Stimulation of sodium and calcium uptake by scorpion toxin in chick embryo heart cells.

Scorpion toxins, the basic miniproteins of scorpion venom, stimulated the passive uptake of Na+ and Ca2+ in chick embryo heart cells. Half-maximum stimulation was obtained for 20-30 nM Na+ and 40-50 nM Ca2+. Scorpion toxin-activated Na+ and Ca2+ uptakes were fully inhibited by tetrodotoxin, a specific inhibitor of the action potential Na+ ionophore in excitable membranes. Half-maximum inhibition was obtained with the same concentration of tetrodotoxin (10 nM) for both Na+ and Ca2+. Scorpion toxin-stimulated Ca2+ uptake was dependent on extracellular Na+ concentration and was not inhibited by C

Correlation of Venom Toxinome Composition of Indian Red Scorpion (Mesobuthus tamulus) with Clinical Manifestations of Scorpion Stings: Failure of Commercial Antivenom to Immune-Recognize the Abundance of Low Molecular Mass Toxins of This Venom.

The Indian red scorpion (Mesobuthus tamulus), with its life-threatening sting, is the world's most dangerous species of scorpion. The toxinome composition of M. tamulus venom was determined by tandem mass spectrometry (MS) analysis of venom protein bands separated by SDS-PAGE. A total of 110 venom toxins were identified from searching the MS data against the Buthidae family (taxid: 6855) of toxin entries in nonredundant protein databases. The Na+ and K+ ion channel toxins taken together are the most abundant toxins (76.7%) giving rise to the neurotoxic nature of this venom. The other minor tox

Characterization of a novel cDNA encoding a short venom peptide derived from venom gland of scorpion Buthus martensii Karsch: trans-splicing may play an important role in the diversification of scorpion venom peptides.

A novel cDNA clone (named BmKT-u) which is a hybrid molecule of the 5'-terminal region of BmKT' cDNA and the 3'-terminal region of an undocumented cDNA (named BmKu), was isolated from a cDNA library made from the venom gland of scorpion Buthus martensii Karsch. BmKT-u codes for a 30 amino acid residue precursor peptide composed of a 20-residue signal sequence, and a putative 10-residue novel mature peptide. Northern blot hybridization showed BmKT-u cDNA is generated from a transcript. RT-PCR experiments excluded the possibility that BmKT-u cDNA is an artifact generated during reverse transcrip

Ayurvedic approaches to scorpion stings: Historical insights, modern scientific perspectives and challenges.

Scorpion stings are medical emergencies that require immediate attention and are frequently overlooked. Commercial antivenom available only … ge of 'Vrishchika Damsha' (scorpion sting) focused only on the history and Ayurvedic perspective on scorpions, as well as on approaches to treating scorpion envenomation in India. The study showed that Agada Tantra, a specialized branch of Ayurvedic Toxicology, focuses on understanding and treating various toxins, including those produced by scorpions (Vris …

Scorpion gut microbiomes as a source of bioactive and rare actinobacteria with nonribosomal peptide potential.

Actinobacteria are well-known for their ability to produce bioactive secondary metabolites, including nonribosomal peptides (NRPs); however, many host-associated reservoirs remain underexplored. In this study, cultivable actinobacteria were isolated from scorpion gut, including both the intestinal tissue and its luminal contents, and evaluated their antimicrobial activity and presence of NRPs genes. Fifty scorpions (Odontobuthus doriae and Mesobuthus eupeus) were collected from Qom Province, Iran. Dissected gut homogenates were plated on selective media, yielding 120 pure isolates. Taxonomic i…

Therapeutic Potential of Scorpion Venom Proteins: Innovations in Cancer Treatment and Future Directions.

Cancer is a significant human health concern due to its increasing mortality rate and profound impact on public health and healthcare systems. The cytotoxic, antiproliferative, immunosuppressive, and apoptogenic properties of scorpion venom proteins and peptides have been observed in various cancer cell lines. Therefore, the purpose of this study was to investigate the potential use of proteins derived from scorpion venom in cancer treatment. In this study, the effects of different scorpion venoms on transmembrane channels, the inhibition of angiogenesis, the inhibition of invasion and metasta

A Fluorescent Protein-Tagged Scorpion Toxin with Picomolar Affinity for the Kv1.3 K + Channel.

The fusion of fluorophores and peptide pore blockers generates valuable molecular tools to identify cells that express the voltage-gated potassium channel Kv1.3. Changes in the Kv1.3 expression are associated with numerous autoimmune and neurological disorders. In this study, Vm24, a potent and selective Kv1.3 inhibitor peptide isolated from scorpion venom, was genetically linked to mCherry, a red fluorescent protein characterized by high photostability, maturation, and tagging tolerance. The recombinant fusion protein (mCherry-Vm24) was expressed utilizing the Pichia pastoris expression syste

Can Scorpion Venom Peptides Be Safely Used in Cardiovascular Therapy: A Systematic Review.

Scorpion venom contains numerous bioactive peptides with potent cardiovascular effects, including bradykinin-potentiating peptides (BPPs), ion channel modulators, and cardioprotective molecules. These peptides show promise for conditions such as hypertension, cardiac injury, and arrhythmias. However, concerns regarding toxicity, immunogenicity, and off-target actions have limited their clinical development. This systematic review evaluates the therapeutic potential and safety of scorpion venom peptides for cardiovascular applications. A systematic search of PubMed, Scopus, Google Scholar, and
