Use of antibiotics in food animals may contribute to development and spread of resistant organisms, particularly so in some countries. The aim of this study was two-fold; first, to establish the prevalence of extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli in chicken production in a region within Romania. Second, to study the relatedness of ESBL-producing E. coli isolates recovered from broilers, abattoir workers where the chickens were slaughtered and from the human clinical specimens from two regional hospitals. The results indicated a very high (69%) rate of carriage of E

BACKGROUND: Extended-spectrum beta-lactamase (ESBL)-producing Enterobacteriaceae is an emerging therapeutic challenge, especially in the treatment of urinary tract infections. Following an outbreak of CTX-M-15 Klebsiella pneumoniae in Uppsala, Sweden, an orphan drug trial on IgY chicken antibodies was undertaken in an attempt to eradicate faecal carriage of ESBL-producing K. pneumoniae and Escherichia coli.
METHODS: Hens were immunised with epitopes from freeze-dried, whole-cell bacteria (ESBL-producing K. pneumoniae and E. coli) and recombinant proteins of two K. pneumoniae fimbriae subunits

Boronic acid transition state inhibitors (BATSIs) are potent class A and C β-lactamase inactivators and are of particular interest due to their reversible nature mimicking the transition state. Here, we present structural and kinetic data describing the inhibition of the SHV-1 β-lactamase, a clinically important enzyme found in Klebsiella pneumoniae, by BATSI compounds possessing the R1 side chains of ceftazidime and cefoperazone and designed variants of the latter, compounds 1 and 2. The ceftazidime and cefoperazone BATSI compounds inhibit the SHV-1 β-lactamase with micromolar affinity that i

We studied the antibiotic resistance among Escherichia coli isolates obtained from fecal samples of dogs and cats treated and untreated with enrofloxacin in veterinary clinics. Resistant patterns of 70 strains and the presence of extended-spectrum beta-lactamase (ESBL) were studied. The genes encoding the following families of beta-lactamases: CTX-M, GES, PER, TEM and SHV, were investigated by PCR-RFLP and sequencing. The strains isolated from enrofloxacin-treated animals were multi-drug-resistant exhibiting resistant patterns including fluorquinolones, beta-lactams, aminoglycosides, tetracycl…

To determine and type the extended-spectrum beta-lactamases (ESBLs) among the family Enterobacteriaceae in a medical center, a total of 668 clinical isolates were screened. Of the 668 isolates, the 80 strains were presumptively defined as ESBL producers according to the result of disk method using ESBL marker antibiotics (aztreonam, ceftazidime, and cefoxitin). These 80 strains were retested with the double-disk synergy test (DDST), the E-test ESBL strip, a 5-microg ceftazidime disk, and agar dilution MICs of ceftazidime with and without clavulonic acid. Isoelectric focusing was performed to c…

Introduction: The combination of ceftazidime-avibactam (CAZ-AVI) with aztreonam (ATM) may be an option for the treatment of infections due to metallo-β-lactamases (MBLs) producing bacteria, as recommended by current guidelines. MBLs protect the pathogen from any available β-lactam/β-lactamase inhibitor (BL/BLI). Moreover, in vitro and clinical data suggest that double carbapenem therapy (DCT) may be an option for such infections. Materials and Methods: This retrospective study was conducted in two mixed intensive care units (ICUs) at the University Hospital of Larissa, Thessaly, Greece, and th

The emergence of multi-beta-lactam resistance is a limiting factor in treating invasive Pseudomonas infections with newer cephalosporins. The in vivo efficacy of ciprofloxacin, a new carboxy-quinolone, was evaluated in experimental aortic valve endocarditis caused by a strain of Pseudomonas aeruginosa which is stably derepressed for beta-lactamase production and is resistant to ceftazidime and multiple other beta-lactam agents. A total of 51 catheterized rabbits with aortic catheters in place were infected with this strain and then received no therapy (controls), ceftazidime (75 mg/kg per day)

Klebsiella spp. present in the food chain have been of much interest during the last few decades due to their implication in the development of antimicrobial resistance. We determined the prevalence of Klebsiella spp. (Klebsiella pneumoniae and Klebsiella oxytoca) in retail poultry meat, along with their resistance profile against antimicrobials. For the detection of the Klebsiella spp. a total of 285 raw meat samples of retailed poultry were collected and cultured. All positive cultures were screened for Klebsiella spp. by PCR to confirm the identity. Their antimicrobial susceptibility patter

Bacterial resistance to antibiotics is increasing globally, with the food-animal sector (FAS) playing a key role. Knowledge of the antimicrobial resistance (AMR) of microbes from the FAS is important in the development of country-specific methods to minimize the AMR burden. In Ghana, there is limited data on the susceptibility of FAS bacteria to frequently used antimicrobials. We evaluated the susceptibility of 58 Escherichia coli isolates obtained from chickens to nine antibiotics and further assessed their potential to produce extended-spectrum beta-lactamase (ESBL). The Kirby-Bauer disc dif

Antimicrobial resistance (AMR) is a growing concern in veterinary and public health, with extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli playing a significant role. This study examined 1,000 healthy and sick dogs from a veterinary clinic in northern Germany and identified 85 ESBL-producing E. coli. Whole-genome sequencing of these isolates revealed seven phylogroups. A (38.8%) and B1 (32.9%) were the most common. Multilocus sequence typing identified 42 sequence types (STs), with the globally occurring lineages ST744 and ST10 being predominant. Single nucleotide polymorphis

Though globally prevalent, Shigella infections rarely progress to bacteremia (shigellemia), particularly in immunocompetent individuals. Here, we report a case of shigellemia with extended-spectrum beta-lactamase (ESBL) in Qatar, involving a 53-year-old immunocompetent male with no significant medical history. The patient presented with a one-day history of frequent loose stools, fever, and mild central abdominal pain. Stool and blood cultures confirmed Shigella sonneiinfection and shigellemia with ESBL production. The patient tested negative for human immunodeficiency virus (HIV), had normal
