C-X-C chemokine receptor type 4 (CXCR4) is highly expressed in a range of pathologies, including cancers like multiple myeloma and non-Hodgkin lymphoma, inflammatory diseases such as rheumatoid arthritis, and viral infections like HIV. Currently, the most advanced radiotracer for CXCR4 imaging in clinics is [68Ga]PentixaFor. However, its structure is prone to modifications, complicating the development of a specific CXCR4 fluorine-18-labeled tracer with good pharmacokinetic properties. This study aimed to screen multiple CXCR4-targeting variable domains of heavy-chain-only antibody (VHH or sin

Rationale: The efficacy of drug delivery to the brain is constrained by the impermeability of the blood-brain barrier (BBB) in healthy tissues and the heterogeneous permeability of the blood-tumor barrier (BTB) in gliomas. Focused ultrasound (FUS) has emerged as a promising technique to transiently modulate vascular permeability, however its effects vary across different brain tissues. This study systematically evaluates the effects of FUS-induced vascular permeability modulation in the gray matter (GM), white matter (WM), and brain tumors, considering their distinct tissue architectures, vasc

Herein partially summarizes one scientist-clinician's wanderings through the jungles of primate aqueous humor outflow over the past ~45 years. Totally removing the iris has no effect on outflow facility or its response to pilocarpine, whereas disinserting the ciliary muscle (CM) from the scleral spur/trabecular meshwork (TM) completely abolishes pilocarpine's effect. Epinephrine increases facility in CM disinserted eyes. Cytochalasins and latrunculins increase outflow facility, subthreshold doses of cytochalasins and epinephrine given together increase facility, and phalloidin, which has no ef

This investigation represents the largest study so far published on human DNA damage and aging. The subject of this investigation is damage, determined as DNA alterations which give rise to complete molecular breaks in the course of treatment of purified DNA solutions with single-strand-specific nucleases. The DNA is derived from milligram samples of human muscle of individuals mostly undergoing surgical treatment. Care has been taken to bring the muscle samples, once shut off from blood circulation to liquid nitrogen temperatures within few seconds. The DNA is prepared by a procedure keeping

The French chemist Michel Eugène Chevreul discovered creatine in meat two centuries ago. Extensive biochemical and physiological studies of this organic molecule followed with confirmation that creatine is found within the cytoplasm and mitochondria of human skeletal muscles. Two groups of investigators exploited these relationships five decades ago by first estimating the creatine pool size in vivo with 14 C and 15 N labelled isotopes. Skeletal muscle mass (kg) was then calculated by dividing the creatine pool size (g) by muscle creatine concentration (g/kg) measured on a single muscle biopsy

Myoclonus dystonia is a childhood-onset hyperkinetic movement disorder with a combined motor and psychiatric phenotype. It represents one of the few autosomal dominant inherited dystonic disorders and is caused by mutations in the ε-sarcoglycan (SGCE) gene. Work to date suggests that dystonia is caused by disruption of neuronal networks, principally basal ganglia-cerebello-thalamo-cortical circuits. Investigation of cortical involvement has primarily focused on disruption to interneuron inhibitory activity, rather than the excitatory activity of cortical pyramidal neurons. Here, we have sought

COVID-19 is characterised by a broad spectrum of clinical and pathological features. Natural killer (NK) cells play an important role in innate immune responses to viral infections. Here, we analysed the phenotype and activity of NK cells in the blood of COVID-19 patients using flow cytometry, single-cell RNA-sequencing (scRNA-seq), and a cytotoxic killing assay. In the plasma of patients, we quantified the main cytokines and chemokines. Our cohort comprises COVID-19 patients hospitalised in a low-care ward unit (WARD), patients with severe COVID-19 disease symptoms hospitalised in intensive c

INTRODUCTION: Autosomal dominant polycystic kidney disease (ADPKD) is characterized by the slow, progressive formation of fluid-filled cysts within the kidneys, ultimately leading to kidney failure. Despite being the most common genetic kidney disease, therapies for ADPKD are limited. Ureteric bud (UB) organoids derived from human induced pluripotent stem cells (hiPSC) have been developed to model ADPKD. Here, we describe a protocol to generate UB organoids in large-scale suspension culture amenable to drug discovery.
METHODS: Day 12 'nephron' organoids derived from wildtype and PKD2-deficien

