Yeast surface display meets immunotherapy: unlocking the potential of T cells.

BACKGROUND AIMS: While T-cell receptors (TCRs) hold immense therapeutic potential, their clinical translation is bottlenecked by the scarcity of high-affinity, specific clones and the instability of recombinant formats. Existing display technologies fall short: phage display often fails to produce properly folded TCRs, and mammalian display is low-throughput. This review is the first to comprehensively synthesize how yeast surface display (YSD) is uniquely positioned to overcome these hurdles and build a new pipeline for TCR based drug discovery. METHODS: We articulate how YSD's eukaryotic ma

Role of the CYR1G2066A mutation in ethanol tolerance of sake yeast Kyokai no. 11.

Sake yeast Kyokai no. 11 (K11) is an ethanol-tolerant mutant of Kyokai no. 7 (K7) and produces a higher ethanol concentration in the sake mash than K7. A previous study revealed that stress-induced genes under the control of STRE elements were upregulated in K11. To elucidate the causal mutation responsible for ethanol tolerance, we compared the genome sequences of the ethanol-tolerant mutants (K11 and K7AT2, a newly isolated ethanol-tolerant mutant of K7) with that of their parental strain, K7. We identified a shared loss of heterozygosity region in the left arm of chromosome X in both mutant

A decoupled transcription platform enables tunable and predictable gene expression in yeast.

Predictable control of gene expression is essential for building genetic circuits and improving metabolic pathways, but conventional promoter libraries often behave unpredictably when genes are combined. Here we develop CRISPR-Activated Promoter-based Orthogonal expression (CAPO), a quantitative platform for controlling multiple genes in yeast. CAPO uses synthetic CRISPR-activated promoters that remain silent until matching guide RNAs recruit dCas9-VPR. We tune each gene by varying guide RNA abundance with defined T7 promoters, while keeping regulatory channels orthogonal. CAPO reaches express

Structural Basis and Molecular Mechanism of Yeast Biosorption: for Food Safety and Nutrition.

Yeast is widely used in food processing, yet its unique cellular structure and the resulting potential for biosorption remains underutilized. A literature search was performed in databases including PubMed and ScienceDirect using a combination of relevant keywords, including yeast biosorption, food contaminants, bioactive compounds, and biosorption enhancement. The search was specifically focused on studies pertaining to food or food-related systems. This narrative review elucidates that rapid, passive surface biosorption mediated by the yeast cell membrane (primarily the mannan layer) conform

The Effects of Dual-Yeast Compound Preparation on the Intestinal Health and Metabolism of Lambs.

Microecological preparations exert beneficial effects on the health of young ruminant animals; however, the mechanism is unclear. As a result, the present study analyzed the effects of yeast cultures on the growth properties, microbiome, and metabolism of weaned lambs. In this study, a total of 20 weaned lambs were randomly, stochastically divided into four teams: the control group (Group A) were fed a basic diet; Group B were fed with Saccharomyces cerevisiae BC strain culture (30 g/head/d); Group C were fed with Kluyveromyces marquez XR4 strain culture (30 g/head/d); Group D were fed with a

Yeast single-cell proteins from CO2 with tandem biological systems.

Production of customized yeast single-cell protein (SCP) from CO2 offers a sustainable avenue to convert a major carbon emission process into a carbon-conserving production route of food and feed. Here, we highlight a specific approach for SCP production from CO2 using sequentially connected yet spatially separated tandem biological systems, which consist of a first unit for CO2 fixation followed by a second unit for precision fermentation toward customized SCPs. We discuss the design principles of the system, review current efforts to convert CO2 using natural and engineered autotrophs, and o

Stabilization of Amylopectin-Xyloglucan Water-in-Water Pickering Emulsions by Yeast Cells.

Pickering water-in-water (W/W) emulsions stabilized by biocompatible particles represent a promising strategy for the design of functional soft materials. In this study, we investigate the ability of Saccharomyces cerevisiae, in both viable (Y-AC) and thermally inactivated (Y-IN) forms, to stabilize amylopectin-xyloglucan (AMP-XG) W/W emulsions, providing the first report of yeast-based Pickering stabilization in this polymer-polymer system. Physicochemical characterization, including FTIR, ζ-potential measurements, and potentiometric titration, revealed that thermal inactivation induces a reo

The transcriptome of dendritic cells redraws the boundaries between pathogenicity and commensalism in yeast.

