The division of a mother cell into two daughter cells is a fundamental process in biology, involving the transfer of genomic information to the next generation of cells. In plant roots, cell divisions occur in the root apical meristem, a specialized tissue located at the plant's root tip. Cell proliferation rates in the root apical meristem shape root growth, thereby contributing to general development as well as the acclimatization to stressful environmental conditions through phenotypic plasticity. Here, we present a protocol for the microscopical analysis of cell proliferation in Arabidopsi

Eye development is a complex process that begins during the first stages of embryonic development; during its formation, a series of cell proliferation, migration, and differentiation events occur. A frequently used laboratory method is tissue staining with two dyes known as hematoxylin-eosin (H&E). H&E staining allows us to observe the arrangement of the cell layers and the formation of characteristic structures. It is fundamental for research in developmental biology since it facilitates a comprehensive analysis of the formation of organs and systems. In this chapter, we will describe the pro

BACKGROUND AND OBJECTIVES: Accurate visualization of thermal damage zone (TDZ) in the dermis is critical for energy-based device (EBD) and burn research. While nitroblue tetrazolium chloride (NBTC) staining and birefringence imaging are commonly used, both rely on detecting loss-of-signal, thereby only inferring TDZ indirectly. Other methods, such as hematoxylin and eosin and Masson's trichrome, are protocol-sensitive and limited in sufficiency and consistency for TDZ visualization. This study aims to validate a novel staining protocol optimized for direct and selective visualization of therma

In this study, we identify a peptide discovered through fluorescence-based screening of a cell-penetrating peptide (CPP) library that preferentially associates with bacterial spores and evaluate its utility as a fluorescence-guided tool for spore isolation. Specifically, an eight-ornithine peptide conjugated with 5(6)-carboxyfluorescein (Orn8-K-FAM) generates a distinct and spatially confined fluorescence signal in spores. Across multiple spore-forming bacterial species, more than 90% of spores were labelled by Orn8-K-FAM, whereas only a minor fraction of vegetative cells from non-spore-formin

Proximity labeling (PL) can identify endogenous proteins in specific subcellular regions. APEX2 (enhanced ascorbate peroxidase 2) enables PL with high temporal resolution by oxidizing biotin phenol (BP) into a radical that promiscuously tags nearby proteins, followed by the isolation and identification of the tagged biomolecules. However, the BP radical has a relatively large diffusion radius, limiting spatial resolution. Replacing the phenol in BP with nitrophenol (NP) could shorten the labeling radius by shortening the radical lifetime, but existing PL heme peroxidases cannot efficiently oxi

Chemical modification of proteins is an important tool for the development of protein-based therapeutics, a rapidly growing field in disease treatment. Currently, no universal approach exists for site-specifically introducing desired functionalities into proteins with minimal disruption. Here, we present a reactive peptide tag/probe pair system for site-specific covalent protein labeling, leveraging the interaction between a lysine-containing histidine tag (KH6 or H6K) and a binuclear nickel (II)-nitrilotriacetic acid (BisNi²+-NTA) probe conjugated to a lysine-reactive N-acyl-N-alkyl sulfonami

Visualization of cellular dynamics in microvascular networks is essential for deciphering physiology and disease. Existing imaging platforms commonly lack the spatiotemporal resolution and sensitivity for single-cell tracking in vivo, particularly beyond the penetration depth of optical microscopy. While recent fluorescence and optoacoustic methods allow detection of single microparticles in vivo, multiplexed imaging of distinct circulating cell populations remains unachieved. Here, we present an optimized labeling protocol using near-infrared (NIR) dyes ICG (indocyanine green) and DiR (1,1'-d

The initial step in diagnosing tuberculosis involves the microscopic examination of sputum samples using acid-fast staining to identify bacilli. However, this conventional method is labor-intensive, requires specialized expertise, is susceptible to errors, and has limited sensitivity. Research literature indicates that deep learning models demonstrate significant potential for detecting acid-fast bacilli (AFB) in sputum smear preparations. This study investigates the effectiveness of deep learning methods in identifying AFB within sputum smear samples. Our objective was to assess the performan

Here, we present a protocol for preserving fluorescent markers using tissue clearing and immunolabeling in mouse tissue such as sciatic nerve, gut, and spinal cord. We describe steps for maintaining tetrahydrofuran (THF) under an inert atmosphere, EZ clearing, immunolabeling, imaging with custom lightsheet microscopy setups, and post-processing. This protocol was refined to ensure stable and reliable clearing across various tissue types. Key improvements include inert atmosphere THF handling, tissue-specific delipidation, and enhanced antibody penetration via extracellular matrix loosening.