Atomic force microscopy (AFM) is a promising method for generating high-spatial-resolution images, providing insightful perspectives on the nanomechanical attributes of soft matter, including cells, bacteria, viruses, proteins, and nanoparticles. AFM is widely used in biological and pharmaceutical sciences because it can scrutinize mechanical properties under physiological conditions. We comprehensively reviewed experimental techniques and fundamental mathematical models to investigate the mechanical properties, including elastic moduli and binding forces, of soft materials. To determine these

The formation of inorganic nanomaterials is highly sensitive to the local environment. When environmental conditions are carefully selected, they enable precise control over the composition, shape and dimensionality of inorganic nanostructures, which, in turn, influences their functional properties. In this study, we explore the impact of single-walled carbon nanotubes (SWNTs) on the sequential transformation of triosmium dodecacarbonyl (Os3(CO)12) into osmium iodide (OsI) and subsequently osmium disulphide (OsS2). Initially, the nanotubes form a van der Waals complex with Os3(CO)12, allowing

Drug-targeted delivery has become a top priority in the world of medicine in order to develop more efficient therapeutic agents. This is important as a critical underlying problem in cancer therapy stems from the inability to deliver active therapeutic substances directly to tumor cells without causing collateral damage. In this work, zinc(II) phthalocyanine (ZnPc) was selected as a sensitizer and was linked to different targeting agents, which would be recognized by overexpressed proteins in cancer cells. As targeting agents, we first selected the two ligands (DAA1106, PK11195) of the translo

Bright, photostable luminescent labels are powerful tools for the in vitro and in vivo imaging of biological events. Semiconductor nanocrystals have emerged as attractive alternatives to commonly used organic lumophores because of their high quantum yields and the spectral tunability that can be achieved through synthetic control. Although conventional synthetic methods generally yield high-quality nanocrystals with excellent optical properties for biological imaging, ligand exchange and biological conjugation are necessary to make nanocrystals biocompatible and biospecific. These steps can su

Huntington's disease is one of several neurodegenerative disorders characterized by the aggregation of polyglutamine (polyQ)-expanded mutant protein. How polyQ aggregation leads to cellular dysfunction is not well understood. Here, we analyzed aberrant protein interactions of soluble oligomers and insoluble inclusions of mutant huntingtin using in-cell single molecule fluorescence spectroscopy and quantitative proteomics. We find that the interactome of soluble oligomers is highly complex, with an enrichment of RNA-binding proteins as well as proteins functioning in ribosome biogenesis, transl

Retinol, a bioactive small molecule of the vitamin A family, contributes epidermal and dermal tissue repair through retinoic acid receptor γ (RARγ)/retinoid X receptor (RXR) receptor-mediated transcriptional regulation of keratinocyte differentiation, extracellular matrix (ECM) remodeling, and barrier restoration. However, the relationship between applied concentration, tissue-level regenerative outcomes, and tolerability remains incompletely characterized. This prospective, randomized, single-blind study compared biophysical effects and tolerance of two retinol concentrations in EU-compliant

BACKGROUND: While blood biomarkers for AD have proven useful in identifying AD pathology from other neurodegenerative conditions, their prognostic value in real-world clinical settings, such as memory clinics, remains unexplored. We aimed to examine the prognostic value of AD plasma biomarkers and their short-term changes within 4 months, for predicting clinical and neuroradiological progression over two years. METHOD: We performed a prospective observational longitudinal cohort study of patients with early stages of AD and followed them for up to 2 years. Plasma was analysed for neurofilament

BACKGROUND: Heart-type fatty acid-binding protein (FABP3) mediates the intracellular transport of fatty acids and is highly expressed in the myocardium. During myocardial infarction, FABP3 leaks from the myocardial cytoplasm into the blood; thus, detection of FABP3 using positron emission tomography (PET) is useful in identifying the site of myocardial damage. 4-(4-Fluoro-2-(1-phenyl-5-(2-(trifluoromethyl)phenyl)-1H-pyrazol-3-yl)phenoxy)butanoic acid (LUF) is a small-molecule organic compound that specifically binds FABP3. We confirmed that fluorine-18-labeled LUF enables clear, high-quality v
