Occurrence of arbovirus infections in two riverine populations in the municipality of Humaitá, Amazonas, Brazil.

BACKGROUND: The riverine communities of the Amazon comprise different social groups that inhabit the rural areas on the banks of rivers and lakes. Residents usually travel by river to rural and urban areas and are then exposed to urbanized diseases such as those caused by arbovirus infection. In Brazil, emerging diseases such as dengue, Zika, chikungunya, and those caused by infection with Oropouche and Mayaro viruses necessitate epidemiological surveillance. This study was aimed at determining the frequency of positivity for immunoglobulin (Ig)G and IgM antibodies against Zika, chikungunya, a

Virus isolations from mosquitoes collected during the 1982 Japanese encephalitis epidemic in northern Thailand.

From 16 June to 15 August, 1982 CDC light traps were used to collect mosquitoes in the province of Kamphaengphet, N. Thailand. 353,042 mosquitoes comprising 59 species were collected and identified, and 345,173 were placed in pools for attempted virus isolation by inoculation of C6/36 Aedes albopictus mosquito cell cultures. Viruses were isolated from 63 mosquito pools. These comprised 56 flaviviruses, identified as 35 isolates of Japanese encephalitis (JE) virus strains, 18 strains of Tembusu (TEM) virus and three untyped flaviviruses (FLA); three alphaviruses, identified as the first isolate…

A sero-epidemiological survey for certain arboviruses (Togaviridae) in Pakistan.

Complement-fixation test reactions to eight viruses of the family Togaviridae were studied in 372 serum samples (157 rodents, 172 domestic animals, 43 humans) from Pakistan. Antibodies to each tested virus were detected. The highest over-all prevalence rates were for West Nile (WN) (7.8%), Japanese encephalitis (JE) (3.2%) and Zika (ZIKA) (2.4%) viruses, followed by Sindbis (SIN), Chikungunya (CHIK), Uganda S (UGS) and Royal Farm (RF) viruses (1.6 to 1.3%). One human serum (male, age 58 years) reacted with Dengue-1 (DEN) virus antigen (titre 1:32). Antibodies to each virus except RF were detec

2-Deoxy-D-glucose attenuates lipopolysaccharide-induced inflammation and restricts Zika, Chikungunya, and Mayaro virus replication in monocyte-derived macrophages.

Re-emerging arthropod-borne viruses such as Mayaro (MAYV), Chikungunya (CHIKV), and Zika (ZIKV) pose a growing global health concern as Aedes mosquito populations expand. These arboviruses infect innate immune cells, particularly monocyte-derived macrophages (MDMs), which support viral replication and serve as reservoirs that facilitate dissemination. Because no effective antiviral treatments are available, strategies that modulate macrophage responses and restrict viral replication are urgently needed. Here, we evaluated the immunomodulatory and antiviral effects of 2-deoxy-D-glucose (2-DG) i

Prospective study of Chikungunya virus acute infection in the Island of La Réunion during the 2005-2006 outbreak.

BACKGROUND: Chikungunya virus (CHIKV) is a recently re-emerged arthropod borne virus responsible for a massive outbreak in the Indian Ocean and India, and extended to Southeast Asia as well as Italy. CHIKV has adapted to Aedes albopictus, an anthropophilic mosquito species widely distributed in Asia, Europe, Africa and America. Our objective was to determine the clinical and biological features of patients at the acute phase of CHIKV infection. METHODS AND FINDINGS: A prospective study enrolled 274 consecutive patients with febrile arthralgia recorded at the Emergency Department of the Groupe

Development and evaluation of antigen capture ELISA for early clinical diagnosis of chikungunya.

The resurgence of chikungunya (CHIK) in the form of unprecedented explosive epidemic after a gap of 3 decades in India and Indian Ocean islands is a point of major public health concern. The laboratory diagnosis is essentially based on virus isolation, IgM ELISA, and reverse transcriptase polymerase chain reaction (RT-PCR). Although PCR-based methods are used for early and accurate diagnosis, the high cost of the assay and requirement of thermal cycler limit its application only to referral laboratories. The antibody-based IgM ELISA is found to be cost-effective, but it takes 5 to 6 days for t

Putative papain-related thiol proteases of positive-strand RNA viruses. Identification of rubi- and aphthovirus proteases and delineation of a novel conserved domain associated with proteases of rubi-, alpha- and coronaviruses.

A computer-assisted comparative analysis of the amino acid sequences of (putative) thiol proteases encoded by the genomes of several diverse groups of positive-stranded RNA viruses and distantly related to the family of cellular papain-like proteases is presented. A high level of similarity was detected between the leader protease of foot-and-mouth-disease virus and the protease of murine hepatitis coronavirus which cleaves the N-terminal p28 protein from the polyprotein. Statistically significant alignment of a portion of the rubella virus polyprotein with cellular papain-like proteases was o

Vector competence of Aedes albopictus from Houston, Texas, for dengue serotypes 1 to 4, yellow fever and Ross River viruses.