Our immune system is constantly exposed to fungi, but it mounts a consistent, disease-related, and species-specific inflammatory response only against a few fungal species. Most of the current understanding of fungus-host interactions is based on a limited number of strains, hence neglecting the fungal intra-specific genetic and phenotypic diversity. To expand our knowledge of the spectrum of immune responses to pathogenic and non-pathogenic fungi, we compared the cytokine and transcriptional profiles of human monocyte-derived dendritic cells exposed to Aspergillus fumigatus, Candida albicans,

A bacterial metabolite rewires fungal metabolism, triggering the hyphae-to-yeast transition.

Fungi transition between cell morphologies to adapt to and colonize environments. For Candida albicans, commensal colonization of humans and virulence depend on a reversible switch between invasive hyphae and disseminating yeast cells. The yeast-to-hyphae transition is well understood, but comparatively little is known about the reverse transition from hyphae back to yeast. By developing an imaging assay to visualize and quantify the hyphae-to-yeast transition, we show that the bacterial natural product gladiolin accelerates the transition. Gladiolin reprograms C. albicans metabolism, causing

Mechanism of the synergistic action of oxythiamine and ketoconazole against the yeast Malassezia pachydermatis.

This article explains the biochemical basis of the synergistic effect of oxythiamine (OT) and ketoconazole (KTC) against Malassezia pachydermatis yeast, which was isolated from dogs exhibiting clinical signs of otitis externa. All strains were incubated on MLNA medium supplemented with OT, KTC, or a mixture of both compounds. We found that the ergosterol content was reduced by the compounds tested, both separately (20%-50%) and in combination (80%). OT alone and in combination with KTC reduced NADPH levels. However, we found no differences in acetyl-CoA levels under the influence of the compou

[Yeast as a Biochemical Model for Diseases Associated with Impaired Intracellular Proteolytic Systems].

The degradation of intracellular proteins is a fundamental biological process necessary for maintaining cellular homeostasis, controlling the cell cycle, regulating signal transduction, and preventing the accumulation of toxic protein aggregates. Disorders of the proteolytic systems are implicated in the pathogenesis of numerous human diseases, including neurodegenerative diseases, lysosomal storage disorders, metabolic disorders, and certain types of cancer. The development of rudimentary and cost-effective models of these diseases for the purpose of evaluating novel pharmaceutical agents and

A case report of childhood onychomycosis caused by the rare yeast Kodamaea ohmeri.

Onychomycosis is an uncommon disease in pediatric patients with dermatophytes and Candida spp. being the main causative agents. Kodamaea ohmeri has recently emerged as a human pathogen, including an onychomycosis causative agent. Here, we report the first case of childhood onychomycosis caused by K. ohmeri in Vietnam, presenting clinically as a white superficial onychomycosis. Fungal identification was confirmed by sequencing of the ITS1-2 region. Antifungal susceptibility testing revealed low minimum inhibitory concentrations for all tested agents, except fluconazole and caspofungin. The pati

Immunostaining the Yeast Nuclear Membrane for Imaging by Super-Resolution Fluorescence Microscopy.

The ability to image biological samples beyond the diffraction limit (super-resolution) has opened a world of new discoveries in molecular biology. Super-resolution fluorescence microscopy requires specific fluorophores that often require immunostaining with labeled antibodies to achieve the necessary parameters for imaging. Budding yeast Saccharomyces cerevisiae is a common model organism used in cell biology, genetics, and molecular biology. However, yeast are difficult to immunostain due to the cell wall that hinders antibody penetration. In this chapter, I describe the methods we use for i

Development of a yeast-based CRISPR genome editing system for feline coronavirus.

INTRODUCTION: Feline infectious peritonitis (FIP), caused by feline coronavirus (FCoV), is a highly lethal disease characterized by systemic organ infection in cats. Current challenges of FIP include the absence of definitive diagnostic criteria, effective vaccines, and targeted therapies. Developing a robust genome editing toolkit is therefore critical to unraveling FCoV replication and pathogenesis mechanisms, elucidating viral protein functions, and identifying promising diagnostic and therapeutic targets. METHODS: In this study, we developed a yeast-based CRISPR genome editing system comp
