Dual stem cell therapy synergistically improves cardiac function and vascular regeneration following myocardial infarction.

Since both myocardium and vasculature in the heart are excessively damaged following myocardial infarction (MI), therapeutic strategies for treating MI hearts should concurrently target both so as to achieve true cardiac repair. Here we demonstrate a concomitant method that exploits the advantages of cardiomyocytes derived from human induced pluripotent stem cells (hiPSC-CMs) and human mesenchymal stem cell-loaded patch (hMSC-PA) to amplify cardiac repair in a rat MI model. Epicardially implanted hMSC-PA provide a complimentary microenvironment which enhances vascular regeneration through prol

Detection, isolation, and identification of Vibrio cholerae from the environment.

Recent molecular advances in microbiology have greatly improved the detection of bacterial pathogens in the environment. These improvements and a downward trend in the cost of molecular detection methods have contributed to increased frequency of detection of pathogenic microorganisms where traditional culture-based detection methods have failed. Culture methods also have been greatly improved, and the confluence of the two suites of methods provides a powerful tool for detection, isolation, and characterization of pathogens. While molecular detection provides data on the presence and type of

This … 7R with agents such as Brilliant Blue G, A-438079, botulinum toxin A, mesenchymal stem cell-derived secretomes, and high-frequency spinal cord stimulation produces marked, P2X7R-dependent analgesic effects. P2X7R stands out as a promising therapeutic target for neuropathic pain. Future priorities should focus on optimizing P2X7R-targeted pharmacodynamics, d …

Synthetic peptide V11A reduces bacterial load and inflammation in pneumococcal meningitis.

INTRODUCTION: Pneumococcal meningitis, caused by Streptococcus pneumoniae, is exacerbated by factors released during bacterial lysis, triggering inflammation and damage to host cells. Bacteriolytic antibiotics increase inflammation meaning that there is a high unmet medical need for non-bacteriolytic antimicrobials as alternative therapeutic options. Previously, an 11-amino acid peptide was discovered in the secretome of Klebsiella pneumoniae, named V11A, and was found to inhibit growth of S. pneumoniae in a bacteriostatic and species-specific manner in vitro. METHODS AND RESULTS: Here it is …

Senescence, Aging and Disease Throughout the Gastrointestinal System.

Senescence is an irreversible cell cycle arrest-characterized by morphologic alterations, genomic instability, and secretome changes-that profoundly affects the tissue structure and function. Accumulating evidence indicates that senescence plays a relevant role in gastrointestinal pathologies: senescence contributes to salivary gland hypofunction, is instrumental in the development of oral submucous fibrosis, drives age-dependent hepatic steatosis, and regulates the clinical progression of steatotic liver disease. Senescence in the biliary tract develops in response to ischemic injury in bilia…

Regenerative Medicine in Urogynecology: Where We Are and Where We Want to Be.

Pelvic floor disorders (PFDs) constitute a major public health issue given their negative effect on quality of life for millions of women worldwide and the associated economic burden. As the prevalence of PFDs continues to increase, novel therapeutic approaches for the effective treatment of these disorders are urgently needed. Regenerative medicine techniques, including cellular therapies, extracellular vesicles, secretomes, platelet-rich plasma, laser therapy, and bioinductive acellular biomaterial scaffolds, are emerging as viable clinical options to counteract urinary and fecal incontinenc…

Astrocyte-derived factors regulate CNS myelination.

The role that astrocytes play in central nervous system (CNS) myelination is poorly understood. We investigated the contribution of astrocyte-derived factors to myelination and revealed a substantial overlap in the secretomes of human and rat astrocytes. Using in vitro myelinating co-cultures of primary retinal ganglion cells and cortical oligodendrocyte precursor cells, we discovered that factors secreted by resting astrocytes, but not reactive astrocytes, facilitated myelination. Soluble brevican emerged as a new enhancer of developmental myelination in vivo, CNS and its absence was linked t…

Extracellular Vesicles: Schistosomal Long-Range Precise Weapon to Manipulate the Immune Response.

Schistosomiasis (Bilharziasis), a neglected tropical disease that affects more than 240 million people around the world, is caused by infection with the helminth parasite Schistosoma. As part of their secretome, schistosomes release extracellular vesicles (EVs) that modulate the host immune response. The EV-harbored miRNAs upregulate the innate immune response of the M1 pathway and downregulate the differentiation toward the adaptive Th2 immunity. A schistosomal egg-derived miRNA increases the percentage of regulatory T cells. This schistosomal-inducible immunoediting process generates ultimat…

Marked Increased Production of Acute Phase Reactants by Skeletal Muscle during Cancer Cachexia.

Loss of skeletal muscle mass in cancer cachexia is recognized as a predictor of mortality. This study aimed to characterize the changes in the muscle secretome associated with cancer cachexia to gain a better understanding of the mechanisms involved and to identify secreted proteins which may reflect this wasting process. The changes in the muscle proteome of the C26 model were investigated by label-free proteomic analysis followed by a bioinformatic analysis in order to identify potentially secreted proteins. Multiple reaction monitoring and Western blotting were used to verify the presence o…

Proteomics at the schistosome-mammalian host interface: any prospects for diagnostics or vaccines?

Since 2004 there has been a remarkable increment in our knowledge of the proteins and glycans that reside at, or are released from the surfaces of schistosomes in the mammalian host. Initial characterization of the soluble proteome permits distinctions to be made between the parasite secretome and its necrotome. The principal proteins secreted by the cercaria to gain access to the skin have been described as well as those released by migrating schistosomula. An inventory of transporters, enzymes and structural proteins has been shown to reside the tegument surface, but also immunoglobulins, co…
