

Protein arginine methyltransferases (PRMTs) catalyse the transference of methyl groups from S-adenosylmethionine to arginine residues in substrate proteins, a post-translational modification widespread among eukaryotes. The change in size and hydrophobicity of the methylated arginine residue impacts on how a protein interacts with other macromolecules and affects several cellular processes, including intracellular signaling, DNA replication and repair, and control of gene expression. As a result, PRMTs became attractive targets for chemotherapy, and several PRMT inhibitors are going through cl

UFMylation: A Potential Modification for Neurological Diseases.

Neurological disorders are the leading health threats worldwide, characterized by impairments in consciousness, cognition, movement, and sensation, and can even lead to death. UFMylation is a novel post-translational modification (PTM) that serves as an important regulatory factor, promoting the complexity of protein structures and enhancing the diversity and specificity of functions. In UFMylation, ubiquitin-fold modifier 1 (UFM1) is covalently transferred to the primary amine of a lysine residue on the target protein through the synergistic action of three enzymes: the activating enzyme E1 o

AMPA receptor palmitoylation restrains ictogenesis in the epileptogenic lesion of patients with hippocampal sclerosis.

Failure of appropriate synaptic regulation in the epileptogeniclesion leads to sudden onset ictogenesis in severe epileptic seizure. Using surgically excised patient brain tissue, our recent biochemical study revealed chronic reduction of synaptic protein expressions in the epileptogenic lesions of patients with hippocampal sclerosis. α-amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA) receptors are the major excitatory neurotransmitter receptors in human brain. Decreased AMPA receptors expression and their phosphorylations in the epileptogenic lesion induce an overall reduction of excita

Cysteine-S-nitrosylation inhibits Rop5-mediated immune evasion in Toxoplasma gondii.

Reactive nitrogen species (RNS) are a mechanism to control microbial infections conserved across the host species of the obligate intracellular parasite Toxoplasma gondii. Cysteine S-nitrosylation (SNO) is a reversible post-translational modification that controls complex cell behaviors by regulating protein interactions and signal transduction events. Here we identified a cluster of T. gondii secreted effector proteins that are SNO-modified in a host inducible nitric oxide synthetase (iNOS)-dependent manner. Among these were the rhoptry protein 5 (ROP5) paralogs, which are the major virulence

Autoantibodies targeting malondialdehyde-modifications in rheumatoid arthritis regulate osteoclasts via inducing glycolysis and lipid biosynthesis.

Proteins subjected to post-translational modifications, such as citrullination, carbamylation, acetylation or malondialdehyde (MDA)-modification are targeted by autoantibodies in seropositive rheumatoid arthritis (RA). Epidemiological and experimental studies have both suggested the pathogenicity of such humoral autoimmunity, however, molecular mechanisms triggered by anti-modified protein antibodies have remained to be identified. Here we describe in detail the pathways induced by anti-MDA modified protein antibodies that were obtained from synovial B cells of RA patients and that possessed r

N-alpha-acetylation of Huntingtin protein increases its propensity to aggregate.

Huntington's disease (HD) is a neurodegenerative disorder caused by a poly-CAG expansion in the first exon of the HTT gene, resulting in an extended poly-glutamine tract in the N-terminal domain of the Huntingtin (Htt) protein product. Proteolytic fragments of the poly-glutamine-containing N-terminal domain form intranuclear aggregates that are correlated with HD. Post-translational modification of Htt has been shown to alter its function and aggregation properties. However, the effect of N-terminal Htt acetylation has not yet been considered. Here, we developed a bacterial system to produce u

Characterization of a new member of kunitz-type protein family from the venom of Persian false-horned viper, Pseudocerastes persicus.

A new member of kunitz-type protein family, PPTI (PseudocerastesPersicusTrypsin Inhibitor), was isolated from the venom of Persian false horned viper Pseudocerastes persicus and characterized. Mass spectrometry and amino acid sequencing revealed that PPTI is a 68 amino acid protein with molecular weight of about 7.6 kDa. The first amino acid residue of PPTI is N-terminally blocked via a post translational modification to pyroglutamyl. Sequence comparison against UniProtKB shows a high sequence similarity of PPTI with kunitz-type proteins, especially serine protease inhibitors and dendrotoxins

Quantitative Succinyl-Proteome Profiling of Camellia sinensis cv. 'Anji Baicha' During Periodic Albinism.