Immunofluorescence staining is a technique that permits visualization of components of various cell preparations. During spermiogenesis, cargo proteins are transported through the intra-manchette transport (IMT) to the basal bodies; then sperm flagella are assembled by intraflagellar transport (IFT). Due to the big complexes formed by IMT and IFT components, it has been challenging to visualize these components in tissue sections. Testicular tissue is digested by a combination of enzymes (DNAse 1 and Collagenase IV) and fixed by paraformaldehyde and sucrose. After being permeabilized with Trit

Objective This study aimed to compare the effects of two distinct intracellular antibody staining protocols-the sequential antibody incubation (A-P-A) method and the one-step antibody incubation (P-A) method-on flow cytometry results. Specifically, it focused on the median fluorescence intensity (MFI), the percentage of positive cells, and the observed differences between experimental groups. Methods The peripheral blood and splenocytes from 12-week-old male Kunming mice under different treatments were analyzed using two intracellular staining methods to assess differences in immune cell subse

Improvement in bacterial detection in smear is needed because pathogens in smears and positive blood cultures (PBCs) can be missed by microscopy if Gram staining is used. This study assessed the efficacy of Wright-Giemsa staining in remedying bacterial detection failures. We conducted a prospective multicenter study involving 5667 sputum specimens and 5502 PBCs collected from five distinct centers. When initial Gram staining did not identify bacteria, Wright-Giemsa staining was performed. The study also analyzed cases of missed detections and the undetected microorganism types. Among the sputu

PURPOSE: An ideal dye for intraocular use should effectively stain the target tissue while being easy to apply and remove. Additionally, it should not have any adverse effects resulting from prolonged contact with the retinal tissue. Recently, concerns have been raised about the safety of some vital dyes during surgical procedures as they may cross the internal limiting membrane and deposit on the retina. In this study, we aimed to investigate whether commercially available vital dyes, VIEW-ILM® and TWIN® (AL.CHI.MI.A. S.r.l., Ponte San Nicolò, Padova, Italy), have the potential to cross the i

Cutaneous graft versus host disease (cGVHD) has substantial clinical and histopathologic overlap with erythema multiforme (EM), Stevens-Johnson syndrome (SJS), and toxic epidermal necrolysis (TEN). This overlap can make it difficult to distinguish these disorders in patients who have received hematopoietic transplants. We sought to evaluate the utility of Dp I/II immunohistochemical stain in differentiating EM/SJS/TEN and cGVHD in a large cohort. Skin biopsy specimens from patients with cGVHD (n = 58) and EM/SJS/TEN (n = 60) were evaluated for Dp I/II expression by immunohistochemistry. We fou

We report about the muscular system and the serotonergic and FMRFamidergic components of the nervous system of the Bucephalidae trematode, Rhipidocotyle campanula, an intestinal parasite of the pike. We use immunocytochemical methods and confocal scanning laser microscopy (CLSM). The musculature is identified by histochemical staining with fluorescently labeled phalloidin. The body wall musculature of R. campanula contains three layers of muscle fibres - the outer thin circular, intermediate longitudinal and inner diagonal muscle fibres running in two opposite directions. The digestive system

BACKGROUND: There are currently nine known gill monogenean species of Diaphorocleidus Jogunoori et al. (Systematic Parasitology 58:115-124, 2004) (Dactylogyridae) which are dispersed within and/or among five fish families in the Neotropical Characiformes: Bryconidae, Acestrorhynchidae, Characidae, Iguanodectidae, and Hemiodontidae. During a fish parasitological study carried out in the Rio Lacantún from Chiapas, Mexico, a new species of Diaphorocleidus on the gill lamellae of Brycon guatemalensis Regan, 1908, was discovered.
OBJECTIVE: Our objective was to morphologically describe the new par

Microenvironment evaluation of intact tissue for analysis of cell infiltration and spatial organization are essential in understanding the complexity of disease processes. The principle techniques used in the past include immunohistochemistry (IHC) and immunofluorescence (IF) which enable visualization of cells as a snapshot in time using between 1 and 4 markers. Both techniques have shortcomings including difficulty staining poorly antigenic targets and limitations related to cross-species reactivity. IHC is reliable and reproducible, but the nature of the chemistry and reliance on the visibl

Renal α-enolase has variable expression in inflammatory and neoplastic diseases. Therefore, in order to define the distribution of α-enolase in renal tissues of cats, an immunohistochemistry assay was validated and described here. Tissues from 29 cats with IRIS Stage 2-4 CKD, 8 control cats < 2 years of age, and 4 control cats> 10 years of age were assessed. Interstitial nephritis was the predominant histopathological finding in the CKD group. The control cats < 2 years of age had moderate α-enolase immunoreactivity in tubular epithelium but staining was absent to mild in glomeruli. In contras
