Tissue engineering of skin: human tonsil-derived mesenchymal cells can function as dermal fibroblasts.

PURPOSE: It is unclear whether dermal fibroblasts are indispensable key players for tissue engineering of dermo-epidermal skin analogs. In this experimental study, we wanted to test the hypothesis that tonsil-derived mesenchymal cells can assume the role of dermal fibroblasts when culturing pigmented skin analogs for transplantation. METHODS: Mesenchymal cells from excised tonsils and keratinocytes, melanocytes, and fibroblasts from skin biopsies were isolated, cultured, and expanded. Melanocytes and keratinocytes were seeded in a ratio of 1:5 onto collagen gels previously populated either wi…

Electrospinning of gelatin for tissue engineering--molecular conformation as one of the overlooked problems.

Gelatin is one of the most promising materials in tissue engineering as a scaffold component. This biopolymer indicates biocompatibility and bioactivity caused by the existence of specific amino acid sequences, being preferred sites for interactions with cells, with high similarity to natural extracellular matrix. The present paper does not aspire to be a full review of electrospinning of gelatin and gelatin containing nanofibers as scaffolds in tissue engineering. It is focused on the still open question of the role of the higher order structures of gelatin in scaffold's bioactivity/functiona…

Poly(lactic-co-glycolic acid) microspheres as an injectable scaffold for cartilage tissue engineering.

Injectable scaffold has raised great interest for tissue regeneration in vivo, because it allows easy filling of irregularly shaped defects and the implantation of cells through minimally invasive surgical procedures. In this study, we evaluated poly(lactic-co-glycolic acid) (PLGA) microsphere as an injectable scaffold for in vivo cartilage tissue engineering. PLGA microspheres (30-80 microm in diameter) were injectable through various gauges of needles, as the microspheres did not obstruct the needles and microsphere size exclusion was not observed at injection. The culture of chondrocytes on…

Comminuted fractures associated with tissue loss can adversely affect bone regeneration. Biomaterials enriched with mesenchymal stem cells (MSCs) employed for supporting osteosynthesis and potentiating osteoconduction are necessary to fill these bone defects. Natural compound biomaterials, similar to bone tissue, have been extensively tested in animal models for clinical use. Bone tissue engineering studies have used critical-size defects in ovine tibia monitored by imaging and histological examinations to evaluate the regenerative process. This study aimed to monitor the regenerative process …

Biopreservation of a tissue engineered nigrostriatal pathway for tract reconstruction in Parkinson's disease.

The typical motor symptoms of Parkinson's disease (PD) are caused by selective loss of dopaminergic neurons in the substantia nigra (SN). Although conventional pharmacotherapies can temporarily alleviate symptoms, no approved therapies exist to slow or reverse the underlying pathologic processes. To address this gap, cell transplantation therapies are being pursued; however, restoration of the original neuroanatomical circuit is not a goal of traditional ectopic intra-striatal neuronal transplantations. To address this, we developed an implantable tissue engineered nigrostriatal pathway (TE-NS

The PEDIAVALVE project - tissue-engineered pulmonary valves for paediatric valvular replacement.

Current paediatric options for pulmonary heart valve (PHV) substitutes are imperfect, requiring numerous reinterventions until maturity. Substitutes with a diameter under 15 mm have a limited service life, underscoring the critical need for small-diameter valves. To address this need, we obtained and tested decellularized, DNA-depleted, small-diameter valvular substitutes using tissue-engineered lamb PHVs. Lamb PHVs were subjected to a decellularization perfusion protocol, yielding a 3D extracellular matrix (ECM)- derived acellular scaffold. Sterility, residual nuclei (histology) and DNA conte

Understanding Degeneration and Healing Pathways for Tissue-Engineered Treatment Strategies in Tendinopathy.

BACKGROUND: The multidisciplinary nature of the tendon tissue engineering field brings challenges to move from our basic understanding of tendon biology towards engineering strategies for tendon disorders. Such pathologies present a high risk of inflammation for young patients and limited tissue regeneration capability in ageing patients, leading to painful symptoms and impaired quality of life. SUMMARY: Among the complications observed in tendinopathy are degenerative changes in the extracellular matrix and aberrant tissue remodelling, accounting for a huge burden in musculoskeletal disorder…

Modeling and Optimization of Nonlinear Viscoelastic Behavior for Tissue-Engineered Blood Vessels.

Vascular tissue engineering technology uses tubular viscoelastic materials as intermediaries to transmit the mechanical stimuli required for the construction of vascular grafts. However, most existing studies rely on elastic models, which fail to capture the time-dependent nature of viscoelastic materials. Moreover, the long fabrication cycles, high costs, and complex parameter measurements in tissue engineering pose significant challenges to experimental approaches. There is thus an urgent need to develop a viscoelastic mechanical model that combines physical interpretability, computational e…

Pediatric Burns: Biological and Tissue Engineered Skin Substitutes-A Systematic Review.

Background/Objectives: Surgical debridement and early excision of burned areas followed by skin autograft is the gold standard of treatment for partial and full-thickness pediatric burns. However, skin autografting might be unfeasible or unlikely to succeed due limited availability of skin donor areas or inadequate conditions. In these circumstances, alternative treatment is required, and Skin Substitutes (SS) cold play a role. Recently, Biological Skin Substitutes (BSS) and Tissue Engineered Skin Substitutes (TESS) are emerging as alternative treatment options, but strong evidence is missing.

Evaluation of plant-derived biomaterials for the development of tissue-engineered corneal substitutes.

Corneal blindness affects over 10 million patients worldwide. Due to the limited supply of donor corneas and frequent graft failure, bioengineered alternatives are crucial. To overcome drawbacks associated with corneal substitutes from synthetic biomaterials, fabrication from plant-derived biomaterials is a potential alternative. Herein, soy protein and glutenin in combination with different crosslinkers were evaluated for fabrication of corneal substitutes. Optical, mechanical, and biochemical properties of fabricated constructs and control rabbit corneas were evaluated in vitro. Soy protein