A combination of virus infection and transmission experiments showed that a Houston, Texas strain of Aedes albopictus is a competent vector for dengue (DEN), yellow fever (YF) and Ross River (RR) viruses. However, at 14 days incubation, DEN virus infection rates in a Puerto Rican strain of Aedes aegypti were significantly higher for each of the four DEN serotypes, except DEN-1, than in Houston Ae. albopictus fed simultaneously on the same virus suspensions. The degree of correlation between disseminated DEN infection rates in Houston Ae. albopictus and transmission to an in vitro system ranged

Replication of alphaviruses in cultures of donkey monocytes.

Representative strains of Venezuelan equine encephalitis virus (VEEV) and Eastern equine encephalitis virus (EEEV) were compared for their ability to grow in cultures of unstimulated leucocytes and monocytes derived from donkey peripheral blood. Replication of epizootic and vaccine strains of VEEV, but not of enzootic strains was observed in this system. Only a minority of monocytes supported virus replication as detected by immunofluorescence, electron microscopy and infectious center assays. EEEV did not appear to replicate in this cell system although virus attached to and was internalized

Experimental viral polymyositis: age dependency and immune responses to Ross River virus infection in mice.

Ross River virus (RRV) causes an age-dependent myositis in mice. Infected 4-week-old mice develop no clinical signs, but 1-week-old mice develop weakness and myositis. Humoral and cell-mediated immune responses to RRV in the two age groups are comparable, and immunosuppression does not alter age-dependent resistance to clinical disease. Immunosuppression of 1-week-old mice protracts clinical signs and reduces muscle inflammation but does not alter muscle necrosis or regeneration. These studies suggest that immune responses do not determine age dependency of RRV myositis and that muscle necrosi

Infection of Rhesus Macaques with O'nyong-nyong virus UVRI-0804 recapitulates key aspects of human clinical disease.

O'nyong-nyong virus (ONNV) is a mosquito-borne alphavirus first isolated in Uganda in 1959. Since its discovery, ONNV has caused several outbreaks in Africa, manifesting clinically as fever, rash, and joint/muscle pain lasting months. Currently, we have a limited understanding of ONNV infection and disease in relevant animal models, which restricts the evaluation of vaccines and therapeutics. In 1967, Binn et al. reported that infection of rhesus macaques (RMs) with ONNV failed to induce viremia in two animals. This may be attributed to the potential attenuation of the virus through extensive …

Coinfection of tick cell lines has variable effects on replication of intracellular bacterial and viral pathogens.

Ticks transmit various human and animal microbial pathogens and may harbour more than one pathogen simultaneously. Both viruses and bacteria can trigger, and may subsequently suppress, vertebrate host and arthropod vector anti-microbial responses. Microbial coinfection of ticks could lead to an advantage or disadvantage for one or more of the microorganisms. In this preliminary study, cell lines derived from the ticks Ixodes scapularis and Ixodes ricinus were infected sequentially with 2 arthropod-borne pathogens, Borrelia burgdorferi s.s., Ehrlichia ruminantium, or Semliki Forest virus (SFV),

Facial hyperpigmentation: causes and treatment.

By midcentury, the U.S.A. will be more ethnically and racially diverse. Skin of colour will soon constitute nearly one-half of the U.S. population, and a full understanding of skin conditions that affect this group is of great importance. Structural and functional differences in the skin, as well as the influence of cultural practices, produce variances in skin disease and presentation based on skin type. In the skin of colour population, dyschromia is a growing concern, and a top chief complaint when patients present to the physician. A thorough understanding of the aetiology and management s

Gustatory receptor expression in the labella and tarsi of Aedes aegypti.

The yellow-fever mosquito, Aedes aegypti, infects a growing number of people every year with dengue, yellow fever and chikungunya viruses. Contact chemoreception in mosquitoes influences a number of behaviors including host-selection, oviposition and feeding. While these behaviors are in many instances well documented, the molecular mechanisms mediating them are not well understood. Here we report the results of sequencing total messenger RNA in the labella and tarsi of both male and female Ae. aegypti to reveal Gustatory Receptor (GR) gene expression profiles in these major gustatory appendag

Underdiagnosis of chikungunya virus infections in symptomatic dutch travelers returning from the Indian ocean area.

A putative underdiagnosis of clinical chikungunya virus infection in Dutch travelers to the Indian Ocean area was addressed by retrospective screening of all sera for which requested dengue virus serology was negative in the period 2007 to 2010. Evidence for a recent infection was observed in 6.5% of 107 patients, indicating a substantial underdiagnosis and the need for increased awareness among physicians.

Diagnostics of Pogosta disease: antigenic properties and evaluation of Sindbis virus IgM and IgG enzyme immunoassays.

Sindbis virus (SINV) is a mosquito-borne causative agent of a fever-rash arthritis, Pogosta disease, as verified recently by virus isolation from acutely ill patients. Pogosta disease occurs annually, but it emerges as unique epidemics every 7 years in Finland; over 10,000 patient samples have been analyzed for SINV antibodies, with over 2000 diagnosed acute SINV infections. However, the performance of these serological tests with a large number of samples has not been described before. The aim of the present study was to characterize and evaluate methods developed for the serodiagnostics of S