Lysine succinylation is a novel dynamic and evolutionarily conserved post-translational modification (PTM) that regulates various biological processes. 'Anji Baicha' is an albino tea variety that exhibits temperature-based variability of leaf colour and amino acid concentrations. However, the mechanism underlying albinism in 'Anji Baicha' has not been investigated at the level of succinylation. Here, we identify 3530 lysine succinylation sites mapped to 2132 proteins in 'Anji Baicha', representing the first extensive data on the lysine succinylome in the tea plant. Eleven conserved succinylati

The antiviral activities of ISG15.

Post-translational protein modification is an important strategy for the regulation of the cell proteome independent of the need for new gene expression. Ubiquitin and ubiquitin-like modifiers mediate the regulation of protein levels, signaling pathways, vesicular trafficking, and many other cellular processes through their covalent conjugation to proteins. Interferon stimulated gene 15 (ISG15) is a ubiquitin-like modifier induced by type I interferon. In addition to conjugating to potentially hundreds of target proteins, ISG15 can be found in an unconjugated form both inside of the cell and r

Overcoming resistance in chronic myelogenous leukemia.

Chronic myeloid leukemia is defined by the acquired genetic mutation, t(9;22), which leads to the fusion-protein BCR-ABL. Prior to the development of imatinib mesylate (Gleevec), treatment was limited and provided only limited survival benefit. Imatinib has dramatically changed the course of the disease and has led to the significantly prolonged survival in the majority of patients. However, there is growing concern for resistance to imatinib and to subsequent second generation tyrosine kinase inhibitors (dasatinib and nilotinib) due to the T315I mutation. With no currently approved effective

A rab-related GTP-binding protein in Schistosoma mansoni.

Schistosomiasis is a parasitic disease initiated by the deposition of eggs in host tissues by the blood fluke Schistosoma. A gene encoding a low-molecular weight GTP-binding protein (LMWGP) was cloned from Schistosoma mansoni using a polymerase chain reaction (PCR)-based strategy. The gene was termed smrab (Schistosoma mansoni rab-related protein). Northern blot analysis hybridizes smrab cDNA with a 1.5-kb band of mRNA; this mRNA is abundantly expressed in male schistosomes, while only slightly in females. The deduced amino acid sequence of smrab shares ca. 70% homology with that of several ra

Crosstalk between ubiquitination and other post-translational modifications in cardiovascular diseases.

Cardiovascular diseases (CVDs) remain a major cause of morbidity and mortality worldwide. Post-translational modifications (PTMs) are increasingly recognized as important regulators of cardiovascular pathophysiology, but how different PTMs interact in CVDs is still incompletely understood. This review focuses on the crosstalk between ubiquitination and other PTMs, including phosphorylation, SUMOylation, acetylation, methylation, O-GlcNAcylation, neddylation, and ADP-ribosylation/PARylation. We summarize representative cardiovascular mechanisms in which these modifications act competitively, co

Phosphorylation, acetylation and ubiquitination: the molecular basis of RUNX regulation.

The RUNX family members play pivotal roles in normal development and neoplasia. RUNX1 and RUNX2 are essential for hematopoiesis and osteogenesis, respectively, while RUNX3 is involved in neurogenesis, thymopoiesis and functions as a tumor suppressor. Inappropriate levels of RUNX activity are associated with leukemia, autoimmune disease, cleidocranial dysplasia, craniosynostosis and various solid tumors. Therefore, RUNX activity must be tightly regulated to prevent tumorigenesis and maintain normal cell differentiation. Recent work indicates that RUNX activity is controlled by various extracell

Accessory gland protein regulates pairing process and oviposition in the subterranean termite Reticulitermes chinensis after swarming.

Swarming and pairing behaviors are significant to population dispersal of termites. Tandem running is a key process in pairing behavior of dealates to find a mate. Succinylation can lead to significant changes in protein structure and function, which is widely involved in metabolism and behavior regulation in many organisms. However, whether succinylation modification regulates termites' tandem running is currently unknown. In this research, we performed quantitative modified proteomics of the subterranean termite Reticulitermes chinensis Snyder before and after alate swarming. The succinylati
